Characterization of biochemical properties of a selenium-independent glutathione peroxidase of Cryptosporidium parvum.

Glutathione peroxidase (GPx; EC 1.11.1.9) is an important antioxidant enzyme that catalyses the reduction of organic and inorganic hydropero … activity against inorganic hydroperoxide, hydrogen peroxide. Recombinant CpGPx was not inhibited by potassium cyanide or by sodium azide. The enzyme effectively protected DNA and protein from oxidative damage induced by hydrogen peroxide, and was functionally expressed in various developmental stages of C. parvum. These results collectively suggest the essential ro …

Mechanism of antagonizing cyanide-induced lethality by alpha-ketoglutaric acid.

alpha-Ketoglutaric acid (alpha-KG) has been shown to be an effective antagonist for cyanide-induced lethality. The mechanism of this antagonism is hypothesized to result from alpha-KG binding with cyanide. Several investigative approaches were taken to determine the existence of this binding. First, mixtures of various molar ratios of alpha-KG:cyanide were injected into a high pressure liquid chromatograph. The addition of cyanide reduced the peak area of alpha-KG at a molar ratio of greater than 1:5. Second, blood from naive male ICR mice was spiked with alpha-KG and cyanide. Headspace above

The hepatic glycogenolysis induced by reversible ischaemia or KCN is exclusively catalysed by phosphorylase a.

1. Ischaemia was applied for 30 min to the liver of Wistar rats and of gsd/gsd rats, which have a genetic deficiency of phosphorylase kinase. The rate of glycogenolysis corresponded closely to the concentration of phosphorylase a. The loss of glycogen from Wistar livers was accounted for by the intrahepatic increase in glucose plus lactate. Further, the accumulation of oligosaccharides was negligible in the gsd/gsd liver. 2. Isolated hepatocytes from Wistar and gsd/gsd rats were incubated for 40 min in the presence of either KCN or glucagon. Again, the production of glucose plus lactate was st

Low-spin ferric forms of cytochrome a3 in mixed-ligand and partially reduced cyanide-bound derivatives of cytochrome c oxidase.

Optical-absorption-, e.p.r.- and m.c.d. (magnetic-circular-dichroism)-spectroscopic measurements were made on liganded derivatives of oxidized and partially reduced cytochrome c oxidase. When NO was added to oxidized cyanide-bound cytochrome c oxidase, no changes occurred in the optical-absorption difference spectrum. In contrast, NO induced reduction of cytochrome a3 and formation of the nitrosylferrohaem species when the oxidized resting enzyme was the starting material. E.p.r. spectroscopy of the NO-treated oxidized cyanide-bound enzyme revealed the presence of a low-spin haem signal at g =

Effect of citrate on the reactions of cytochrome c with reductants and cyanide.

The rate of reduction of cytochrome c by ascorbate is decreased in the presence of anions. This decrease is due to two factors: (a) nonspecific changes in ionic strength which occur when the total ion concentration or the charge on the anion is altered and (b) "specific" binding of anions to a site on cytochrome c which directly inhibits the reaction. The reaction between cyanide ion and cytochrome c is also affected by anions: increasing the ionic strength decreases the apparent association rate constant for cyanide binding. Substitution of citrate for morpholinopropane sulphonate in isoionic

Cysteine residues in the C-terminal tail of the human BK(Ca)alpha subunit are important for channel sensitivity to carbon monoxide.

In the presence of oxygen (O(2)), carbon monoxide (CO) is synthesised from heme by endogenous hemeoxygenases, and is a powerful activator of … ansiently transfected into HEK 293 cells before subsequent analysis in inside-out membrane patches. Potassium cyanide (KCN) completely abolished activation of wild type BK(Ca) channels by the CO donor, tricarbonyldichlororuthenium (II) dimer, at 100microM. In the absence of KCN the CO donor increased wild-type channel activity in a concentration-dependent manner, w …

Characterization of MXR activity in the sea anemone Bunodosoma cangicum exposed to copper.

Transmembrane proteins of the ABC family contribute to a multiple xenobiotic resistance (MXR) phenotype in cells, driving the extrusion of t … ecreased in treatment groups exposed to copper concentrations of 15.6μg/L and 10mM energy depleting potassium cyanide. Metabolic activity increased in cells exposed to 7.8μgCu/L, but 15.6μgCu/L was similar to 0 and 7.8μg/L. The presence of copper decreased the ABC proteins expression. The present study improves the knowledge of MXR in anemone cells and shows that t …

Thermodynamic investigation by heat capacity measurements of ferrimagnetic A2Mn[Mn(CN)6] (A=K, Rb, Cs) Prussian blue compounds.

Heat capacity measurements of a new series of Prussian blue analogs of A2Mn[Mn(CN)6] (A=K, Rb, Cs) composition were performed using thermal relaxation calorimetry. The Cs compound has a face-centered cubic structure with a linear Mn-C≡N-Mn linkage, while the monoclinic Rb and K compounds have nonlinear Mn-C≡N-Mn linkages. For all of the compounds, large broad thermal anomalies associated with magnetic transitions were observed in the temperature dependence of the heat capacity. The systematic changes in the heat capacity for the three compounds under magnetic fields of up to 7 T were found to

Mitochondrial phospholipid hydroperoxide glutathione peroxidase plays a major role in preventing oxidative injury to cells.

Phospholipid hydroperoxide glutathione peroxidase (PHGPx) is synthesized as a long form (L-form; 23 kDa) and a short form (S-form; 20 kDa). … etected in L9 cells that overexpressed the S-form. Cell death caused by mitochondrial injury due to potassium cyanide (KCN) or rotenone (chemical hypoxia) was considerably suppressed in the M15 cells, whereas the L9 cells and control RBL-2H3 cells (S1 cells, transfected with the vector alone) succumbed to the cytotoxic effects of KCN. Flow cytometric analysis showe …

Circulatory Failure During Noninhaled Forms of Cyanide Intoxication.

Our objective was to determine how circulatory failure develops following systemic administration of potassium cyanide (KCN). We used a noninhaled modality of intoxication, wherein the change in breathing pattern would not influence the diffusion of CN into the blood, akin to the effects of ingesting toxic levels of CN. In a group of 300 to 400 g rats, CN-induced coma (CN i.p., 7 mg/kg) produced a central apnea within 2 to 3 min along with a potent and prolonged gasping pattern leading to autoresuscitation in 38% of the animals. Motor deficits and neuronal necrosis were nevertheless observed i…

Lactating mouse mammary cell uptake of 59Fe-transferrin. The effect of KCN, 2,4-dinitrophenol, ethanol and ATP.

1. Amounts of KCN of 0.2, 1.0 and 2.0 X 10(-11) mol in a probe do not prevent the uptake of transferrin iron from lactating mouse mammary gland cells. 2. The effect of 5, 10 and 100 X 10(-11) mol in a probe 2,4-dinitrophenol dissolved in 20% ethanol is also negative on the iron-transferrin uptake from the cell. 3. There is a statistically significant decrease of uptake of transferrin-iron under the influence of 20% ethanol in the incubation mixture (25, 50 and 100 microliters). The effect of ethanol is diminished by 2,4-dinitrophenol in the solution. 4. The uptake of 59Fe-transferrin from cell

Effects of temperature, pH elevators, and energy production inhibitors on horseradish peroxidase transport through endocytic vesicles.

We investigated the effects of reduced temperature, the pH elevators NH4Cl, monensin, and HEPES (N-2-hydroxy-ethylpiperazine-N'-2-ethanesulfonic acid) buffer, as well as the metabolic poisons NaF/KCN on transport of the fluid phase pinocytic marker, horseradish peroxidase (HRP), to lysosomes in Chinese hamster ovary (CHO) cells. In cell fractionation experiments, these agents appeared to block HRP transit at specific point(s) from "early" to "late" (i.e., low to high density) prelysosomal vesicles and lysosomes. Reduced temperature (17 degrees C) most strongly inhibited HRP transport from low

Oxidative interactions between fatty acid peroxy radicals and quinones: possible involvement in cyanide-resistant electron transport in plant mitochondria.

The interactions between duroquinol, linoleic acid, and lipoxygenase have been followed spectrophotometrically in the uv (210-340 nm) in a simple reaction medium (5 mM [4-(2-hydroxyethyl)-1-piperazineethanesulfonic acid] buffer, pH 6.0). Duroquinol is oxidized by reacting with the peroxy radicals of linoleic acid generated in the presence of lipoxygenase. This oxidation is insensitive to cyanide but is sensitive to salicylhydroxamic acid, propylgallate, and disulfiram, the known inhibitors of lipoxygenase and of the cyanide-resistant electron transport pathway of plant mitochondria. This react

Involvement of cytochrome b5 and a cyanide-sensitive monooxygenase in the 4-demethylation of 4,4-dimethylzymosterol by yeast microsomes.

According to Ohba et al. (Ohba, M., Sato, R., Yoshida, Y., Nishino, T. and Katsuki, H. (1978) Biochem. Biophys. Res. Commun. 85, 21-27), yeast microsomes catalyze the removal of three methyl groups attached to the C-4 and C-14 positions of [1,7,15,22,26,30-14C]lanosterol (4,4,14 alpha-trimethyl-5 alpha-cholesta-8,24-dien-3 beta-ol) in the presence of NADPH, NAD+ and molecular oxygen, concomitant with the liberation of 14CO2 derived from C-30 (one of the two methyl groups at the C-4 position). In this process the methyl group at the C-14 position is first removed in a cyanide-insensitive reacti

Glia activation and its role in oxidative stress.

Glia activation and neuroinflamation are major factors implicated in the aetiology of most neurodegenerative diseases (NDDs). Several agents and toxins have been known to be capable of inducing glia activation an inflammatory response; most of which are active substances that can cause oxidative stress by inducing production of reactive oxygen species (ROS). Neurogenesis on the other hand involves metabolic and structural interaction between neurogenic and glia cells of the periventricular zone (PVZ); a region around the third ventricle. This study investigates glia activation (GFAP), cell pro
