OBJECTIVE: The purpose of this investigation was to confirm the presence of subepithelial nerve plexuses associated with taste buds in the human tongue and to study the histomorphologic features of each plexus.
STUDY DESIGN: Through use of a light microscope, several hundred microslides representing biopsies of the human tongue were reviewed so that the histomorphologic character of subepithelial nerve plexuses could be detected and studied. Tissue sections were stained with hematoxylin-erythrosin, Bodian, and Masson's trichrome, as well as with immunohistochemical stains for neuron-specific …

We have studied the in vitro effect of sodium saccharin (NaSacch) on the rat adipocyte adenylyl cyclase complex. NaSacch (2.5-50 mM) inhibited significantly in a dose-dependent manner basal and isoproterenol-stimulated cAMP accumulation on isolated rat adipocytes. Similarly, NaSacch (2.5-50 mM) inhibited forskolin-stimulated adenylyl cyclase activity measured in the presence of Mg(2+)-ATP on adipocyte, astrocyte and thyrocyte membrane fractions. In contrast, NaSacch did not inhibit but slightly increased the forskolin-stimulated adenylyl cyclase activity measured in the presence of Mn(2+)-ATP

Except for taste bud cells, all sensory receptor cells and neurons have been shown to originate from neurogenic ectoderm (i.e., neural tube, neural crest, or ectodermal placodes). Descriptive studies on taste buds indicate that they, however, may arise from local epithelium. To determine whether taste receptor cells originate from neurogenic ectoderm or from local epithelium, the tongues of X chromosome-inactivation mosaic mice were examined. Results of this analysis show that taste bud cells and their surrounding epithelium always match in terms of the mosaic marker. This suggests that taste …

Taste sensitivity and responsivity, two-tastant and taste-mixture discrimination performance, and taste preferences were examined prior to and after the selective desalivation of 48 male Long-Evans rats. Altered preference behavior was observed in rats after removal of the major salivary glands, as well as after removal of only the submandibular-sublingual complexes. In 9 of 12 desalivated rats, decreased sensitivity and increased responsivity to near-threshold sodium chloride solutions were observed, although these changes were less than one-half an order of magnitude. No between-group differ…

Taste perception, taste preferences, and food preferences are three interrelated constructs underlying food-related behavior and contribute to individual dietary choices. Both genetic and environmental factors contribute to shaping these traits. Taste perception refers to interindividual differences in biological sensitivity to basic taste modalities. Taste preferences represent the hedonic evaluation of taste modalities, including salty, sweet, bitter, sour, or umami, and fatty tastes, and are closely related to, but distinct from, taste perception. Recent evidence in sensory genetics has ide…

Mechanisms ensuring reliable behavioral control amid complex, noisy sensory environments are often attributed to the central brain, yet computations occurring at the extreme sensory periphery remain poorly understood. Recent evidence hints at complex computations at the earliest stages of taste detection, but whether peripheral circuits of gustatory receptor neurons (GRNs) exist to perform them remains unknown. Here, using transsynaptic mapping and paired electrophysiological recordings in Drosophila, we reveal a feedforward circuit of monosynaptically connected GRNs. When driven by tastants, …

The perception of taste is a complex physiological process that extends far beyond the simple detection of flavor molecules, serving as a critical interface between nutrient sensing, metabolic regulation, and feeding behavior. Emerging evidence reveals that this process is profoundly modulated by endocrine and neuromodulatory systems, creating a sophisticated gut-brain-taste axis that integrates peripheral gustatory signals with central homeostatic and hedonic mechanisms. Hormones such as glucagon-like peptide-1, leptin, ghrelin, and CCK not only regulate appetite and energy balance but also d…

Oral perception during food and beverage consumption results from the integration of multiple sensory processes and is inherently complex. Current knowledge of oral perception is primarily derived from studies of stimuli and compounds that evoke specific sensory modalities, including taste, mouthfeel-related somatosensory sensations, and chemesthesis. Difficulties are associated with defining and characterizing each sensation because they involve multiple chemosensory systems. This review focuses on kokumi substances, taste modulators that have been increasingly examined in recent years, and o…

Chemotherapy-induced taste disorders and food aversion are important adverse effects that lead to reduced food intake and a deteriorated quality of life. Sex differences have been proposed to influence their development, but findings remain inconsistent, and no clinical consensus has been established. Further basic research is required to clarify these discrepancies underlying mechanisms. In this study, conditioned taste aversion (CTA) was evaluated in C57BL/6J mice after intraperitoneal administration of cisplatin, prepared in physiological saline, using a two-bottle choice test with distille…

Buds are continuously renewed sensory organs in which development, adult maintenance, and repair share overlapping molecular circuitry. During embryogenesis, WNT/β-catenin signaling promotes taste placode formation and placodal Shh expression, while SHH refines papilla spacing and restricts neighboring papilla formation. SOX2 functions as a taste-competence and progenitor maintenance factor. In adults, LGR5/LGR6-RSPO-WNT signaling sustains progenitor activity, and gustatory neurons are an important source of RSPO2; available genetic evidence is consistent with a neuron-derived contribution to …

BACKGROUND: The taste receptor type 1 (TAS1R) family includes class C G protein-coupled receptors that mediate sweet and umami taste perception through well-defined receptor complexes and downstream signaling pathways. Recently, TAS1Rs have been shown to be expressed not only in the oral cavity, but also in a wide variety of extraoral tissues, raising the possibility that they function as systemic nutrient sensors. However, the receptor composition and the molecular mechanisms by which extraoral TAS1Rs function remain unclear.
HIGHLIGHT: In the oral cavity, TAS1Rs function as heterodimeric co…

BACKGROUND: Endoderm-derived organs, including the gastrointestinal tract, play central roles in nutrient digestion and absorption. They simultaneously function as an "internalized external environment" that continuously interfaces with exogenous substances, thereby constituting a frontline of host defense. Chemosensory cells within the gastrointestinal epithelium, such as enteroendocrine cells and tuft cells, use molecular mechanisms analogous to those of taste reception to detect nutrients and xenobiotic compounds. However, because these populations are sparse, a comprehensive understanding …

Myo-inositol, a sugar alcohol produced by most plants, serves as a nutrient and feeding stimulant for many phytophagous insects. Inositol-sensitive taste sensilla have been characterized in many Lepidoptera larvae, but their molecular bases remain unclear. In this study, we determined the gustatory receptors (GRs) for myo-inositol in larva of Helicoverpa armigera, a worldwide crop pest. First, electrophysiological analyses revealed that medial sensilla styloconica strongly responded to myo-inositol and ribose, with weaker responses to xylose, and one GRN inside sensillum may mediate the respon…

Autism spectrum disorder (ASD) is a neurodevelopmental condition characterized by persistent challenges in social communication and restricted/repetitive behaviors, including sensory atypicalities. Taste processing is critical for assessing the sensory and hedonic properties of food; however, it remains understudied in ASD, which may limit our understanding of the disorder's links to selective eating and nutritional deficits. This scoping review on autistic gustatory functioning followed a predefined protocol. We conducted searches across four databases and identified 37 studies involving huma…

Saliva plays a crucial role in digestion, including taste perception, food breakdown, chewing, swallowing, and bolus formation. Saliva is mainly produced by three pairs of major glands: parotid, submandibular, and sublingual glands. To evaluate the effect of each salivary gland on taste preference, we conducted a 48-h two-bottle preference test using mouse models in which the parotid glands (PGs), submandibular glands (SMGs), or sublingual glands (SLGs) were surgically removed. The taste preferences for the five basic tastes of the PG- and SLG-removed mice were similar to those of the control …

Increasing pollution of heavy metals poses great risks to animals globally. Their survival likely relies on an ability to detect and avoid h … natonium. Detection of HMIs in food requires Gr66a + gustatory neurons but is independent of bitter-taste receptors. In these neurons, the ionotropic receptors IR76b, IR25a, and IR7a are required for the perception of heavy metals. Furthermore, IR47a mediates the activation of a distinct group of non-Gr66a + gustatory neurons elicited by HMIs. Together, …

Parosmia is a well-documented consequence of smell loss associated with Covid 19. Those who experience this qualitative olfactory disorder during recovery find that the odour of common foods and household items is distorted, and the experience can range from merely altered to profoundly disgusting. This can lead to a greatly altered relationship with food, including the physical symptoms of loss of appetite, nausea, vomiting, as well as wider reaching issues such as withdrawal from social situations, emotional distancing from others in their close social circles, and anhedonia. There is at pre
