Acute nicotine affects auditory brainstem responses in a frequency-, age-, and sex-dependent manner in mice.

Age-related hearing loss (presbycusis) is a widespread sensory disorder associated with diminished quality of life and increased risk of cognitive decline, with limited treatment options. While nicotinic acetylcholine receptor modulation is a potential treatment for improving auditory function, it is unclear if nicotine affects auditory responses in an age- and sex-dependent manner. To test this, we recorded tone-evoked auditory brainstem responses (ABR) from young (2-3 months) and old (16-22 months) male and female mice (FVB strain) following an injection of saline or nicotine (0.5 mg/kg, i.p…

Sudden hearing loss in the elderly.

In elderly subjects suffering from presbycusis a clinical picture of sudden hearing loss, tinnitus and vertigo may be precipitated by a superimposed serous otitis media. Such an abrupt and dramatic sensory deficiency may be erroneously ascribed to an acute cochlear lesion and given grave prognostication. This situation was observed in 12 patients during a period of 2 years. Appropriate diagnostic evaluation revealed the true nature of the disease and simple therapeutic measures promptly restored the auditory function to its previous level.

[Socio-presbyacusis: definition and prevention].

Socio-presbyacusis is a hot topic with an increasing incidence and high social costs. The main etiological factors are: genetics, smoking and drinking habits, diet, metabolic syndrome, chronic otitis and noise exposure (professional, environmental and recreative). Despite its multifactorial origin, the pathogenetic mechanism of cochlear damage is based on the increased production of Reactive Oxygen Species (ROS). The clinical characteristic feature of socio-presbyacusis is represented by slow cochlear degeneration causing a reduction of central hearing inputs. The result is a gradual social is

Event-Related Potentials as Markers of Age-Related Changes of Spatial Hearing in Normal-Hearing Adults.

Previous studies have shown that older adults can witness a decline in the integration of their spatial auditory cues even when they don't present pathological audiometric thresholds. Nonetheless, when spatial hearing in aging is discussed, most of the focus concerns elderly individuals with hearing impairments or presbycusis, rather than those with clinically normal hearing. In fact, auditory aging (even without presbycusis) may still result in a decline in spatial sensitivity, potentially contributing to the deterioration of key binaural functions crucial for speech understanding, such as bi…

cAMP-MFN2 signaling suppresses cochlear cell senescence and age-related hearing loss.

INTRODUCTION: Age-related hearing loss (presbycusis) is the most common sensory deficit in the elderly, yet it lacks effective pharmacological treatments. The decline of the cyclic AMP (cAMP) signaling pathway is implicated in aging, but its functional role in the auditory system remains largely unknown. METHODS: To investigate whether enhancing cAMP signaling could counteract presbycusis and elucidate the underlying molecular mechanism, we used a D-galactose-induced cellular senescence model and an in vivo aged mouse model. The effects of the cAMP analog dibutyryl-cAMP (dbcAMP) on cellular s…

Ultrastructural Synaptic Differences in the Central Inferior Colliculus in the 3xTG Mouse Across Three Disease Stages.

Numerous studies support a mechanistic link between hearing loss and increased risks of cognitive decline and dementia. Hearing loss is widely viewed as a modifiable risk factor for Alzheimer's disease (AD). During normal aging the inferior colliculus (IC), a large auditory midbrain nucleus, undergoes numerous changes to neurotransmission, and these changes contribute to the development of central gain and presbycusis. Recent reports also implicate the IC as a nucleus that undergoes processing changes during AD. We used transmission electron microscopy (EM) to examine the synaptic ultrastructu…

Insights into early cochlear damage induced by potassium channel deficiency.

Hearing loss (HL) is the most common sensory disorder, caused by genetic mutations and acquired factors like presbycusis and noise exposure. A critical factor in HL development is the dysfunction of potassium (K+) channels, essential for sensory cell function in the organ of Corti (OC). Inner and outer hair cells (IHCs and OHCs) convert sound into electrical signals, while supporting cells (SCs) maintain ionic and structural balance. KCNQ4 channels, located in the basal membrane of OHCs, regulate K+ efflux. Mutations in KCNQ4 are linked to progressive HL (DFNA2), noise-induced hearing loss, an…

The reverse stethoscope technique: a rapid intervention for old age hearing loss.

OBJECTIVES: Presbycusis is highly prevalent, affecting between a third and two-thirds of elderly populations. Effective communication between patient and physician has been shown to directly correlate with the quality of patient care. The Reverse Stethoscope Technique (RST) involves placing the earpieces into the patient's ears and speaking into the diaphragm. Here, we aim to show the RST is a simple and effective method to communicate with patients suffering from presbycusis in inpatient settings. METHODS: Medical inpatients aged >60 years old without cognitive impairment were included in th…

Preventive Effect of Cocoa Flavonoids via Suppression of Oxidative Stress-Induced Apoptosis in Auditory Senescent Cells.

Presbycusis or Age-related hearing loss (ARHL) is a sensorineural hearing loss that affects communication, leading to depression and social isolation. Currently, there are no effective treatments against ARHL. It is known that cocoa products have high levels of polyphenol content (mainly flavonoids), that are potent anti-inflammatory and antioxidant agents with proven benefits for health. The objective is to determine the protective effect of cocoa at the cellular and molecular levels in Presbycusis. For in vitro study, we used House Ear Institute-Organ of Corti 1 (HEI-OC1), stria vascularis (…

Inputs to a physiologically characterized region of the inferior colliculus of the young adult CBA mouse.

Presbycusis is a sensory perceptual disorder involving loss of high-pitch hearing and reduced ability to process biologically relevant acoustic signals in noisy environments. The present investigation is part of an ongoing series of studies aimed at discerning the neural bases of presbycusis. The purpose of the present experiment was to delineate the inputs to a functionally characterized region of the dorsomedial inferior colliculus (IC, auditory midbrain) in young, adult CBA mice. Focal, iontophoretic injections of horseradish peroxidase were made in the 18-24 kHz region of dorsomedial IC of…

Electron microscopic findings in presbycusic degeneration of the basal turn of the human cochlea.

Three human temporal bones with presbycusis affecting the basal turn of the cochlea were studied by light and electron microscopy. Conditions in two ears examined by light microscopy were typical of primary neural degeneration, with a descending audiometric pattern, loss of cochlear neurons in the basal turn, and preservation of the organ of Corti. Ultrastructural analysis revealed normal hair cells and marked degenerative changes of the remaining neural fibers, expecially in the basal turn. These changes included a decrease in the number of synapses at the base of hair cells, accumulation of …

Central auditory decline precedes cochlear deficits in a D-galactose mimetic model of aging.

Age-related hearing loss reflects a mixture of concurrent peripheral cochlear and central auditory pathway degeneration. Disentangling their … lds, reduced peripheral wave amplitudes, and reduced EFRs at fast modulation rates, consistent with presbycusis. These findings demonstrate a clear temporal dissociation in which central auditory decline in the D-gal model precedes age-related peripheral cochlear and neural loss.
