Efficacy of a nonadjuvanted recombinant FeLV vaccine and two inactivated FeLV vaccines when subject to consistent virulent FeLV challenge conditions.

The purpose of this study was to compare the efficacy of three FeLV vaccines, under identical conditions in a laboratory challenge model that closely mimics natural infection. Four groups of cats (n = 20 per group) were administered two doses of vaccine, 21 days apart, starting at 9-10 weeks of age (Purevax® FeLV, Versifel® FeLV, Nobivac® feline 2-FeLV, and a placebo). Cats were challenged 3 weeks later with a virulent, heterologous FeLV isolate. FeLV antigenemia was determined at weekly intervals from 3 to 15 weeks postchallenge. Circulating proviral DNA was determined on terminal PBMC sample

Alternative inactivated poliovirus vaccines adjuvanted with Quillaja brasiliensis or Quil-a saponins are equally effective in inducing specific immune responses.

Inactivated polio vaccines (IPV) have an important role at the final stages of poliomyelitis eradication programs, reducing the risks associated with the use of attenuated polio vaccine (OPV). An affordable option to enhance vaccine immunogenicity and reduce costs of IPV may be the use of an effective and renewable adjuvant. In the present study, the adjuvant activity of aqueous extract (AE) and saponin fraction QB-90 from Quillaja brasiliensis using poliovirus antigen as model were analyzed and compared to a preparation adjuvanted with Quil-A, a well-known saponin-based commercial adjuvant. Comparative immunogenicity and cross-clade protective efficacy of mammalian cell-grown inactivated and live attenuated H5N1 reassortant vaccines in ferrets.

Continued H5N1 virus infection in humans highlights the need for vaccine strategies that provide cross-clade protection against this rapidly evolving virus. We report a comparative evaluation in ferrets of the immunogenicity and cross-protective efficacy of isogenic mammalian cell-grown, live attenuated influenza vaccine (LAIV) and adjuvanted, whole-virus, inactivated influenza vaccine (IIV), produced from a clade 1 H5N1 6:2 reassortant vaccine candidate (caVN1203-Len17rg) based on the cold-adapted A/Leningrad/134/17/57 (H2N2) master donor virus. Two doses of LAIV or IIV provided complete prot

Analysis of reversions in the 5'-untranslated region of attenuated poliovirus after sequential administration of inactivated and oral poliovirus vaccines.

Replication of Sabin strains used in oral poliovirus vaccine (OPV) in the intestines of vaccine recipients leads to reversions that increase virus neurovirulence. Previously, a small study reported that prior immunization with inactivated poliovirus vaccine (IPV) resulted in faster accumulation of revertant virus, thus potentially increasing the risk of vaccine-associated paralytic poliomyelitis. We studied the impact that prior immunization with IPV and OPV has on shedding of revertant virus by healthy infants. By polymerase chain reaction (PCR), we amplified full-length poliovirus genomes di

[Comparative study of inactivated cultured vaccines against tick-borne encephalitis manufactured in Russia and in Austria by the "Immuno" firm].

Inactivated culture vaccine against tick-borne encephalitis (TBE) FSME-Immun-Inject produced in Austria ("Immuno") was compared to vaccine produced in the Institute of Poliomyelitis and viral encephalitis by immunological activity, safety, reactogenicity. In mice, no significant differences in protective activity of the vaccines were found when test-strains were diverse strains of TBE virus. Seroconversion in the sera from the vaccinated two times was not significantly different by the level of antibodies determined for both eastern and western serotypes of TBE virus.

Antibody responses to mumps virus proteins in natural mumps infection and after vaccination with live and inactivated mumps virus vaccines.

Paired sera from 20 patients with acute mumps infection, 16 from persons vaccinated with live attenuated mumps virus vaccine, and 12 from persons vaccinated with formalin-inactivated virus vaccine were studied for mumps antibodies by single radial hemolysis (SRH), hemagglutination inhibition (HI), and by enzyme immunoassays (EIA) specific for whole virus, envelope glycoprotein, and nucleocapsid antibodies. Mumps patients had diagnostic rises in serum mumps antibodies in 90-100% of the cases depending on the method of assay. Vaccination resulted in seroconversion in 75-88% (live vaccine) and in

Immunogenicity of an inactivated vaccine against senecavirus a and duration of the immune response in pigs.

Senecavirus A (SVA) infection causes vesicular disease in pigs, resulting in significant economic losses for swine producers and trade restrictions on pork exports. Due to its clinical similarity to other vesicular diseases, implementing control measures, such as vaccination, is essential to mitigate the impact of the disease. In this study, we assessed the immunogenicity of an inactivated SVA vaccine derived from a representative strain circulating in Brazil and the duration of vaccine-induced immunity in swine. Vaccinated 6- to 8-week-old pigs exhibited a robust humoral response, evidenced b

sIFITM1, sIFITM3, and sViperin antiviral proteins as inactivated CSFV vaccine adjuvants.

Vaccine adjuvants are now widely utilized in vaccine formulations. The IFN-Stimulated Genes (ISGs) family, play crucial roles in immune regulation and exhibit broad-spectrum antiviral activity. However, limited studies have investigated the potential of ISGs as vaccine adjuvants. Here, three swine ISGs fusion proteins were induced and purified from Escherichia coli, including IFITM1, IFITM3 and Viperin (sIFITM1, sIFITM3, and sViperin). Furthermore, sIFITM1, sIFITM3, and sViperin inhibited the replication of pseudorabies virus (PRV) in swine (PK-15 and 3D4/21) and murine (NIH/3 T3 and C57/B6-L)

Development and evaluation of an experimental inactivated vaccine against lumpy skin disease.

BACKGROUND AND AIM: Lumpy skin disease (LSD), caused by the LSD virus (LSDV), results in severe economic losses, reduced productivity, and restricted livestock trade. Although live attenuated vaccines are available, they pose risks such as viral shedding, recombination, and reversion to virulence. Inactivated vaccines, being safer alternatives, are particularly suitable for disease-free regions. This study aimed to develop an inactivated oil-adjuvanted vaccine using a local LSDV isolate and evaluate its immunogenicity and protective efficacy in rabbits. MATERIALS AND METHODS: Scab samples wer

The SPR analysis of the interaction of inactivated poliovirus vaccine attenuated strains with antibodies.

The interaction of inactivated poliovirus vaccine strains with oriented antibodies immobilized to protein A via Fc fragments has been investigated. Using an SPR biosensor, the kinetic and equilibrium parameters of the interaction of vaccine attenuated polioviruses of the Sabin strains type 1 and type 2, inactivated by various methods were determined. The strongest interaction was observed between polyclonal antibodies to Sabin strain type 2 poliovirus and Sabin strain type 2 poliovirus inactivated with β-propiolactone, KD = 1.04⋅10-11 M, as well as the interaction of monoclonal antibodies to S

Comparison of structure and immunogenicity of CVB1-VLP and inactivated CVB1 vaccine candidates.

Coxsackievirus B1 (CVB1) is a common cause of acute and chronic myocarditis, dilated cardiomyopathy and aseptic meningitis. However, no CVB-vaccines are available for human use. In this study, we investigated the immunogenicity of virus-like particle (VLP) and inactivated whole-virus vaccines for CVB1 when administrated to mice via either subcutaneous or intranasal routes formulated with and without commercial and experimental adjuvants. Here, the potential of utilizing epigallocatechin-3-gallate (EGCG) as a mucosal adjuvant synergistically with its ability to inactivate the virus were investi

Frequency of Ipsilateral Axillary Lymphadenopathy After the Inactivated COVID-19 Vaccine.

OBJECTIVE: During COVID-19 vaccine development studies, vaccines' efficacy and safety profiles should be carefully investigated. Only a few studies have shown that the COVID-19 vaccine can cause axillary lymphadenopathy on the injection arm. This study aimed to investigate the incidence of axillary lymphadenopathy and imaging findings using B-mode and Doppler ultrasonography (US) examinations in volunteers who had recently been vaccinated against COVID-19. METHODS: The ipsilateral and contralateral axillae of 101 volunteers who received the COVID-19 vaccine were evaluated using B-mode and Dop

An inactivated multivalent influenza A virus vaccine is broadly protective in mice and ferrets.

Influenza A viruses (IAVs) present major public health threats from annual seasonal epidemics and pandemics and from viruses adapted to a variety of animals including poultry, pigs, and horses. Vaccines that broadly protect against all such IAVs, so-called "universal" influenza vaccines, do not currently exist but are urgently needed. Here, we demonstrated that an inactivated, multivalent whole-virus vaccine, delivered intramuscularly or intranasally, was broadly protective against challenges with multiple IAV hemagglutinin and neuraminidase subtypes in both mice and ferrets. The vaccine is co

Evaluation of determinants of the serological response to the quadrivalent split-inactivated influenza vaccine.

The seasonal influenza vaccine is only effective in half of the vaccinated population. To identify determinants of vaccine efficacy, we used data from > 1,300 vaccination events to predict the response to vaccination measured as seroconversion as well as hemagglutination inhibition (HAI) titer levels one year after. We evaluated the predictive capabilities of age, body mass index (BMI), sex, race, comorbidities, vaccination history, and baseline HAI titers, as well as vaccination month and vaccine dose in multiple linear regression models. The models predicted the categorical response for > 75

Subacute Thyroiditis After Receiving Inactivated Virus Vaccine for COVID-19.

The spread of SARS-COVID 19 infection has resulted in accelerated efforts at development and dissemination of vaccines throughout the globe. These vaccines have different mechanisms of actions and their efficacy and side effects are being monitored. There have been rare reports in literature of thyroid dysfunction after COVID-19 vaccine administration. Sub-acute thyroiditis is one such complication which can arise as a rare side-effect of vaccination. This has also been reported as a symptom of COVID-19 infection. Clinical features include fever, neck pain, palpitations and weight loss. We rep
