Inhibition of influenza virus-induced NF-kappaB and Raf/MEK/ERK activation can reduce both virus titers and cytokine expression simultaneously in vitro and in vivo.

Influenza virus (IV) infection can cause severe pneumonia and death. Therapeutic actions are limited to vaccines and a few anti-viral drugs. These target viral functions thereby selecting resistant variants. During replication IV activates the Raf/MEK/ERK-cascade and the transcription factor NF-kappaB. Both result in virus supportive and anti-viral effects by promoting viral genome transport for virus assembly and by inducing expression of pro-inflammatory host factors. Apart from tissue damage caused by the virus lytic replication, an imbalanced overproduction of anti-viral cytokines can caus

Synergistic activation of simian immunodeficiency virus and human immunodeficiency virus type 1 transcription by retinoic acid and phorbol ester through an NF-kappa B-independent mechanism.

The activation of human immunodeficiency virus type 1 (HIV-1) expression in latently infected cells by exogenous agents is believed to be important in the progression of AIDS. Most factors that are known to activate HIV-1 gene expression increase the binding of NF-kappa B or NF-kappa B-like transcription factors to the HIV-1 core enhancer region. In this report, we demonstrate that retinoic acid (RA) treatment of promonocytic U937 cells stimulates expression from the simian immunodeficiency virus (SIVmac) long terminal repeat (LTR). Furthermore, RA and phorbol 12-myristate 13-acetate (PMA) syn

Antiviral Activity of Dendritic Heparan Sulfate Mimetics against Respiratory Syncytial Virus and Herpes Simplex Virus‑1.

Heparan sulfate (HS) can act as an attachment receptor for many different animal viruses. Virus binding to HS occurs through electrostatic interactions between the negatively charged sulfate and carboxyl groups of HS and positively charged amino acids present in viral proteins. Soluble heparin, a glycosaminoglycan related to HS, can inhibit infection by many viruses, but the clinical use of heparin is limited by its anticoagulant activity. Synthetic HS mimetics that lack anticoagulant activity have been developed and can inhibit virus infection in vitro and in vivo. Building on this precedent,

Prior Dengue Virus Exposure is Associated with Enhanced Zika Virus-Specific ADCC Activity in a Longitudinal Human Cohort.

BACKGROUND: Prior dengue virus (DENV) infection is common in regions affected by Zika virus (ZIKV) and may shape immune responses to subsequent ZIKV infection. Although cross-reactive neutralizing and disease-enhancing antibodies have been extensively studied, the effect of prior DENV immunity on antibody-dependent cellular cytotoxicity (ADCC) responses to ZIKV remains unclear. METHODS: Longitudinal serum samples were obtained and analyzed from a prospective cohort of 46 PCR-confirmed ZIKV cases enrolled at two U.S. sites between 2016-2018 (follow-up visits 6-409 days). ZIKV cases were classi

Inactivation of Vaccinia Virus via Nitric Oxide-Plasma Activated Water: A Potential Way to Inactivate Mpox Virus.

Mpox virus (Poxviridae family) an emerging environmental bio-contaminant caused a non-zoonotic human infection in 2022. The recent surge mirrors early COVID-19 trends, highlighting the need for effective viral inactivation to prevent outbreaks and reduce environmental risks. Our study explores an eco-friendly and non-toxic antiviral approach using nitric oxide (NOx)-plasma activated water (PAW) for environmental decontamination. Vaccinia virus (VACV) chosen as a surrogate model due to their genetic similarities with Mpox virus (MPXV). Results demonstrated that NOx-PAW was non-toxic to host cel

The inflammasome pathway is activated by dengue virus non-structural protein 1 and is protective during dengue virus infection.

Dengue virus (DENV) is a medically important flavivirus causing an estimated 50-100 million dengue cases annually, some of whom progress to severe disease. DENV non-structural protein 1 (NS1) is secreted from infected cells and has been implicated as a major driver of dengue pathogenesis by inducing endothelial barrier dysfunction. However, less is known about how DENV NS1 interacts with immune cells and what role these interactions play. Here we report that DENV NS1 can trigger activation of inflammasomes, a family of cytosolic innate immune sensors that respond to infectious and noxious stim

Identification of Embryonic Chicken Proteases Activating Newcastle Disease Virus and Their Roles in the Pathogenicity of Virus Used as In Ovo Vaccine.

In ovo vaccination is an attractive immunization approach for chickens. However, most live Newcastle disease virus (NDV) vaccine strains used safely after hatching are unsafe as in ovo vaccines due to their high pathogenicity for chicken embryos. The mechanism for viral pathogenicity in chicken embryos is poorly understood. Our previous studies reported that NDV strain TS09-C was a safe in ovo vaccine, and the F protein cleavage site (FCS) containing three basic amino acids (3B-FCS) was the crucial determinant of the attenuation of TS09-C in chicken embryos. Here, five trypsin-like proteases t

Suppression of viral RNA polymerase activity is necessary for persistent infection during the transformation of measles virus into SSPE virus.

Subacute sclerosing panencephalitis (SSPE) is a fatal neurodegenerative disease caused by measles virus (MV), which typically develops 7 to 10 years after acute measles. During the incubation period, MV establishes a persistent infection in the brain and accumulates mutations that generate neuropathogenic SSPE virus. The neuropathogenicity is closely associated with enhanced propagation mediated by cell-to-cell fusion in the brain, which is principally regulated by hyperfusogenic mutations of the viral F protein. The molecular mechanisms underlying establishment and maintenance of persistent i

Cross-Reaction, Enhancement, and Neutralization Activity of Dengue Virus Antibodies against Zika Virus: A Study in the Mexican Population.

Zika virus (ZIKV), an emerging mosquito-borne flavivirus, has quickly spread in many regions around the world where dengue virus (DENV) is endemic. This represents a major health concern, given the high homology between these two viruses, which can result in cross-reactivity. The aim of this study was to determine the cross-reacting antibody response of the IgM and IgG classes against the recombinant envelope protein of ZIKV (rE-ZIKV) in sera from patients with acute-phase infection of different clinical forms of dengue, i.e., dengue fever (DF) and dengue hemorrhagic fever (DHF) (before the ar

Dengue virus infection-enhancing activity of undiluted sera obtained from patients with secondary dengue virus infection.

BACKGROUND: Dengue virus (DENV) infection-enhancing activity of pre-existing antibody has been suggested to be one of the factors responsible for the pathogenesis of the severe form of DENV infection, dengue haemorrhagic fever. Although studies of infection-enhancement activity using diluted serum sera are informative, infection-enhancement activity determined using undiluted sera may better reflect in vivo conditions. METHODS: We determined whether serum samples collected from dengue patients possessed the ability to enhance DENV infection without dilution using FcγR-expressing cells. RESUL

A case of severe chronic active infection with Epstein-Barr virus: immunologic deficiencies associated with a lytic virus strain.

Infectious mononucleosis (IM) is a self-limiting, lymphoproliferative disease induced by primary infection with the Epstein-Barr virus (EBV). Infection with EBV leads in general to lifelong asymptomatic persistence of the virus. We report the case of a woman who acquired IM at the age of 15 years and then suffered from recurrent high fever, fatigue, and signs of immunologic disorder for more than 12 years until she died of liver failure. In an attempt to describe and to define the course of chronic active infection with EBV, we performed immunologic and molecular assays that demonstrated lytic

Occurrence of Epstein-Barr virus deoxyribonuclease enzyme activity-neutralizing antibodies. An index of Epstein-Barr virus disease.

The occurrence of Epstein-Barr virus (EBV) specific DNase antibodies was measured in the sera of 400 patients. Three hundred sera were positive (greater than or equal to 20) for early antigen diffuse component (EA-D) antibodies, as measured by indirect immunofluorescence (IF). One hundred sera were EA-D antibody-negative but had IF IgG titers (greater than 40) to the viral capsid antigen (VCA). Twenty-nine of the anti-EA-D-positive sera, and 14 of the anti-EA-D-negative sera showed specific inhibition of EBV DNase. The mean age of positive male patients (39 years) for DNase antibodies was sign

Hepatitis B virus DNA in the sera of HBsAg carriers: a marker of active hepatitis B virus replication in the liver.

Sera and liver biopsies from 30 Italian patients, carriers of HBsAg for at least 3 years, were examined for markers of hepatitis B virus (HBV) infection by serological assays and immunofluorescence. Biopsies were analyzed for HBcAg, HBsAg, and delta antigen by immunofluorescence; sera were assayed for HBsAg/anti-HBs, HBcAg/anti-HBc, HBeAg/anti-HBe, delta/anti-delta, HBV-specific DNA polymerase activity and the presence of HBV DNA. HBcAg, HBeAg, and DNA polymerase tests were positive in the sera of 71, 86, and 57%, respectively, of carriers with intrahepatic HBcAg. HBV DNA was detected in 100%

In Vitro Evaluation of the Anti-Chikungunya Virus Activity of an Active Fraction Obtained from Euphorbia grandicornis Latex.

Chikungunya virus (CHIKV) is classified as a pathogen with the potential to cause a pandemic. This situation becomes more alarming since no approved drug exists to combat the virus. The present research aims to demonstrate the anti-CHIKV activity of molecules present in the latex of Euphorbia grandicornis. Therefore, a biodirected assay was carried out to find the molecules with anti-CHIKV activity. Extractions with hexane, dichloromethane, and methanol and subsequent purification by column chromatography were carried out to later evaluate cytotoxic activity by 3-(4,5-dimethyl-2-thiazolyl)-2,5

Chikungunya virus-induced persistent allodynia is accompanied by joint microenvironmental changes and involves cartilage degradation and TRPA1 activation.

BACKGROUND AND PURPOSE: Chikungunya virus (CHIKV) causes debilitating chronic joint pain. CHIKV-induced pain mechanisms are not completely understood and models designed to resemble CHIKV chronic pain are critical for the development of novel therapeutic approaches. EXPERIMENTAL APPROACH: Mechanical allodynia was induced by injection of either inactivated CHIKV or its viral RNA into the mouse ipsilateral knee. Both ipsilateral and contralateral joints were collected for histological analysis, collagen Type I content and immunohistochemistry. The effects of paracetamol and sodium diclofenac we

Distinct Activation of Defense-Related Gene Networks in Rosetted Versus Symptomless Shoots in Rose Rosette Virus-Infected Roses.

Rose rosette virus (RRV) causes witches' broom disease in roses. This study examined whether differences in disease in these spatially distinct domains are associated with local changes in defense-related hormones and transcriptional reprogramming. The levels of defense-related phytohormones salicylic acid (SA), jasmonic acid (JA), 9-lipoxygenase (9-LOX), 13-lipoxygenase (13-LOX), and derived oxylipins were different in these distinct tissues. We curated differentially expressed genes involved in the biosynthesis, storage, and signaling of SA, JA, and oxylipins to reveal how specific defense p

Secondary dengue virus infection is associated with endothelial activation and hypotension in an outpatient cohort from the Philippines.

BACKGROUND: Secondary dengue virus (DENV) infection is a known risk factor for severe clinical manifestations. Antibody-dependent enhancement of viral pathogenesis explains this phenomenon; however, the underlying mechanisms remain incompletely defined. OBJECTIVES: To compare the frequency of hypotension, endothelial activation, systemic inflammation, and thrombocytopenia in patients with primary and secondary DENV infection. METHODS: This was a cross-sectional study among children and young adults aged 1-26 years conducted at an outpatient clinic in the Philippines. Secondary infection was

Fuzheng Jiedu decoction alleviates H1N1 virus-induced acute lung injury in mice by suppressing the NLRP3 inflammasome activation.

ETHNOPHARMACOLOGICAL RELEVANCE: Severe influenza, marked by excessive cytokine production, is a major contributor to death in hospitalized individuals. Fuzheng Jiedu decoction (FZJDD), an effective traditional Chinese herbal recipe, has demonstrated promising results in combating the COVID-19 pandemic by reducing mortality and improving Symptoms, and has exhibited anti-inflammatory properties in both clinical trials and laboratory research. Given that pneumonia is a common outcome of SARS-CoV-2 and H1N1 virus infections, we hypothesized that FZJDD may also have therapeutic effects on influenza

Severe fever with thrombocytopenia syndrome virus induces platelet activation and apoptosis via a reactive oxygen species-dependent pathway.

Источник пометил публикацию как отозванную

Severe fever with thrombocytopenia syndrome (SFTS) is an emerging tick-borne infectious disease caused by the SFTS virus (SFTSV) and with a high fatality rate. Thrombocytopenia is a major clinical manifestation observed in SFTS patients, but the underlying mechanism remains largely unclear. Here, we explored the effects of SFTSV infection on platelet function in vivo in severely infected SFTSV IFNar-/- mice and on mouse and human platelet function in vitro. Results showed that SFTSV-induced platelet clearance acceleration may be the main reason for thrombocytopenia. SFTSV-potentiated platelet
