Phosphorylation-guarded light-harvesting complex II contributes to broad-spectrum blast resistance in rice.

Environmental conditions are key factors in the progression of plant disease epidemics. Light affects the outbreak of plant diseases, but the underlying molecular mechanisms are not well understood. Here, we report that the light-harvesting complex II protein, LHCB5, from rice is subject to light-induced phosphorylation during infection by the rice blast fungus Magnaporthe oryzae We demonstrate that single-nucleotide polymorphisms (SNPs) in the LHCB5 promoter control the expression of LHCB5, which in turn correlates with the phosphorylation of LHCB5. LHCB5 phosphorylation enhances broad-spectr

Suppressing an AUX/IAA gene GhIAA43 expression activates the SA-mediated immune pathway and enhances Verticillium wilt resistance in cotton.

Salicylic acid (SA) and auxin often exert antagonistic effects during pathogen challenge, and SA's inhibitory effect on auxin-mediated signaling pathway plays a crucial role in plant defense mechanisms. However, whether SA-mediated immunity can be enhanced through the inhibition of auxin signaling pathway remains unclear. Cotton GhIAA43, a member from the Aux/IAA family, was a resistance-related gene against Verticillium dahliae attack. Here, we clarified the role of GhIAA43 in regulating the auxin and SA-mediated pathways to improve the plant resistance by using the RNA interference (RNAi)-Gh

Aspartic Acid Catalyzed by Asparaginase Provides Rice with Broad-Spectrum Resistance.

Amino acid metabolism plays a key role in regulating interactions between plants and pathogens. Asparaginase hydrolyzes asparagine, a major … al for their enzyme activity and immune function, and identified aspartic acid as a key molecule in plant immunity. This study reveals a novel mechanism linking asparagine metabolism to rice immunity and provides a target for breeding disease-resistant crops.

Discovery of immunostimulatory CpG-DNA and its application to tuberculosis vaccine development.

DNA containing an unmethylated CpG motif has a potent immunostimulatory effect on the vertebrate immune system. Because such CpG motifs are relatively common in bacterial DNA, but rare in mammalian animal and plant DNA, they may be an evolutionary adaptation augmenting innate immunity, most likely in response to pathogens that replicate within the host cells, such as viruses and intracellular bacteria. Microbial infection induces innate immunity by triggering pattern-recognition systems. The infected cells produce proinflammatory cytokines that directly combat microbial invaders and express co

A CsWRKY46-CsPBL9-CsARI1 tripartite regulatory module coordinates H2O2 production and callose deposition in citrus fruit immunity.

Plant immunity against pathogens involves multiple immune responses and intricate regulatory networks. However, how immune networks are deployed in fruit remains poorly understood. Here, we show that citrus fruit immune responses, including hydrogen peroxide (H2O2) production and callose deposition, are multiply regulated by transcriptional activation, phosphorylation, and ubiquitination. Citrus sinensis genes encoding nicotinamide adenine dinucleotide phosphate (NADPH) oxidase CsRBOHG and callose synthase CsCalS5, responsible for H2O2 production and callose deposition, respectively, are trans…

A central circadian oscillator confers defense heterosis in hybrids without growth vigor costs.

Plant immunity frequently incurs growth penalties, which known as the trade-off between immunity and growth. Heterosis, the phenotypic superiority of a hybrid over its parents, has been demonstrated for many traits but rarely for disease resistance. Here, we report that the central circadian oscillator, CCA1, confers heterosis for bacterial defense in hybrids without growth vigor costs, and it even significantly enhances the growth heterosis of hybrids under pathogen infection. The genetic perturbation of CCA1 abrogated heterosis for both defense and growth in hybrids. Upon pathogen attack, th…

Plant virus-derived siRNAs stay a step ahead of viral short-range spread.

Whether mobile virus-derived siRNAs can pre-immunize uninfected plant tissues has been a long-standing assumption in plant biology. In this issue, Albertini et al. demonstrate that they indeed establish an AGO1-dependent immune zone of adjacent uninfected short-range cells, and this mobile pre-immunization is impacted by viral suppressors of RNA silencing.

Plant viruses and the microbiome: a complex network shaping plant health and disease resistance.

Plant viruses and microbiomes are pivotal to plant health, growth, and their ability to withstand environmental challenges. However, the extent to which viruses engage in direct or indirect crosstalk with plant-associated microbial communities remains largely unexplored. Traditionally, plant viruses are considered as harmful agents that disrupt normal plant functions, but new research indicates they may also engage in complex interactions with the plant's microbiome. On the other hand, the plant microbiome, comprising a diverse assembly of bacteria, fungi, and other microbes, can influence the

Food Waste Compost as a Tool of Microbiome-Assisted Agri-Culture for Sustainable Protection of Vegetable Crops Against Soil-Borne Parasites.

A low-scale Food Waste Compost (FWC1), characterized by optimal physic-chemical parameters and high organic matter percentages, was used as a fertilizer and a bio-stimulant for vegetable plants. Groups of treated plants were inoculated with active juveniles of root-knot nematodes to detect the effect on plant defense. Optimal amounts of compost mixed with soil increased plant biomass 30% compared to untreated plants. Moreover, when plants were inoculated, treated roots contained about 50% less sedentary forms (SFs) of nematodes and a lower reproduction rate of the parasites than untreated plan

A plant pathogen effector blocks stepwise assembly of a helper NLR resistosome.

Helper NLRs function as central nodes in plant immune networks. Upon activation, they oligomerize into inflammasome-like resistosomes to initiate immune signaling, yet the dynamics of resistosome assembly remain poorly understood. Here, we show that the virulence effector AVRcap1b from the Irish potato famine pathogen Phytophthora infestans suppresses immune activation by directly engaging oligomerization intermediates of the tomato helper NLR SlNRC3. Cryo-EM structures of SlNRC3 in AVRcap1b-bound and unbound states reveal that AVRcap1b bridges multiple protomers, stabilizing a stalled interme

Fungal pathogens of plants: deciphering the mechanisms of Fusarium wilt.

Fusarium oxysporum (Fo) is among the most dangerous soilborne pathogens, causing Fusarium wilt and root rots in over 100 plant species worldwide. Some pathogen strains can also infect immunocompromised animals and humans. Consequently, studying the molecular mechanisms associated with pathogen virulence and the plant immune response at different stages of disease development is of paramount importance. The process of host recognition by the pathogen and all stages of the infection process involve a wide repertoire of specific signaling molecules, effector proteins, receptor complexes, as well

An empirical test of the biodiversity hypothesis: Exposure to plant diversity is associated with a reduced risk of childhood acute lymphoblastic leukemia.

The biodiversity hypothesis posits that declining biodiversity may be responsible, at least in part, for the global increase in immune diseases. However, few studies have been able to demonstrate a link between exposure to biodiversity and specific health outcomes. We test whether exposure to plant diversity protects against childhood acute lymphoblastic leukemia (ALL) by promoting immune maturation. Our sample consisted of all children born in New Zealand from 1998 to 2013 (n = 899,126; 264 ALL cases), which we followed from birth to age five. We calculated plant-diversity metrics using the G

Transformative potential of plant-based nanoparticles in cancer diagnosis and treatment: bridging traditional medicine and modern therapy.

Cancer is a primary global health concern, with an estimated 35.3 million cancer cases expected worldwide, representing a 76.6% increase in 2022, and 20 million by 2050, resulting from genetic mutation and environmental factors that cause uncontrolled cell growth. Other factors including smoking, unhealthy diets, physical inactivity, exposure to carcinogens, UV radiation, and aging increase DNA damage. Current cancer treatments like chemotherapy, radiation therapy, immunotherapy, and surgery are effective, but those have significant effects like lack of specificity, development of drug resista

Cotton SNARE complex component GhSYP121 regulates salicylic acid signaling during defense against Verticillium dahliae.

Syntaxin of plant (SYP) plays a crucial role in SNARE-mediated membrane trafficking during endocytic and secretory pathways, contributing to the regulation and execution of plant immunity against pathogens. Verticillium wilt is among the most destructive fungal diseases affecting cotton worldwide. However, information regarding SYP family genes in cotton is scarce. Through genome-wide identification and transcriptome profiling, we identified GhSYP121, a Qa SNARE gene in Gossypium hirsutum. GhSYP121 is notably induced by Verticillium dahliae, the causal agent of Verticillium wilt in cotton, and…

The Bacterial Volatile Organic Compound N,N-Dimethylhexadecylamine Induces Long-Lasting Developmental and Immune Responses throughout the Life Cycle of Arabidopsis thaliana.

N,N-dimethylhexadecylamine (DMHDA) is a bacterial volatile organic compound that affects plant growth and morphogenesis and is considered a cross-kingdom signal molecule. Its bioactivity involves crosstalk with the cytokinin and jasmonic acid (JA) pathways to control stem cell niches and induce iron deficiency adaptation and plant defense. In this study, through genetic analysis, we show that the DMHDA-JA-Ethylene (ET) relations determine the magnitude of the defensive response mounted during the infestation of Arabidopsis plants by the pathogenic fungus Botrytis cinerea. The Arabidopsis mutan

The RXLR Effector PcAvh1 Is Required for Full Virulence of Phytophthora capsici.

Plant pathogens employ diverse secreted effector proteins to manipulate host physiology and defense in order to foster diseases. The destruc … amiana attenuates resistance to P. capsici and results in dwarfism, suggesting that PP2Aa regulates plant immunity and growth. Collectively, these results suggest that PcAvh1 contributes to P. capsici infection, probably through its interaction with host PP2Aa.

miR403a and SA Are Involved in NbAGO2 Mediated Antiviral Defenses Against TMV Infection in Nicotiana benthamiana.

RNAi (RNA interference) is an important defense response against virus infection in plants. The core machinery of the RNAi pathway in plants include DCL (Dicer Like), AGO (Argonaute) and RdRp (RNA dependent RNA polymerase). Although involvement of these RNAi components in virus infection responses was demonstrated in Arabidopsis thaliana, their contribution to antiviral immunity in Nicotiana benthamiana, a model plant for plant-pathogen interaction studies, is not well understood. In this study, we investigated the role of N. benthamiana NbAGO2 gene against TMV (Tomato mosaic virus) infection.
