[Bioterrorism].

Biological terrorism is intentionally to use infectious substances for developing diseases or death in animals or humans, leading to disaster and panic in our community. Bioterrorism-associated diseases are mostly rare or eradicated infectious diseases, and recently, we do not have experience to make a clinical and laboratory diagnosis. In particular, these infectious diseases have incubation periods from infection to development of the disease. The staff working at public health institutions, including legal medicine, must be involved in as a first responder when bioterrorism would happen. Th

Distinct influenza interfaces in cattle: repeated spillovers of highly pathogenic avian influenza A(H5N1) from wild birds and enzootic circulation of influenza D virus.

Cattle now occupy two epidemiologically distinct influenza interfaces. This Mini Review comparatively synthesizes epidemiological, pathological, experimental-transmission, genomic, and surveillance evidence for highly pathogenic avian influenza A(H5N1) and influenza D virus (IDV), asking how source ecology, tissue tropism, shedding, transmission, persistence, and zoonotic evidence differ between the two viruses. Genetically distinct H5N1 viruses have spilled over repeatedly from wild birds into United States dairy cattle. In lactating cows, infection is prominent in mammary alveolar epithelium

Mapping the geodispersal and evolutionary dynamics of regional Nipah virus strain in Bangladesh.

Nipah virus (NiV) is a zoonotic pathogen causing severe respiratory and neurological disease in humans, with high fatality rates, primarily transmitted from fruit bats to humans through bat-tainted raw date palm sap and fruits. NiV outbreaks have occurred almost annually across 34 of 64 districts in Bangladesh. The study investigated the geodispersal patterns and evolutionary phylodynamics of NiV, aiding in more effective outbreak management strategies in Bangladesh. Phylodynamic and geographic evolutionary analyses were conducted using the nucleoprotein (N) gene sequence (n = 23) isolated fro

Evaluating vectors for the design of a spillover-disrupting Lassa virus transmissible vaccine.

Lassa fever is a viral zoonotic disease that sickens tens of thousands of people each year when it spills over from its rodent reservoir throughout West Africa. Despite the burden this persistent spillover places on public health, stopping it has proven to be an intractable challenge despite decades of investment and effort. A revolutionary solution is the development of a transmissible vaccine that can spread through the rodent population resulting in immune coverage sufficient to eliminate Lassa virus transmission. Here, we evaluate the feasibility of a transmissible vaccine constructed from

Machine learning assessment of zoonotic potential in avian influenza viruses using PB2 segment.

BACKGROUND: Influenza A virus (IAV) is a major global health threat, causing seasonal epidemics and occasional pandemics. Particularly, Influenza A viruses from avian species pose significant zoonotic threats, with PB2 adaptation serving as a critical first step in cross-species transmission. A comprehensive risk assessment framework based on PB2 sequences is necessary, which should encompass detailed analyses of specific residues and mutations while maintaining sufficient generality for application to non-PB2 segments. RESULTS: In this study, we developed two complementary approaches: a regr

Amplification-free detection of zoonotic viruses using Cas13 and multiple CRISPR RNAs.

Zoonotic viruses such as hantaviruses and influenza A viruses present a threat to humans and livestock. There is thus a need for methods that are rapid, sensitive and relatively cheap to detect infections with these pathogens early. Here, we use an amplification-free clustered regularly interspaced short palindromic repeats-associated protein 13 (CRISPR-Cas13)-based assay, which is simple, cheap and field-deployable, to detect the presence or absence of genomic hantavirus or influenza A virus RNA. In addition, we evaluate whether the use of multiple CRISPR RNAs (crRNAs) can improve the sensiti

Retrospective Screening for Zoonotic Viruses in Encephalitis Cases in Austria, 2019-2023, Reveals Infection with Lymphocytic Choriomeningitis Virus but Not with Rustrela Virus or Tahyna Virus.

Zoonotic viruses may be neglected as etiologies of meningoencephalitis in humans. We performed retrospective testing of cerebrospinal fluid from encephalitis cases in biobank material for three zoonotic or potentially zoonotic viruses: rustrela virus (Rubivirus strelense, Matonaviridae); Tahyna virus (Orthobunyavirus tahynaense, Peribunyaviridae); and lymphocytic choriomeningitis virus ("LCMV", Mammarenavirus choriomeningitidis, Arenaviridae). The cohort consisted of 443 samples, received for routine diagnostic testing year-round between January 2019 and February 2023, and were negative for he

Long-term surveillance for hepatitis E virus in an Italian two-site farrow-to-finish swine farm.

In humans, hepatitis E virus (HEV) is responsible for an acute enterically transmitted hepatitis, which can become chronic in immune-compromised patients. Genotypes 3 and 4 (HEV-3 and HEV-4) are zoonotic, and domestic pigs and wild boar are the main reservoirs. The occurrence of autochthonous cases in Europe, which have been increasing over the last 10 years, has been associated with food-borne zoonotic transmission of HEV-3, mainly linked to consumption of undercooked or raw pork products (sausages containing liver) and wild boar meat. Zoonotic HEV-3 strains are widespread on pig farms, but l

Genomic Surveillance of Yellow Fever Virus Epizootic in São Paulo, Brazil, 2016 - 2018.

São Paulo, a densely inhabited state in southeast Brazil that contains the fourth most populated city in the world, recently experienced its largest yellow fever virus (YFV) outbreak in decades. YFV does not normally circulate extensively in São Paulo, so most people were unvaccinated when the outbreak began. Surveillance in non-human primates (NHPs) is important for determining the magnitude and geographic extent of an epizootic, thereby helping to evaluate the risk of YFV spillover to humans. Data from infected NHPs can give more accurate insights into YFV spread than when using data from hu

Estimation of Rift Valley fever virus spillover to humans during the Mayotte 2018-2019 epidemic.

Rift Valley fever (RVF) is an emerging, zoonotic, arboviral hemorrhagic fever threatening livestock and humans mainly in Africa. RVF is of global concern, having expanded its geographical range over the last decades. The impact of control measures on epidemic dynamics using empirical data has not been assessed. Here, we fitted a mathematical model to seroprevalence livestock and human RVF case data from the 2018-2019 epidemic in Mayotte to estimate viral transmission among livestock, and spillover from livestock to humans through both direct contact and vector-mediated routes. Model simulation

Development of a DNA-based microarray for the detection of zoonotic pathogens in rodent species.

The demand for diagnostic tools that allow simultaneous screening of samples for multiple pathogens is increasing because they overcome the limitations of other methods, which can only screen for a single or a few pathogens at a time. Microarrays offer the advantages of being capable to test a large number of samples simultaneously, screening for multiple pathogen types per sample and having comparable sensitivity to existing methods such as PCR. Array design is often considered the most important process in any microarray experiment and can be the deciding factor in the success of a study. Th

West Nile virus epizootiology, central Red River Valley, North Dakota and Minnesota, 2002-2005.

West Nile virus (WNV) epizootiology was monitored from 2002 through 2005 in the area surrounding Grand Forks, North Dakota. Mosquitoes were tested for infection, and birds were surveyed for antibodies. In 2003, WNV was epidemic; in 2004, cool temperatures precluded WNV amplification; and in 2005, immunity in passerines decreased, but did not preclude, WNV amplification.

Experimental and Natural Infections of Tick-Borne Encephalitis Virus in Dogs.

Dogs are frequently infected with the tick-borne encephalitis virus (TBEV). However, to date, only a few clinically manifest cases of tick-borne encephalitis (TBE) have been reported in dogs. In this study, three-month-old beagle dogs were infected with TBEV through a subcutaneous injection. Body temperature, clinical signs, blood haematology, blood biochemistry, and immune responses were monitored for up to 28 days postinfection (p.i.). No changes in body temperature or clinical signs were observed in the infected dogs. Most haematology and blood biochemistry parameters were unchanged after t

West Nile Virus Epidemic in Germany Triggered by Epizootic Emergence, 2019.

One year after the first autochthonous transmission of West Nile virus (WNV) to birds and horses in Germany, an epizootic emergence of WNV was again observed in 2019. The number of infected birds and horses was considerably higher compared to 2018 (12 birds, two horses), resulting in the observation of the first WNV epidemy in Germany: 76 cases in birds, 36 in horses and five confirmed mosquito-borne, autochthonous human cases. We demonstrated that Germany experienced several WNV introduction events and that strains of a distinct group (Eastern German WNV clade), which was introduced to German

Potential effects of hepatitis E virus infection in swine on public health in China.

Hepatitis E virus (HEV), a zoonotic pathogen, is the main cause of acute hepatitis worldwide. Swine serves as the main reservoir, and its infection is mainly transmitted via fecal-oral route. Due to huge consumption of pork in China, close human-swine interactions at pig farms likely contribute to high risk in zoonotic transmission of HEV. Thus, we aim to investigate the HEV prevalence in pig farm in seven provinces across the east to west China and estimate the potential effects of swine HEV on public health in China. In this study, serum samples of pig were collected for detection of anti-HE
