A Translational Readiness Checklist for Stem-Cell-Based Trials in Movement Disorders: A Practical Roadmap.

Stem cell-based therapies for movement disorders show inconsistent translation due to variations in product characterisation, delivery, endpoints, safety, and follow-up. Regulatory guidance provides principles but lacks a unified trial framework. The objective was to create a practical framework that translates scientific, regulatory, manufacturing, ethical, and clinical expectations into an auditable pre-First Patient In (FPI) checklist for stem cell trials. A structured review of the literature, clinical trials, and guidance documents was conducted. Searches were performed in PubMed/MEDLINE,

Shesha, a WhatsApp Chatbot for Linking Household Contacts to Tuberculosis Treatment or Preventive Therapy in South Africa: Design and Development.

BACKGROUND: Literature on the development of mobile health (mHealth) tools for public health interventions is scarce. This scarcity creates a knowledge gap, and new tools may repeat the mistakes of past implementations. OBJECTIVE: In this paper, we describe the development of Shesha, a WhatsApp-based chatbot designed to facilitate linkage to care for household contacts of people being treated for tuberculosis (TB). Shesha facilitates linkage by providing TB test results, TB preventive treatment (TPT) information, nudges, reminders, and personalized support. We developed Shesha to address the

Pullout strength of anterior spinal instrumentation: a product comparison of seven screws in calf vertebral bodies.

A lot of new implant devices for spine surgery are coming onto the market, in which vertebral screws play a fundamental role. The new screws developed for surgery of spine deformities have to be compared to established systems. A biomechanical in vitro study was designed to assess the bone-screw interface fixation strength of seven different screws used for correction of scoliosis in spine surgery. The objectives of the current study were twofold: (1) to evaluate the initial strength at the bone-screw interface of newly developed vertebral screws (Universal Spine System II) compared to establi

Evaluating the effects of inhibitory control training as a universal prevention strategy for internalizing complaints in middle childhood.

Internalizing complaints or inwardly directed psychological difficulties such as anxiety, worry and low mood can significantly affect children's emotional well-being and daily functioning. If left untreated, these difficulties can persist into adulthood. Growing evidence has reported the critical role of cognitive control (ability to regulate attention and memory processes) in the development and maintenance of emotional problems such as anxiety. Cognitive control training has shown promise in reducing emotional vulnerability in adults and adolescents. However, studies with children are scant.

Highly Conserved Influenza A Nucleoprotein as a Target for Broad-Spectrum Intervention: Characterization of a Monoclonal Antibody with Pan-Influenza Reactivity.

Influenza A viruses remain a persistent global health challenge due to their rapid antigenic evolution, zoonotic potential, and pandemic threat. Universal countermeasures targeting conserved viral components are urgently needed to enhance diagnostic, surveillance, and therapeutic capabilities. Here, we report the generation and characterization of a high-affinity monoclonal antibody (2D8 mAb) against the nucleoprotein (NP) of the H9N2 avian influenza virus, a subtype with increasing relevance to human infections. Importantly, 2D8 mAb exhibited robust cross-reactivity with a broad spectrum of i

Beyond physical access: disability-inclusive primary health care as a test of health equity in Sub-Saharan Africa.

Disability inclusion in primary health care is not a peripheral issue but a fundamental test of whether health systems in sub-Saharan Africa are advancing health equity and universal health coverage. Persons with disabilities experience persistent and overlapping barriers across the care pathway, including physical inaccessibility, unaffordable services, inaccessible information, poor provider communication, stigma, weak referral systems, and limited access to rehabilitation and assistive technology. These barriers are intensified by poverty, gender inequality, rural residence, and under-resou

Structural vaccinology: structure-based design of influenza A virus hemagglutinin subtype-specific subunit vaccines.

There is a great need for new vaccine development against influenza A viruses due to the drawbacks of traditional vaccines that are mainly prepared using embryonated eggs. The main component of the current split influenza A virus vaccine is viral hemagglutinin (HA) which induces a strong antibody-mediated immune response. To develop a modern vaccine against influenza A viruses, the current research has been focused on the universal vaccines targeting viral M2, NP and HA proteins. Crystallographic studies have shown that HA forms a trimer embedded on the viral envelope surface, and each monomer

A Mammalian Surface Display Platform to Optimize the Antigenicity of Viral Proteins.

Vaccine development often involves modifying native viral proteins to enhance their stability and antigenicity, as seen in approved Covid-19 vaccines and multiple HIV vaccine candidates currently under investigation. High throughput screening on the surface of mammalian cells enables the rapid evaluation of oligomeric, glycosylated viral proteins and the identification of mutations that improve their properties for vaccine design. Here, we developed an experimental platform that uses the PiggyBac transposon system to display libraries of viral protein variants on the surface of mammalian cells
