Characterization of a novel epigenetic effect of ionizing radiation: the death-inducing effect.

The detrimental effects associated with exposure to ionizing radiation have long been thought to result from the direct targeting of the nucleus leading to DNA damage; however, the emergence of concepts such as radiation-induced genomic instability and bystander effects have challenged this dogma. After cellular exposure to ionizing radiation, we have isolated a number of clones of Chinese hamster-human hybrid GM10115 cells that demonstrate genomic instability as measured by chromosomal destabilization. These clones show dynamic and persistent generation of chromosomal rearrangements multiple

Prognostic relevance of previous exposure to ionizing radiation in well-differentiated thyroid cancer.

BACKGROUND AND AIMS: Although exposure to ionizing radiation is a well-known risk factor for well-differentiated thyroid cancer, its effects on tumor behavior remain unclear. This study was undertaken to address this question. MATERIALS AND METHODS: We randomly selected 426 patients who underwent thyroidectomy for cancer between 1964 and 2000 and divided them into two groups: previously exposed (ExR); not exposed to radiation (nExR). Data were retrospectively collected. RESULTS: There were 340 female patients and 86 male, age 9-89 years with mean follow-up of 56 months. The ExR group (n=68)

Dose-response relationship for parathyroid adenoma after exposure to ionizing radiation in infancy.

Several authors have suggested that there is an excess risk of hyperparathyroidism, adenomas or hyperplasia after exposure to ionizing radiation. There is still, however, some uncertainty about this association, because these diseases are often asymptomatic and escape clinical detection if not specially searched for. This study is based on a pooled Swedish cohort of 27,925 persons with skin hemangiomas. The majority received radiation treatment in infancy between 1920 and 1965 in Stockholm and Gothenburg. The mean age at treatment was 6 months and the median thyroid dose was 0.20 Gy (range 0-2

Sensitivity of nucleotide excision repair-deficient human cells to ionizing radiation and cyclophosphamide.

Nucleotide excision repair (NER)-deficient rodent and human cells (such as those derived from patients with xeroderma pigmentosum, XP) are hypersensitive to UV light. Some of these cell lines, specifically certain rodent mutants with severe defects in the ERCC1 and XPF genes, are dramatically sensitive to crosslinking agents such as phosphoramide mustard (PM). These crosslink-sensitive rodent mutants also exhibit sensitization to gamma-rays under hypoxic (but not under aerated) conditions. Like their rodent counterparts, human XP cells are highly sensitive to UV light; however, none of the hum

Dose dependence effects of ionizing radiation on bile acid metabolism in the rat.

PURPOSE: To study radiation dose-related changes of individual or total bile acids in various physiological fluids in order to identify potential bio-indicators of radiation-induced gastrointestinal injuries. MATERIALS AND METHODS: Wistar rats were sham- or whole-body gamma irradiated (1-12Gy). Total and individual bile acids were quantified, 3 days after exposure, in bile collected after catheterization of the bile duct. Total bile acid concentrations were also measured in plasma and colonic contents 1, 2 and 3 days after irradiation. These concentrations were determined by an enzymatic meth

Differing responses of Nijmegen breakage syndrome and ataxia telangiectasia cells to ionizing radiation.

Nijmegen breakage syndrome (NBS) is a rare autosomal recessive disorder. Originally thought to be a variant of ataxia telangiectasia (AT), the cellular phenotype of NBS has been described as almost indistinguishable from that of AT. Since the gene involved in NBS has been cloned and its functions studied, we sought to further characterize its cellular phenotype by examining the response of density-inhibited, confluent cultures of human diploid fibroblasts to irradiation in the G(0)/G(1) phase of the cell cycle. Both NBS and AT cells were markedly sensitive to the cytotoxic effects of radiation

Retrospective dosimetry based on cytogenetic study of individuals chronically exposed to ionizing radiation.

Analysis of chromosomal aberrations (CAA) was carried out in 27 radiation workers with cumulative exposures of approximately 500 mSv received over a period of 2-3 decades. A similar study was carried out in 20 age- and gender-matched healthy individuals who served as controls. The average age of the exposed group was 50 years, and that of the controls was 51 years. The absorbed radiation dose was calculated by using the linear component of the in vitro dose response curve established for 60Co gamma-rays. In the controls we found only 3 dicentrics in 5,500 metaphases analyzed. In the exposed gr

Ionizing radiation enhances double-strand-break repair in rapamycin-treated ataxia telangiectasia lymphoblasts.

PURPOSE: Unlike normal cells, ataxia telangiectasia (A-T) cells do not exhibit enhanced double-strand-break (dsb)-repair fidelity after ionizing radiation (IR) exposure. DNA-repair induction and other responses to IR are mediated by signalling pathways that are defective in A-T cells. Ataxia telangiectasia mutated (ATM) protein, the mutated protein in A-T cells, is homologous to members of the P13-kinase, including the rapamycin target FRAP. Rapamycin kills normal cells more readily in normal than in A-T cells, and inhibits the FRAP target p70 S6 kinase (p70S6K) more readily in normal than in

Is micronuclei yield variability a problem for overexposure dose assessment to ionizing radiation?

Scoring of micronuclei (Mni) in cytokinesis-blocked human blood peripheral lymphocytes after an accidental radiation overexposure appears an easier and faster alternative for biological dosimetry than dicentrics analysis. However, an increase of Mni rate could be difficult to interpret particularly at low doses, because of the known variability of individual dose response and the unknown background frequency. Moreover, in case of nuclear emergency, there are the added problems of large samples numbers for processing and so screening time. In this paper, we wish to propose some solutions using

Vascular activation of adhesion molecule mRNA and cell surface expression by ionizing radiation.

During cutaneous inflammatory reactions the recruitment of circulating leukocytes into the tissue critically depends on the regulated expression of endothelial cell adhesion molecules (CAMs). Various proinflammatory stimuli upregulate endothelial CAMs, including cytokines and UV irradiation. We have investigated the effects of ionizing radiation (IR) on endothelial CAM expression. Organ cultures of normal human skin as well as cultured human dermal microvascular endothelial cells (HDMEC) were exposed to IR. Expression of three major endothelial CAMs was studied in skin organ cultures by immuno

Ionizing radiation enhances IL-6 and IL-8 production by human endothelial cells.

Irradiation exposure is known to induce an inflammatory reaction. Endothelial cells play a crucial role both in the inflammatory process and in radiation damage. Therefore, supernatants and cell lysates of (60)Co-irradiated human umbilical vein endothelial cells (HUVEC) have been assessed for the presence of pro-inflammatory cytokines. After gamma irradiation, interleukin (IL)-1alpha, IL-1beta and tumor necrosis factor (TNF)-alpha remained undetectable in both cell supernatants and cell lysates. However, a dose-dependent increase in the production of IL-6 and IL-8 has been demonstrated up to 6

Enhanced expression of the Kin17 protein immediately after low doses of ionizing radiation.

Kin17 is a mammalian nuclear protein sharing a slight sequence homology with the bacterial RecA protein. Kin17 has a zinc-finger motif and binds efficiently to curved DNA, a genomic topology associated with illegitimate recombination junctions. We investigated the relationship between the level of Kin17 protein and genomic alteration due to either impaired wild-type p53 functions or exposure to gamma rays. We used BP cells, a rodent epithelial cell system. The cell lines used were syngeneic and harbored wild-type or mutant p53 alleles and exhibited different sensitivities to gamma irradiation.

Fibroblast growth factor-4 enhanced G2 arrest and cell survival following ionizing radiation.

Fibroblast growth factors (FGFs) bind to cell membrane receptors and activate signal transduction pathways related to cell growth, angiogenesis, and tumorigenesis. FGFs have been shown to be abundantly expressed in some of the human tumors, which are known to be poorly responsive to radiation therapy. Using adrenal cortical carcinoma cells genetically engineered to express FGF-4, we have tested cellular survival following exposure to ionizing radiation. We report here that FGF-4 enhances cellular capacity to survive ionizing radiation. Furthermore, cell cycle analysis shows a pronounced increa

Alterations in gastric mucus secretion in rhesus monkeys after exposure to ionizing radiation.

The aim of the present study was to evaluate the effect of gamma-irradiation on soluble gastric mucus. Six conscious chair-adapted rhesus monkeys were studied once before and twice after exposure to ionizing irradiation (800 rads). Using a marker (99mTc-DTPA) dilution technique, acidic glycoprotein (AG), neutral glycoprotein (NG), ion, and fluid output were determined during a basal period and after the administration of an 80-ml water load. Irradiation significantly increased the outputs of both AG and NG during the basal period. After the water load, NG output remained elevated but irradiati

Radiogenic male breast cancer with in vitro sensitivity to ionizing radiation and bleomycin.

A cytogenetically normal man with gynecomastia and a family history of diverse cancers developed adenocarcinoma of the breast 30 years following thymic irradiation. In vitro experiments measuring colony-forming ability of cultured skin fibroblasts from family members implied that the patient had a small but significant increase in sensitivity to ionizing radiation, and a moderate increase in sensitivity to bleomycin, a radiomimetic drug. Enhanced radiosensitivity of fibroblasts from the patient's mother, and bleomycin sensitivity of fibroblasts from the sister suggested, but did not prove, tha
