Spermatic cord torsion: loss of spermatogenesis despite return of blood flow.

Previous work in animal models has recapitulated and refined the clinical observation that longer periods of testicular torsion are associat … of blood flow, testis weights were significantly reduced by half by 7 days after torsion repair and spermatogenesis was clearly disrupted upon histological examination 30 days after torsion repair. Increasing the time of torsion to 2 or 4 h did not inhibit return of blood flow to the testis as measured 48 h after torsion repair. Unless the testis is infarcted, te …

Optimization of oxygen concentration and glycosphingolipid effects on spermatogenesis in mouse testicular culture†.

Our previous research highlighted the importance of hormones, free fatty acids, lysophospholipids, and antioxidants in supporting in vitro spermatogenesis in mice through the development of a chemically defined medium (CDM). While it was possible to induce round spermatids through the culture of testicular tissue in medium containing these factors, challenges remained with the low efficiency of spermatogenesis and the differentiation into elongating spermatids. This study aimed to further improve the in vitro spermatogenesis system by exploring optimal oxygen concentrations and identifying add…

Testicular Morphology and Spermatogenesis in Potamotrygon motoro: Insights Into Reproduction of Freshwater Stingrays.

This study investigated the spermatogenesis of the freshwater stingray Potamotrygon motoro using microscopic analyses. The testes of this species were described as being composed of germinal zones, a degenerated zone, and the epigonal organ, consisting of connective tissue and lymphomyeloid cells. A cystic pattern of spermatogenesis was observed, in which each cyst is formed and maintained by Sertoli cells that undergo morphological and positional changes throughout the process. After the release of spermatozoa into the duct, the peripheral cysts formed a degenerating layer. Spermiogenesis, th…

Smek1-Dependent Transcriptional Repression of Usp10 Is Essential for the Progression of Murine Spermatogenesis.

Meiotic progression is critically dependent on precise regulatory networks to ensure genomic stability. SMEK1 is recognized as a regulatory subunit of the protein phosphatase 4 complex. However, its potential phosphatase-independent functions in mammalian meiosis remain largely unexplored. Given the association of genetic variants near the SMEK1 locus with human infertility, we sought to define its specific role and mechanism in murine spermatogenesis. We generated a germ cell-specific Smek1 knockout mice model by crossing Smek1f/f mice with Stra8GFP-Cre mice. The phenotypic consequences were …

LncRNA THOR regulates spermatogenesis by interacting with and stabilizing Sycp3 protein in zebrafish.

THOR (testis-associated highly conserved oncogenic long non-coding RNA) is a highly conserved and testis-enriched lncRNA across vertebrates that plays diverse roles in various cancers. However, its physiological function and regulatory mechanism in testes remain largely unknown. Here, we investigated the genomic location and expression pattern of THOR in the model organism zebrafish, and generated a homozygous THOR knockout model using CRISPR-Cas9 technology. Loss of THOR in zebrafish impaired spermatogenesis, leading to oligospermia (38.7% reduction in sperm count), reduced sperm motility, sp…

Protein palmitoylation in spermatogenesis and sperm function: emerging roles and regulatory mechanisms†.

Spermatogenesis, a complex developmental process, is spatiotemporally regulated by post-translational modifications. Among these, dynamic protein palmitoylation emerges as a central coordinator of membrane-associated events, critically governing acrosome formation, tail assembly, blood-testis barrier integrity, and epididymal maturation. This review synthesizes current knowledge on how palmitoylation, catalyzed by zinc finger Asp-His-His-Cys enzymes and reversed by thioesterases, regulates protein localization, stability, and interactions to ensure normal sperm production and function. Disrupt…

B3GALT4-dependent gangliosides are required for normal late spermatogenesis and male fertility.

Gangliosides play important roles in membrane organization and cellular signaling. Although their involvement in spermatogenesis has been suggested, the specific functions of individual ganglioside species during each stage of germ cell development remain poorly understood. In this study, we generated beta-1,3-galactosyltransferase 4(B3galt4) knockout (KO) mice to investigate the role of B3GALT4-dependent ganglioside in male germ cell development. Immunostaining analyses demonstrated that B3galt4 deficiency caused selective alterations in ganglioside distribution. Although testis size did not …

Utilization of the QuPath open-source software platform for analysis of mammalian spermatogenesis†.

The adult mammalian testis is filled with seminiferous tubules, which contain somatic Sertoli cells along with germ cells undergoing all phases of spermatogenesis. During spermatogenesis in postnatal mice, male germ cells undergo at least 17 different nomenclature changes as they proceed through mitosis as spermatogonia (=8), meiosis as spermatocytes (=6), and spermiogenesis as spermatids (=3). Adding to this complexity, combinations of germ cells at each of these stages of development are clumped together along the length of the seminiferous tubules. Due to this, considerable expertise is req…

A Review and New Insights Into Spermatogenesis From Single-Cell RNA Sequencing.

As the most primitive germ cells in the testis, spermatogonial stem cells (SSCs) not only constantly renew themselves to ensure their quantity, but also differentiate into mature sperm cells to complete spermatogenesis and transmit genetic information to the next generation. Successful spermatogenesis is inseparable from niche regulation, which provides factors that enhance the self-renewal of SSCs to maintain their numbers and directs the appropriate differentiation of spermatogonia. Some progress has been achieved in the definition and isolation of SSCs. However, a high degree of cellular he…

Functional Study of Four Histone Genes Involved in the Spermatogenesis of Cynoglossus semilaevis.

Chinese tongue sole (Cynoglossus semilaevis) is an important mariculture fish in China, and female individuals present a growth advantage. However, genetic females (ZW) can sex reverse to phenotypic males, designated pseudomales. The pseudomale shows abnormal spermatogenesis and produces only Z sperm. Histone is pivotal in spermatogenesis, and post-translational modification could regulate its function. A comparison of testis phosphorylated and ubiquitinated proteins revealed 8 and 12 differentially phosphorylated and ubiquitinated histones in the testes of male and pseudomale Chinese tongue s…

Genetic deficiency of EXOSC10 ribonuclease disrupts spermatogenesis and male fertility in mice.

Male germ cell meiosis is essential for the generation of haploid spermatozoa in mammals. Oocyte-specific KO of Exosc10 results in female su … iotic recombination. These disruptions lead to heightened apoptosis, defective meiosis, compromised spermatogenesis, and resultant male sterility. Single-cell RNA-Seq analysis further corroborates these meiotic defects, demonstrating that Exosc10 deficiency disrupts RNA metabolism, misregulates meiotic gene expression, and impairs rRNA processing. Consequently, t …

Mechanisms and reversibility of nicotine-induced spermatogenesis impairment and DNA methylation changes.

Smoking among men of childbearing age poses a significant threat to their reproductive health. Nicotine, the primary bioactive compound in tobacco, adversely affects sperm characteristics, but mechanisms underlying its effects and if these effects are reversible upon cessation are unclear. We assessed the impact of nicotine exposure and its cessation on spermatogenesis and DNA methylation. Our findings revealed that nicotine exposure reduces sperm quality and leads to testicular damage. However, these effects can be reversed to some degree following nicotine cessation. In spermatogenesis, nico…

Functional Analysis of Forkhead Transcription Factor Fd59a in the Spermatogenesis of Drosophila melanogaster.

Spermatogenesis is critical for insect reproduction and is regulated by many different genes. In this study, we found that Forkhead transcription factor Fd59a functions as a key factor in the spermatogenesis of Drosophila melanogaster. Fd59a contains a conversed Forkhead domain, and it is clustered to the FoxD subfamily with other FoxD members from some insect and vertebrate species. Mutations in Fd59a caused swelling in the apical region of the testis. More importantly, fewer mature sperm were present in the seminal vesicle of Fd59a mutant flies compared to the control flies, and the fertilit…

A convenient research strategy for functional verification of epigenetic regulators during spermatogenesis.

Spermatogenesis is a fundamental process that requires a tightly controlled epigenetic event in spermatogonial stem cells (SSCs). The mechanisms underlying the transition from SSCs to sperm are largely unknown. Most studies utilize gene knockout mice to explain the mechanisms. However, the production of genetically engineered mice is costly and time-consuming. In this study, we presented a convenient research strategy using an RNA interference (RNAi) and testicular transplantation approach. Histone H3 lysine 9 (H3K9) methylation was dynamically regulated during spermatogenesis. As Jumonji doma…

Lactate dehydrogenase a-like 6B is not essential for spermatogenesis and male fertility in mice.

Lactate is recognized as a principal energy substrate for male germ cells. Lactate dehydrogenases (LDHs) catalyze the reversible conversion between pyruvate and lactate, which are required for male fertility. Among them, lactate dehydrogenase A-like 6B (Ldhal6b) has been identified as a testis-specific LDH gene. However, its precise roles in spermatogenesis remain to be elucidated. In this study, we used a Ldhal6b knockout mouse model to assess its impact on spermatogenesis. Our findings reveal that Ldhal6b knockout mice exhibit normal development and no significant alterations in male fertili…
