Alterations in Cisplatin Pharmacokinetics and Its Acute/Sub-chronic Kidney Injury over Multiple Cycles of Cisplatin Treatment in Rats.

Cisplatin (CDDP)-induced acute kidney injury (AKI) is a major clinical concern. CDDP treatment is generally conducted with multiple cycles; the magnitude of the CDDP-induced AKI may be altered by these cycles. Moreover, sub-chronic kidney injury (sCKI) induced by repeated CDDP treatment is often associated with renal interstitial fibrosis, potentially leading to chronic kidney disease. Therefore, it is suggested that the management of not only AKI but also sCKI induced by CDDP in multiple cycles plays an important role in the outcome of CDDP-based chemotherapy. This study investigated the alte

Chymase is activated in the pulmonary inflammation and fibrosis induced by paraquat in hamsters.

The involvement of chymase has been implicated in fibrotic response to tissue injuries. Besides its direct action, chymase indirectly promotes fibrotic response by generating angiotensin (Ang) II from Ang I. In the present study, we examined whether chymase and angiotensin converting enzyme (ACE), that also generates Ang II, were activated in the pulmonary inflammation and fibrosis induced by paraquat (PQ) in hamsters. In an acute PQ intoxication group, PQ was administered subcutaneously and the lungs were excised four days after administration. In a chronic PQ intoxication group, PQ was admin

Acute and subacute oral toxicity of copper oxide nanoparticles in female albino Wistar rats.

The extensive use of copper oxide nanoparticles (CuO-NPs) in various industries and their wide range of applications have led to their accumulation in different ecological niches of the environment. This excess exposure raises the concern about its potential toxic effects on various organisms including humans. However, the hazardous potential of CuO-NPs in the literature is elusive, and it is essential to study its toxicity in different biological models. Hence, we have conducted single acute dose (2000 mg/kg) and multiple dose subacute (30, 300 and 1000 mg/kg daily for 28 days) oral toxicity

Taring the scales: Weight-of-evidence framework for biocompatibility evaluations.

ISO 10993-1:2018 describes evaluating the biocompatibility profile of a medical device from a risk-based approach. This standard details the battery of information that should be considered within the assessment of a device, including raw material composition data, manufacturing processes, and endpoint testing. The ISO 10993/18562 series requires worst-case assumptions and exposure scenarios to be used in the evaluation, which may result in an over-estimation of patient safety risk. Currently, biocompatibility assessments evaluate each data set independently, and the consequence of this indivi

Epigenetic toxicity of environmental chemicals upon exposure during development - Bisphenol A and valproic acid may have epigenetic effects.

As of 2017, chemical substances registered in Chemical Abstracts Service (CAS) exceed 100 million, which is increasing yearly. The safety of chemical substances is adequately managed by regulations based on scientific information from toxicity tests. However, there are substances reported to have "biological effects" even though they are judged to be nontoxic in conventional toxicity tests. Therefore, it is necessary to consider a new concept on toxicity, "epigenetic toxicity". In this review, we explain about epigenetic toxicity using bisphenol A (BPA) and valproic acid (VPA) as examples. We …

The activity of methacrylate esters in skin sensitisation test methods II. A review of complementary and additional analyses.

Allergic contact dermatitis is an important occupational health issue, and there is a need to identify accurately those chemicals that have the potential to induce skin sensitisation. Hazard identification was performed initially using animal (guinea pig and mouse) models. More recently, as a result of the drive towards non-animal methods, alternative in vitro and in silico approaches have been developed. Some of these new in vitro methods have been formally validated and have been assigned OECD Test Guideline status. The performance of some of these recently developed in vitro methods, and of

Examination of microcystin neurotoxicity using central and peripheral human neurons.

Microcystins (MC) are a group of cyanobacterial toxins that comprises MC-LF and other cyclic heptapeptides, best known as potent hepatotoxicants. Cell culture and epidemiological studies suggest that MC might also affect the nervous system when there is systemic exposure, e.g., via drinking water or food. We asked whether in vitro studies with human neurons could provide estimates on the neurotoxicity hazard of MC-LF. First, we used LUHMES neurons, a well-established test system for neurotoxicants and neuropathological processes. These central nervous system cells express OATP1A2, a presumed c

Chemical sensitization and allergotoxicology.

Chemical sensitization remains an important environmental and occupational health issue. A wide range of substances have been shown to possess the ability to induce skin sensitization or respiratory sensitization. As a consequence, there is a need to have appropriate methods to identify sensitizing agents. Although a considerable investment has been made in exploring opportunities to develop methods for hazard identification and characterization, there are, as yet, no validated nonanimal methods available. A state of the art of the different in vitro approaches to identify contact and respirat

A Miniaturized Two-Electrode Detection System for Convenient and Rapid Ferricyanide-Mediated Chemical Toxicity Screening.

Toxicity bioassays play a crucial role in chemical risk assessment; however, conventional methods usually require specialized equipment, and limited accessibility is a significant drawback. Here, we present a user-friendly version using a simple two-electrode ferricyanide-mediated scaling of microbial respiration for rapid, convenient, and reliable toxicity assessment. The electrochemical cell is designed for continuous pulse-amperometric analysis, eliminating the need for disassembly/assembly and enabling a straightforward protocol to obtain a comprehensive evaluation of bacterial viability a

Deep Learning-Enabled Unbiased Precision Toxicity Assessment of Zebrafish Organ Development.

Precise assessment of toxicological effects remains a key bottleneck in biomedical and environmental health assessments. Traditional toxicology relies on macroscopic end points and manual image analysis, which limit sensitivity to structural damage and introduce subjective bias. We developed an automated deep learning approach based on U-Net for the precise assessment of toxic effects and established a general framework for objective toxicological analysis. Our U-Net model can perform pixel-level segmentation and morphological quantification on thousands of biological images in 1 min without b
