Effects of Piperine on Obese and Lean Nonalcoholic Fatty Liver Disease in Mice.

Nonalcoholic fatty liver disease (NAFLD) is commonly associated with overweight and obesity. But an increasing number of cases involve lean individuals, who tend to experience worse liver outcomes. In this study, we established mouse models of both obese and lean NAFLD by feeding mice a Western diet (WD) ad libitum and a WD with 30% caloric restriction (CR), respectively, to investigate the effects of piperine. After 12 weeks, hepatocyte lipid deposition and liver injury were similar in lean and obese NAFLD mice. Piperine treatment significantly reduced serum ALT levels and hepatic triglycerid

Synthesis and performance optimization of multi-bioactive compound-loaded nanocarriers for antimicrobial applications.

Curcumin (Cur), quercetin (Que), and piperine (Pip) are well-known plant-derived compounds with diverse pharmacological activities and health benefits. However, their clinical application is limited due to hydrophobicity and poor bioavailability. Polylactic-co-glycolic acid nanoparticles(PLGA NPs) provide efficient drug loading and sustained release, making them an ideal platform for controlled drug delivery. Cur, Que, and Pip co-loaded PLGA NPs (Cur-Que-Pip-PLGA NPs) were prepared using the emulsification-solvent evaporation method. Their physicochemical properties were characterized by trans

Structure optimization of natural product piperine to obtain novel and potent analogs with anti-inflammation pain and urate-lowering effect.

Hyperuricemia is a metabolic disorder syndrome caused by a disorder of purine metabolism in the body, followed by urate crystal deposition leading to gouty arthritis, urate nephropathy, and kidney stones, collectively known as gout. Promoting uric acid stone dissolution by continuously reducing blood uric acid levels and reducing acute gout attacks in patients by controlling inflammatory pain reactions are identified as a potential therapy for gout. Starting from the natural product piperine, three analogs of novel piperine derivates were designed and synthesized to improve the anti-inflammati

Selective Cytotoxicity of Piperine Over Multidrug Resistance Leukemic Cells.

Multidrug resistance (MDR) is the main challenge in the treatment of chronic myeloid leukemia (CML), and P-glycoprotein (P-gp) overexpression is an important mechanism involved in this resistance process. However, some compounds can selectively affect MDR cells, inducing collateral sensitivity (CS), which may be dependent on P-gp. The aim of this study was to investigate the effect of piperine, a phytochemical from black pepper, on CS induction in CML MDR cells, and the mechanisms involved. The results indicate that piperine induced CS, being more cytotoxic to K562-derived MDR cells (Lucena-1

Role of endocannabinoid signalling in the dorsolateral periaqueductal grey in the modulation of distinct panic-like responses.

Panic attacks, a major feature of panic disorder, can be modelled in rats by exposing animals to stimuli that induce escape reactions, such as the elevated T-maze or the activation of the dorsolateral periaqueductal grey. Since the cannabinoid CB1 receptor modulates various types of aversive responses, this study tested the hypothesis that enhancement of endocannabinoid signalling in the dorsolateral periaqueductal grey inhibits panic-like reactions in rats. Local injection of the CB1 agonist, arachidonoyl 2-Chloroethylamide (0.005-0.5 pmol), attenuated the escape response from the open arm of

TRPV4: an exciting new target to promote alveolocapillary barrier function.

Transient receptor potential (TRP) channels are emerging as important players and drug targets in respiratory disease. Amongst the vanilloid-type TRP channels (which includes the six members of the TRPV family), target diseases include cough, asthma, cancer, and more recently, pulmonary edema associated with acute respiratory distress syndrome. Here, we critically evaluate a recent report that addresses TRPV4 as a candidate target for the management of acute lung injury that develops as a consequence of aspiration of gastric contents, or acute chlorine gas exposure. By use of two new TRPV4 inh

Involvement of serotonergic system in the antidepressant-like effect of piperine.

Piperine is a major alkaloid of black pepper (Piper nigrum Linn.) and long pepper (P. longum Linn.), and its antidepressant-like effect has been previously demonstrated. The purpose of this study was to explore the possible contribution of the serotonergic system in the antidepressant-like effect of piperine in mice. The results showed that piperine significantly reduced the immobility time in the forced swim test and tail suspension test in mice. The anti-immobility effect of piperine in the forced swim test and tail suspension test was completely abolished by pre-treating the mice with pCPA

Effect of anaesthesia and cardiopulmonary bypass on blood endocannabinoid concentrations during cardiac surgery.

BACKGROUND: The endocannabinoid system (ECS) is an endogenous signalling system which includes the endocannabinoids anandamide (AEA) and 2-arachidonoylglycerol (2-AG) and specific G-protein-coupled endocannabinoid receptors (CB1 and CB2). Recent studies have described important roles of the peripheral ECS in human atherosclerosis, cardiometabolic disorders, heart failure, and systemic inflammation. We sought to study changes in plasma endocannabinoid concentrations during cardiac surgery (CS) under general anaesthesia with isoflurane/sufentanil, and during cardiopulmonary bypass (CPB). METHOD

TRPV1 is involved in stretch-evoked contractile changes in the rat autonomous bladder model: a study with piperine, a new TRPV1 agonist.

AIM: Vanilloids like capsaicin and resiniferatoxin (RTX) have been used for more than a decade in the treatment of neurogenic detrusor overactivity. Recently, the vanilloid molecule piperine (PIP) has been shown to have similar pharmacological properties as these drugs. In this study, we looked at PIP-effects on autonomous bladder contractile activity, with particular interest for its selectivity for the transient receptor potential channel 1 (TRPV1) receptor. Additionally, we studied the role of TRPV1 in volume-induced contractile changes using selective and non-selective TRPV1 antagonists.

UCM707, a potent and selective inhibitor of endocannabinoid uptake, potentiates hypokinetic and antinociceptive effects of anandamide.

To date, UCM707, N-(3-furylmethyl)eicosa-5,8,11,14-tetraenamide, has the highest potency and selectivity in vitro as inhibitor of the endocannabinoid transporter, which might make this compound useful in potentiating endocannabinoid transmission, with minimal side-effects, in the treatment of several disorders. However, there is no information about how UCM707 behaves in vivo as regards certain classic effects of endocannabinoids, such as hypomotility and antinociception. In the present work, we tested in rats the dose-response effects of UCM707 in the open-field and hot-plate tests, and, in p

Piperine reverses colistin resistance in multidrug resistant Gram-negative pathogens by membrane disruption and ROS damage.

The global spread of colistin (COL) resistance, particularly mediated by mcr-1, threatens the efficacy of this last-line antibiotic against multidrug-resistant (MDR) Gram-negative pathogens. This development underscores the urgency of developing innovative therapeutic strategies to counteract antimicrobial resistance mechanisms. This study demonstrates that piperine (PIP), a natural alkaloid, functions as a potent adjuvant that restores COL efficacy through a multi-target mechanism. The combination of PIP and COL exhibits significant synergism against clinically relevant pathogens, including m

Improving Paclitaxel permeation and anticancer efficacy through Soluplus-based solid dispersion with Piperine.

OBJECTIVE: This study aimed to develop and optimize a Soluplus-based Paclitaxel-Piperine (PTX-PPN) solid dispersion to enhance PTX solubility, intestinal permeation, and anticancer efficacy in metastatic breast cancer cells. SIGNIFICANCE: PTX suffers from extremely poor aqueous solubility and dose-limiting toxicity associated with Cremophor® EL-based formulations. PPN is a well-documented bioenhancer and P-glycoprotein inhibitor that may modulate PTX permeation and cytotoxicity. METHODS: PTX-PPN solid dispersions were prepared via freeze-drying using different PTX-PPN-Soluplus ratios (1:1:1,

Neuroprotective Role of Piperine-Encapsulated Casein Micelles in Mitigating Intracellular Dyshomeostasis in Alzheimer's Disease Pathology.

Alzheimer's disease remains a complex neurodegenerative disorder characterized by multifactorial mechanisms that undermine the efficacy of monotherapeutic strategies. Multi-targeted therapies have emerged as promising strategies, particularly those addressing mitochondrial dysfunction, a key contributor to AD pathophysiology. This study explores the neuroprotective efficacy of piperine encapsulated casein micelles (PIP@CMs) against Aβ(1-42)-induced-neurotoxicity in differentiated SH-SY5Y cells, an established in vitro model of Alzheimer's pathology. The formulation was synthesized and characte

Cannabinoid Receptor-2 Activation in Keratinocytes Contributes to Elevated Peripheral β-Endorphin Levels in Patients With Obstructive Jaundice.

BACKGROUND: Cholestatic diseases are often accompanied by elevated plasma levels of endogenous opioid peptides, but it is still unclear whether central or peripheral mechanisms are involved in this process, and little is known about the change of pain threshold in these patients. The purpose of this study was to determine the preoperative pain threshold, postoperative morphine consumption, and central and peripheral β-endorphin levels in patients with obstructive jaundice. This study also tests the hypothesis that activation of the cannabinoid receptor-2 (CB2R) in skin keratinocytes by endocan

Essential fatty acids deficient diet modulates N-Acylethanolamide profile in rat's tissues.

No data are available on whether a diet deficient of the essential fatty acids is able to modulate tissue levels of endocannabinoids and congeners. Male rats fed for 12 weeks a diet deficient of essential fatty acids, palmitic and oleic acids (EFAD), replaced with saturated fatty acids (SAFA), showed lowered n-3 and n-6 PUFAs levels in plasma, liver and adipose tissue, with concomitant steep increase of oleic and mead acids, while in hypothalamus no changes in PUFA concentration were detected and only palmitoleic acid was found increased. We found a reduction of anandamide and palmitoylethanol

Innovative Therapeutic Potential of Cannabinoid Receptors as Targets in Alzheimer's Disease and Less Well-Known Diseases.

The discovery of cannabinoid receptors at the beginning of the 1990s, CB1 cloned in 1990 and CB2 cloned in 1993, and the availability of selective and potent cannabimimetics could only be justified by the existence of endogenous ligands that are capable of binding to them. Thus, the characterisation and cloning of the first cannabinoid receptor (CB1) led to the isolation and characterisation of the first endocannabinoid, arachidonoylethanolamide (AEA), two years later and the subsequent identification of a family of lipid transmitters known as the fatty acid ester 2-arachidonoylglycerol (2-AG)

Sex-specific autistic endophenotypes induced by prenatal exposure to valproic acid involve anandamide signalling.

BACKGROUND AND PURPOSE: Autism spectrum disorder (ASD) is more commonly diagnosed in males than in females. Prenatal exposure to the antiepileptic drug valproic acid (VPA) is an environmental risk factor of ASD. Male rats prenatally exposed to VPA show socio-emotional autistic-like dysfunctions that have been related to changes in the activity of the endocannabinoid anandamide. Here, we have investigated if prenatal VPA induced sex-specific autistic endophenotypes involving anandamide signalling. EXPERIMENTAL APPROACH: We studied sex-specific differences in the ASD-like socio-emotional, cogni
