The challenge of implant integration in partial meniscal replacement: an experimental study on a silk fibroin scaffold in sheep.

PURPOSE: To restore meniscal function after excessive tissue damage, a silk fibroin implant for partial meniscal replacement was developed and investigated in an earlier sheep model. After 6 months implantation, it showed promising results in terms of chondroprotection and biocompatibility. To improve surgical fixation, the material was subjected to optimisation and a fibre mesh was integrated into the porous matrix. The aim of the study was the evaluation of this second generation of silk fibroin implants in a sheep model. METHODS: Nine adult merino sheep received subtotal meniscal replaceme…

Efficacy of silk fibroin-nano silver against Staphylococcus aureus biofilms in a rabbit model of sinusitis.

BACKGROUND: Staphylococcus aureus biofilms contribute significantly to the recalcitrant nature of chronic rhinosinusitis. In previous studies, it has been shown that silk fibroin-nano silver solution can eliminate S. aureus biofilms in vitro, which suggests a potential role of this novel agent in the treatment of biofilm-associated diseases, such as sinusitis. OBJECTIVE: The aim of this study was to investigate the efficacy of silk fibroin-nano silver solution as a topical anti-biofilm agent in a rabbit model of sinusitis. METHODS: Biofilm-associated sinusitis models were established in 24 N…

Full covered self-expandable metal stents for the treatment of anastomotic leak using a silk thread.

To evaluate the safety and effectiveness of fixation of the fully covered self-expandable metal stent (SEMS) placement using a silk thread for complete closure of an anastomotic leak. An anastomotic leak is a life-threatening complication after gastrectomy. Although the traditional treatment of choice was surgical re-intervention, an endoscopic SEMS can be used alternatively.During the study period, we retrospectively reviewed consecutive patients who received a modified covered SEMS capable of being fixed using a silk thread (Shim technique) due to an anastomotic leak after gastrectomy to pre…

Comparative acoustic performance and mechanical properties of silk membranes for the repair of chronic tympanic membrane perforations.

The acoustic and mechanical properties of silk membranes of different thicknesses were tested to determine their suitability as a repair material for tympanic membrane perforations. Membranes of different thickness (10-100μm) were tested to determine their frequency response and their resistance to pressure loads in a simulated ear canal model. Their mechanical rigidity to pressure loads was confirmed by tensile testing. These membranes were tested alongside animal cartilage, currently the strongest available myringoplasty graft as well as paper, which is commonly used for simpler procedures. …

The role of photo-electric properties of silk cocoon membrane in pupal metamorphosis: A natural solar cell.

Silkworm metamorphosis is governed by the intrinsic and extrinsic factors. One key intrinsic factor is the temporal electrical firing of the neuro-secretory cells of the dormant pupae residing inside the silk cocoon membrane (SCM). Extrinsic factors are environmental like temperature, humidity and light. The firing pattern of the cells is a function of the environmental factors that eventually controls the pupal development. How does the nervous organization of the dormant pupae sense the environment even while enclosed inside the cocoon shell? We propose that the SCM does this by capturing th…

Anterior cruciate ligament reconstruction in a rabbit model using silk-collagen scaffold and comparison with autograft.

The objective of the present study was to perform an in vivo assessment of a novel silk-collagen scaffold for anterior cruciate ligament (ACL) reconstruction. First, a silk-collagen scaffold was fabricated by combining sericin-extracted knitted silk fibroin mesh and type I collagen to mimic the components of the ligament. Scaffolds were electron-beam sterilized and rolled up to replace the ACL in 20 rabbits in the scaffold group, and autologous semitendinosus tendons were used to reconstruct the ACL in the autograft control group. At 4 and 16 weeks after surgery, grafts were retrieved and anal…

Influence of the protocol used for fibroin extraction on the mechanical properties and fiber sizes of electrospun silk mats.

Silk fibroin (SF) was regenerated using three of the most common protocols described in the bibliography for the dissolution of raw SF (LiBr 9.3M, CaCl2 50 wt.% or CaCl2:EtOH:H2O 1:2:8 in molar ratio). The integrity of regenerated SF in aqueous solution was analyzed by SDS-PAGE and different profiles of degradation were observed depending on the protocol used. This fact was found to affect also the aqueous solubility of the freeze dried protein. These different SFs were used to produce electrospun mats using SF solutions of SF 17 wt.% in 1,1,1,1',1',1'-hexafluoro-2-propanol (HFIP) and signific…

Effects of different post-spin stretching conditions on the mechanical properties of synthetic spider silk fibers.

Spider silk is a biomaterial with impressive mechanical properties, resulting in various potential applications. Recent research has focused on producing synthetic spider silk fibers with the same mechanical properties as the native fibers. For this study, three proteins based on the Argiope aurantia Major ampullate Spidroin 2 consensus repeat sequence were expressed, purified and spun into fibers. A number of post-spin draw conditions were tested to determine the effect of each condition on the mechanical properties of the fiber. In all cases, post-spin stretching improved the mechanical prop…

Utilising silk fibroin membranes as scaffolds for the growth of tympanic membrane keratinocytes, and application to myringoplasty surgery.

BACKGROUND: Chronic tympanic membrane perforations can cause significant morbidity. The term myringoplasty describes the operation used to close such perforations. A variety of graft materials are available for use in myringoplasty, but all have limitations and few studies report post-operative hearing outcomes. Recently, the biomedical applications of silk fibroin protein have been studied. This material's biocompatibility, biodegradability and ability to act as a scaffold to support cell growth prompted an investigation of its interaction with human tympanic membrane keratinocytes. METHODS …

Comparison of methods for the repair of acute tympanic membrane perforations: Silk patch vs. paper patch.

We investigated the effects of repairing large tympanic membrane (TM) perforations in rats with a thin silk patch compared with the commonly used paper patch. We performed bilateral myringotomies of 1.8 mm in diameter on 50 adult Sprague-Dawley rats with intact TMs. The perforations in the right ears of 40 rats were treated with a silk patch, and the perforations in the left ears of the same rats were treated with a paper patch. Ten rats acted as controls. The mean healing times of the TM perforations on the silk-patch-treated ears and the paper-patch-treated ears were 7.2+/-1.48 and 9.1+/-1.1…

A multifunctional composite hydrogel with natural polymer silk fibroin and gelatin for promoting chronic wound healing.

The skin, being the largest organ of the human body, plays a pivotal role as a protective barrier against infections. Due to persistent infection and inflammation, chronic wounds pose a significant challenge. In the study, GelMA and SilMA hydrogels were synthesized using natural biopolymers gelatin and silk fibroin as raw materials. Honokiol and cobalt ions are mixed in proportion to form a composite hydrogel. Honokiol has powerful anti-inflammatory, antioxidant and chronic wound healing capabilities, while cobalt ions can promote vascular regeneration by activating the hypoxia-inducible facto…

Assessing the Immunological Safety of Silk Fibroin Nanoparticles: An In Vitro Study With Primary Human Immune Cells.

Biomaterials for drug delivery must be biodegradable, nontoxic, and must not trigger inflammatory or immune responses. Silk fibroin (SF) is a natural polymer with exceptional mechanical strength and notable biological properties. In this study, the immunogenicity and inflammatory responses of silk fibroin nanoparticles (SFNPs) were characterized in primary human cells for the first time. SFNPs with a mean particle size of 213 ± 9 nm were synthesized and characterized. Primary human immune cells were exposed to SFNPs at concentrations ranging from 0.1 to 0.01 mg/mL. Cell viability remained abov…

The influence of substrate and ethanol treatment on properties and quantitative analysis of retention of recombinant spider silk coatings.

Silk coatings have significant application potential in medical materials due to their biocompatibility. Through processing of silk materials and functionalization, new coating properties can be gained. Following initial protein adsorption on a surface, an ethanol post-treatment can be used to enhance properties. Ethanol is thought to induce changes in protein structure, but very little is known on its effect on the binding stability and retention of coatings. Here, we study the effect of ethanol post-treatment and substrate material on recombinant silk coating adhesion, stability, structural …

The Impact of S-220-Loaded Silk Hydrogels on In Vitro Neurite Outgrowth in 2 and 3 Dimensions.

Silk hydrogels have attracted increasing interest as biocompatible substrates for neural cell culture and regenerative research. Recent research has shown that activation of exchange protein directly activated by cAMP 2 (Epac2) promotes neurite outgrowth in several neuronal models. Here, we investigated the effects of Bombyx mori silk hydrogel, alone or in combination with S-220, a non-hydrolysable cAMP analogue that selectively activates Epac2, on neurite outgrowth. Epac2 activation significantly and dose-dependently enhanced both the total and longest neurite length. Neurons cultured on silk…
