Effects of inulin-type oligosaccharides (JSO) from Cichorium intybus L. on behavioral deficits induced by chronic restraint stress in mice and associated molecular alterations.

Depression and anxiety are serious psychiatric disorders with significant physical and mental health impacts, necessitating the development of safe and effective treatments. This study aimed to evaluate the efficacy of Jiangshi oligosaccharide (JSO), a type of inulin-based oligosaccharide, in alleviating anxiety and depression and to investigate the underlying molecular mechanisms. Using a mouse model of chronic restraint stress (CRS), JSO was administered orally at doses of 50, 100, and 200 mg/kg for 21 days. Behavioral tests, including the novelty-suppressed feeding test (NSFT), open field t

Sleep fails to depotentiate amygdala-reactivity to negative emotional stimuli in youth with elevated symptoms of anxiety.

Sleep-related problems often precede escalating anxiety in early adolescence. Pushing beyond broad sleep-mental health associations and toward mechanistic theories of their interplay can inform etiological models of psychopathology. Recent studies suggest that sleep depotentiates neural (e.g., amygdala) reactivity during reexposure to negative emotional stimuli in adults. Persistent amygdala reactivity to negative experiences and poor sleep characterize anxiety, particularly at the transition to adolescence. We propose that sleep depotentiates amygdala reactivity in youth but fails to do so am

Role of the orphan nuclear receptor NR4A1 in mediating alcohol withdrawal-associated anxiety-like behaviors.

BACKGROUND: Anxiety induced by alcohol withdrawal is a crucial target for treating alcohol dependence (AD). Anxiety has been linked to prefrontal cortex neuron apoptosis. Studies have also indicated that nuclear receptor subfamily 4 group A member 1 (NR4A1) is associated with both anxiety and apoptosis. However, it remains uncertain whether NR4A1 is involved in anxiety-like behaviors induced by alcohol withdrawal through the regulation of the apoptosis of prefrontal cortex (PFC) neurons. METHODS: We established an alcohol preference mouse model using the CPP paradigm, followed by 16 h of alco

Baicalein alleviates anxiety symptom in Parkinson's disease by targeting Sema3A-mediated parvalbumin interneuron dysfunction.

BACKGROUND: Anxiety is a common non-motor symptom of Parkinson's disease (PD), affecting 20 %-50 % of the patients, with an unmet need for effective therapies. Although perineuronal net (PNN) abnormalities have been implicated in both PD and anxiety, the underlying mechanisms and potential treatments remain poorly characterised. PURPOSE: In this study, we investigated whether baicalein ameliorates PD-associated anxiety via modulation of parvalbumin (PV) interneuron function by targeting semaphorin 3A (Sema3A). METHODS: Unilateral 6-hydroxydopamine (6-OHDA) lesioning of the substantia nigra p

Neuropeptide S receptor gene -- converging evidence for a role in panic disorder.

Animal studies have suggested neuropeptide S (NPS) and its receptor (NPSR) to be involved in the pathogenesis of anxiety-related behavior. In this study, a multilevel approach was applied to further elucidate the role of NPS in the etiology of human anxiety. The functional NPSR A/T (Asn¹⁰⁷Ile) variant (rs324981) was investigated for association with (1) panic disorder with and without agoraphobia in two large, independent case-control studies, (2) dimensional anxiety traits, (3) autonomic arousal level during a behavioral avoidance test and (4) brain activation correlates of anxiety-related em

Panic patients reveal idiographic associations between anxiety symptoms and catastrophes in a semantic priming task.

In the present study, a modified semantic priming paradigm was used to test whether panic patients more strongly associate catastrophes with anxiety symptoms than nonclinical subjects. Subjects named catastrophic target words (e.g. infarction) and target words neutral to anxiety themes (e.g. weekend) that followed auditive prime sentences immediately (i.e.0ms) or with a delay (i.e. 1500ms). Prime sentences described the perception of anxiety symptoms (e.g. You feel tense) or sensations neutral to anxiety (You feel relaxed). Consistent with an earlier study [Schniering C.A., & Rapee, R.M. (1997
