Quantitative phosphoproteomics profiling reveals RNA binding proteins GhRS2Z32 and GhGRP7 response to cold stress in cotton.

Cold stress severely restricts the growth and development of cotton. Plants must adjust their developmental and physiological processes to cope with such stress. Protein phosphorylation, a vital reversible post-translational modification, dominates intracellular signal transduction upon environmental stimuli. In this study, we analyzed the phosphoproteome of cotton under cold stress to delineate how phosphorylation modifications mediate cold responses. Phosphoproteomic analysis showed that differentially phosphorylated proteins were significantly enriched in pathways associated with post-trans

SIRT5-mediated desuccinylation of MTHFD2 enhances chemoresistance in breast cancer cells by reducing therapy-induced senescence.

Protein lysine succinylation is a crucial post-translational modification that regulates nearly all aspects of eukaryotic and prokaryotic cell, including gene transcription, cell metabolism and redox homeostasis. Among them, metabolic disorders caused by dysfunctional post-translational modifications induce aging and aged-related diseases, including cancer. This study quantified the dynamic changes in protein succinylation in response to DNA damage stress induced by etoposide (ETOP) in tumor cells. A total of 4354 lysine succinylation sites on 1259 proteins were identified, many of which have

Increased susceptibility to organic dust exposure-induced inflammatory lung disease with enhanced rheumatoid arthritis-associated autoantigen expression in HLA-DR4 transgenic mice.

Immunogenetic as well as environmental and occupational exposures have been linked to the development of rheumatoid arthritis (RA), RA-associated lung disease, and other primary lung disorders. Importantly, various inhalants can trigger post-translational protein modifications, resulting in lung autoantigen expression capable of stimulating pro-inflammatory and/or pro-fibrotic immune responses. To further elucidate gene-environment interactions contributing to pathologic lung inflammation, we exploited an established model of organic dust extract (ODE) exposure with and without collagen-induce

Vitamin D-binding protein as a biomarker of active disease in acute intermittent porphyria.

UNLABELLED: Acute intermittent porphyria (AIP) is an autosomal dominant metabolic disorder caused by a deficiency of hepatic porphobilinogen deaminase (PBGD). The disease is characterized by life threatening acute neurovisceral attacks. The aim of this study was to identify metabolites secreted by the hepatocytes that reflect differential metabolic status in the liver and that may predict response to the acute attack treatment. Plasma vitamin D binding protein (VDBP) from a mouse model of AIP displayed an abnormal migration in 2D-electrophoresis that is efficiently recovered upon gene therapy

Oligosaccharide structures of von Willebrand factor and their potential role in von Willebrand disease.

Oligosaccharides make up approximately 20% of the mass of VWF and although their structures are well established, their functional role remains unclear. Modification of the VWF oligosaccharide structures has been shown to result in increased plasma clearance of the protein. A mutation which alters cell type-specific expression of the Galgt2 glycosyltransferase gene in the RIIIS/J mouse results in an autosomal dominant partial quantitative deficiency of VWF. Increased plasma clearance of VWF has been demonstrated in some individuals with a partial quantitative deficiency of the protein and it i

A Bird's-Eye View of the Wings Apart-Like Protein in Cell Biology and Implications for Disease and Therapeutics.

Wings apart-like protein (WAPL) has emerged as a key player in maintaining genome integrity through its regulation of cohesin dynamics, which govern chromatin architecture and gene expression. WAPL mainly acts as a cohesin release factor and ensures proper chromosomal segregation during mitosis by promoting sister chromatid resolution. Owing to its prominent role in cell biology, WAPL dysregulation can cause genomic instability and disrupt chromosomal cohesion, leading to diseases such as cancer. The precise regulatory mechanisms controlling WAPL activity, including post-translational modifica

Molecular mechanisms and functions of protein acetylation in sepsis and sepsis-associated organ dysfunction.

Sepsis, a life-threatening condition characterized by organ dysfunction due to dysregulated host response to infection, remains a global health challenge with high morbidity, mortality, and long-term sequelae. The development of sepsis-associated organ dysfunction (SAODs) substantially worsens prognosis. Despite extensive studies, the pathophysiological mechanisms underlying sepsis and SAODs remain unclear. Protein acetylation is a widespread and reversible post-translational modification regulated by acetyltransferases and deacetylases that occurs on both histone and non-histone proteins. Thi

Thioredoxin and ventricular remodeling.

Increasing bodies of evidence indicate that reactive oxygen species (ROS) produced by mitochondria and other sources play an essential role in mediating ventricular remodeling after myocardial infarction and the development of heart failure. Antioxidants scavenge ROS, thereby maintaining the reduced environment of cells and inhibiting ventricular remodeling in the heart. Thioredoxin not only functions as a major antioxidant in the heart but also interacts with important signaling molecules and transcription factors, thereby modulating various cellular functions. The activity of thioredoxin is

Single-nucleotide RNA m6A mapping in bovine preimplantation development reveals site-specific regulation of RPL12 at zygotic genome activation.

RNA N6-methyladenosine (m6A) is a key regulator of gene expression during early embryogenesis. Using SAC-seq (m6A-selective allyl chemical labeling and sequencing), an antibody-independent m6A profiling method, we generated the first single-nucleotide-resolution m6A map of bovine oocytes and preimplantation embryos. We observed both coordinated and uncoupled relationships between m6A modification and expression of protein-coding and noncoding genes. Integrative analysis of the transcriptome, m6A epitranscriptome, and translatome revealed dynamic m6A remodeling, particularly in ribosomal protei

Chemical Synthesis of Ubiquitinated Proteins and Ubiquitin Probes for Biochemical and Functional Analysis.

Protein ubiquitination is a pivotal post-translational modification that plays vital roles in eukaryotic cellular regulation. Ubiquitination is orchestrated by a cascade of enzymes: E1 activating enzymes, E2 conjugating enzymes, and E3 ligases collectively facilitate ubiquitin conjugation, while ubiquitin-binding proteins interpret the ubiquitin signal. Deubiquitinases (DUBs) counterbalance this process by removing ubiquitin from substrate proteins, thereby reversing ubiquitination. In recent years, chemical protein synthesis-enabling the preparation of proteins with atomic-level precision-has

Development of a Chemoproteomic Platform to Identify Sites of (Homo)citrullination within Complex Proteomes.

Citrullination and homocitrullination of arginine and lysine can significantly impact protein structure and function. Citrullination of arginine is an enzymatic modification catalyzed by Protein Arginine Deiminases (PADs). Homocitrullination of lysine is a nonenzymatic modification that occurs in the presence of high concentrations of cyanate. Both post-translational modifications are elevated in Rheumatoid Arthritis (RA) and other inflammatory diseases. Moreover, autoantibodies targeting these PTMs are associated with the development of RA. Identifying arginine and lysine residues that are hy

The role of S-palmitoylation of C4BPA in regulating murine sperm motility and complement resistance.

The epididymis and epididymosomes are crucial for regulating sperm motility, a key factor in male fertility. Palmitoylation, a lipid modification involving the attachment of palmitic acid to cysteine residues, is essential for protein function and localization. Additionally, this modification plays a vital role in the sorting of proteins into exosomes. This study investigates the role of S-palmitoylation at the Cys15 residue of the C4b binding protein alpha chain (C4BPA) in murine sperm motility. Our findings revealed high expression of C4BPA mRNA in the caput epididymis, with the protein pres

SUMOylation of SIRT1 activating PGC-1α/PPARα pathway mediates the protective effect of LncRNA-MHRT in cardiac hypertrophy.

Long noncoding RNA-Myosin heavy chain associated RNA transcript (LncRNA-MHRT) has been reported to prevent pathological cardiac hypertrophy. However, the underlying inhibition mechanism has not been fully elucidated. Further, whether MHRT inhibits hypertrophy by regulating post-translational modification of certain proteins remains unclear. Therefore, this study aims to find potential role of MHRT in inhibiting cardiac hypertrophy via regulating modification of certain proteins. Here, Angiotensin II (Ang II) -treated neonatal rat cardiomyocytes and transverse aortic constriction (TAC) mice wer

An Overview of Mass Spectrometry-Based Methods to Analyze Post-Translational Modifications.

Post-translational modifications (PTM) are the covalent modifications of proteins after their biosynthesis. PTMs change the structure and functions of proteins, which eventually regulate various biological processes that are closely related to the onset and development of diseases. Hundreds of different forms of PTMs have been discovered so far and mass spectrometry (MS) has become one of the most important tools to analyze PTMs. With the development of new methods in sample preparation, advances in MS instrumentation, and inventions of new bioinformatic tools, MS-based methods can identify mo

What the kinome reveals about resistance to foodborne pathogens.

While the genetic similarities between vertebrates have been remarked on with each new complete genome sequenced, the physiological and phenotypic differences between agricultural species remain clear even to the casual observer. As one moves along the path of molecular biology, from DNA to RNA to protein, the distinctions between species increase. The separation between species further increases when the active sites of protein are considered. Phenotype is dictated by gene expression patterns, response to environmental stimuli, epigenetic alterations, protein translation, protein folding and

Hydrogen sulfide action in the regulation of plant autophagy.

Hydrogen sulfide is a signalling molecule with a well-established impact on both plant and animal physiology. Intense investigation into the regulation of autophagy by sulfide in Arabidopsis thaliana has revealed that the post-translational modification of persulfidation/S-sulfhydration plays a key role. In this review focused on plants, we discuss the nature of the sulfide molecule involved in the regulation of autophagy, the final outcome of this modification and the persulfidated autophagy proteins identified so far. A detailed outline of the actual knowledge of the regulation mechanism of
