CONTEXT AND OBJECTIVE: Viperid venom-induced chronic local-toxicity continues even after anti-snake venom treatment. Therefore, traditional antidote Albizia lebbeck L. (Fabaceae) seed extract was tested against Echis carinatus S. (Viperidae) venom (ECV)-induced local toxicity to evaluate its complementary remedy.
MATERIALS AND METHODS: Soxhlet extraction of A. lebbeck seeds was performed with the increasing polarity of solvents (n-hexane to water); the extract was screened for phytochemicals (alkaloids, anthraquinones, flavonoids, glycosides, phenolics, saponins, steroids and tannins). In pre…

In this report, we present a study on the antigenic cross-reactivity of various venoms from the most dangerous Egyptian snakes and scorpions belonging to families Elapidae, Viperidae and Buthidae. The study was carried out with special reference to bonding properties between venoms and antivenoms and their involvement in the formation of specific and/or cross-reactive interactions. The homologous polyclonal antivenoms showed high reactivity to the respective venoms and cross-reacted with varying degrees to other non-homologous venoms. Assorting the antivenoms according to their susceptibility …

Crotalidae and Viperidae snake venoms contains several kinds of metalloproteinases which cause localized hemorrhage by direct action on blood vessel walls. We report here the entire amino acid sequence and the disulfide bridge locations of HT-2, one of the hemorrhagic toxins isolated from the venom of Crotalus ruber ruber (red rattlesnake). The non-reduced protein was first cleaved at methionine residues to provide a set of 8 fragments, which covered the entire sequence of HT-2. The disulfide bridge locations of HT-2 were also determined by using these primary fragments. The unambiguous sequen…

Thrombocytopenia is a common manifestation in viperid snakebite envenomings, contributing to systemic bleeding and hemodynamic alterations. Despite its relevance, this effect is not evaluated when assessing the neutralizing ability of antivenoms. In this study we present an in vivo method in mice for the quantification of thrombocytopenic effect. Representative venoms of Costa Rican viperid snake genera (Atropoides, Bothrops, Bothriechis, Cerrophidion, Crotalus, Lachesis, Metlapilcoatlus, and Porthidium) were administered by the intramuscular route in mice and platelet counts were carried out

BACKGROUND: Venom systems are ideal models to study genetic regulatory mechanisms that underpin evolutionary novelty. Snake venom glands are thought to share a common origin, but there are major distinctions between venom toxins from the medically significant snake families Elapidae and Viperidae, and toxin gene regulatory investigations in elapid snakes have been limited. Here, we used high-throughput RNA-sequencing to profile gene expression and microRNAs between active (milked) and resting (unmilked) venom glands in an elapid (Eastern Brown Snake, Pseudonaja textilis), in addition to compar…

Echis carinatus (EC) is known as saw-scaled viper and it is endemic to the Indian subcontinent. Envenoming by EC represents a major cause of snakebite mortality and morbidity in the Indian subcontinent. Zinc (Zn++) dependent snake venom metalloproteases (SVMPs) present in Echis carinatus venom (ECV) is well known to cause systemic hemorrhage and coagulopathy in experimental animals. An earlier report has shown that ECV activates neutrophils and releases neutrophil extracellular traps (NETs) that blocks blood vessels leading to severe tissue necrosis. However, the direct involvement of SVMPs in

The excessive production of inflammatory mediators results in an overactive immune response leading to the worsening of various human diseases. Thus, there is a still need to identify molecules able to regulate the inflammatory response. Lebecetin, a C-type lectin protein isolated from Macrovipera lebetina snake venom, was previously characterized as a platelet aggregation inhibitor and antitumor active biomolecule. In the present work, we investigated its effect on the production of some cytokines linked to inflammatory response and the underlying mechanisms in lipopolysaccharide (LPS)-induce

Gloydius tsushimaensis is an endemic species inhabiting only Tsushima, a remote Japanese island, and is a distinct species from Gloydius blomhoffii widely distributed throughout mainland Japan and Gloydius brevicaudus and Gloydius ussuriensis which are geographically distributed in South Korea. This is the first multicenter retrospective study of G. tsushimaensis bites in Japan. A study of seventy-two patients who visited the former Izuhara Hospital, the former Naka Tsushima Hospital, Tsushima Hospital, and Kamitsushima Hospital during the fourteen years from January 1, 2005, to December 31, 2

The expense of production and distribution of snakebite antivenom, as well as its relatively infrequent use, has caused antivenom to be increasingly difficult to obtain and ultimately producing an alarming global shortage. Unused, expired antivenom may represent a significant, untapped resource to ameliorate this crisis. This study examines the efficacy of expired antivenom over time using in vitro, whole blood clotting, and platelet function statistics. Representatives from three years for four different global brands of polyvalent antivenom were chosen and tested against their corresponding

Snake venoms are composed of a complex mixture of active proteins that induce toxic effects, such as edema, hemorrhage, and death. Lachesis muta has the highest lethality indices in Brazil. In most cases, antivenom fails to neutralize local effects, leading to disabilities in victims. Thus, alternative treatments are under investigation, and plant extracts are promising candidates. The objective of this work was to investigate the ability of crude extracts, fractions, or isolated products of Erythroxylum ovalifolium and Erythroxylum subsessile to neutralize some toxic effects of L. muta venom.

Every year, dogs are presented to veterinary clinics in the Netherlands after having been bitten by a viper. The viper is the only venomous snake native to the Netherlands. Clinical signs after an acquired viper bite can range from none (after a 'dry' bite) to very mild up to life threatening following a 'wet' bite. To prevent mortality it is important to monitor the animals for a period of time and provide adequate treatment. Clotting disorders and multiple organ failure can occur several days to a week after the viper bite, appropriate follow up is therefore important. In the Netherlands, a

This article examines an international controversy over the most visible scientific event of Brazilian physiology in the nineteenth century. In 1881, Brazilian scientist João Baptista Lacerda stated that he had found an efficient antidote to the poison of Brazilian snakes: permanganate of potash (nowadays, potassium permanganate). His findings were given great publicity in Brazil and traveled rapidly around the world. Scientists, especially in France, contradicted Lacerda's claims. They argued that permanganate of potash could not be a genuine antidote to snake bites since it could not neutral

The composition of snake venoms shows a high degree of variation at all taxonomic levels, and natural selection for diet has been implicated as a potential cause. Saw-scaled vipers (Echis) provide a good model for studying this phenomenon. The venoms of arthropod feeding species of Echis are significantly more toxic to natural scorpion prey than those of species which feed predominantly upon vertebrate prey. Although testing venom activity on natural prey is important for our understanding of the evolution of venom, natural prey species are often difficult to obtain in sufficient numbers for t

It has been a common belief that snake venom may help in the digestion of its prey, although direct examples and supporting evidence have not been sufficient. To address this, the present study examined whether preinjecting natural amounts of pit viper venom into experimental mice may accelerate their digestion by the snakes or gain energy benefit as compared to the control without the envenomation. Live adults of two Asian pit viper species Trimeresurus gracilis and T. stejnegeri stejnegeri, which inhabit the cold and warm environment respectively, were the subjects studied herein. A natural

The processes that drive the evolution of snake venom variability, particularly the role of diet, have been a topic of intense recent research interest. Here, we test whether extensive variation in venom composition in the medically important viper genus Echis is associated with shifts in diet. Examination of stomach and hindgut contents revealed extreme variation between the major clades of Echis in the proportion of arthropod prey consumed. The toxicity (median lethal dose, LD(50)) of representative Echis venoms to a natural scorpion prey species was found to be strongly associated with the

This study examined the effect of Echis pyramidum (EP) venom on time-course of lipid peroxidation in different vital organs of mice. Adult male Swiss albino mice were injected with EP venom (2 mg/kg, i.p.); control mice received vehicle alone (normal saline). Mice were killed at 1, 3, 6, 12, and 24 h post-envenomation. The liver, lung, kidney, heart, and brain (cerebrum and cerebellum) were collected for the estimation of malondialdehyde (MDA), an index of lipid peroxidation. The results of this study showed that a single injection of EP venom caused a significant lipid peroxidation in all the

The venom of the Armenian adder (Vipera raddei Boettger, 1890) was tested for its ability to induce histopathological changes in rabbits after long-term (once every 6 days for 30 days) intramuscular injection of the venom (0.35 mg/kg approx. 0.5 LD 50), by light microscopic examination of some organs (liver, heart, kidney, adrenal, lung, spleen). V. raddei (Vr) venom induces changes including necrosis and edematous appearance with cellular infiltration and vacuolation. The injury of kidneys includes significant changes of the glomerular apparatus. Lung sections showed infiltration of erythrocy

Snake-venom phospholipases A2 (PLA2s) represent a good model for studies of structure-function relationships, mainly because of their small size and diverse pharmacological and toxicological activities. To obtain new members of the abundant PLA2 family, the venom of the viper Vipera nikolskii was fractionated for the first time and two new proteins, VN5-3 and VN4-3, were isolated. Both proteins show phospholipase A2 activity and may possess neurotoxic activity. Based on the determined partial amino-acid sequences, the new proteins can be classified as basic Asp49 phospholipases A2. They were c
