Inactivation of yeast inorganic pyrophosphatase by organic solvents.

A number of application for enzymes in organic solvents have been developed in chemical processing, food related conversions and analyses. The only unsolved problem related to nonaqueous enzymology is the notion that enzymes in organic solvent are mostly far less active than in water. Therefore, studies concerning the mechanisms by which enzymes are inactivated by organic solvents would reveal a clear understanding of the structure-function relationship of this phenomenon. Here we analyzed the effects of a series of alcohols (methanol, ethanol, 1-propanol and 2-propanol) and acetone on the act

Enhanced organically bound chromium yeast production.

This paper describes continuous and fed-batch fermentation protocols for enhanced production of organically bound chromium yeast. During continuous fermentation, several inorganic chromium compounds were evaluated. Sodium chromate demonstrated the best chromium incorporation into yeast biomass without any precipitation in the fermentation medium. During fed-batch fermentation, several sodium chromate concentrations were evaluated at 1.1, 3.5, 5.9, 7.1, 8.3, and 10.8 g/L with continuous or single-dose addition. For single-dose addition of sodium chromate, some precipitation was observed for all

Hereditary spastic paraplegia: mitochondrial metalloproteases of yeast.

Hereditary spastic paraplegia (HSP) is a genetically heterogenous group of inherited neurodegenerative disorders. Recently, an autosomal recessive form of HSP was mapped to 16q24.3, and subsequently the defective gene associated to HSP was identified and designated SPG7. The SPG7 gene product was predicted to encode a protein of 795 amino acids, and is called paraplegin. Paraplegin is highly homologous to a class of well studied yeast ATP-dependent zinc metalloproteases, which show 55%, 55% and 52% identity, respectively, to Afg3p, Rca1p and Yme1p. Mutation of either Afg3p, Rca1p or Yme1p in y

Ethanol tolerance of immobilized brewers' yeast cells.

A method based on the survival of yeast cells subjected to an ethanol or heat shock was utilized to compare the stress resistance of free and carrageenan-immobilized yeast cells. Results demonstrated a significant increase of yeast survival against ethanol for immobilized cells as compared to free cells, while no marked difference in heat resistance was observed. When entrapped cells were released by mechanical disruption of the gel beads and submitted to the same ethanol stress, they exhibited a lower survival rate than entrapped cells, but a similar or slightly higher survival rate than free

Multiple-copy integration in the yeast Yarrowia lipolytica.

Using an EcoRI-BglII fragment of the G unit of the rDNA of Y. lipolytica and a set of 11 deletions in the URA3 promoter, we have constructed several plasmids to test gene amplification in the rDNA. These plasmids contain the rDNA fragment for integration, defective versions of the URA3 gene, the XPR2 gene encoding alkaline extracellular protease (AEP) as a reporter gene, and part of the pBR322 plasmid for selection and replication in E. coli. Among these plasmids, one corresponds to a deletion which allows multiple integration into the rDNA (plasmid pINA773). Two other plasmids (pINA767 and pI

Yeast metallothionein function in metal ion detoxification.

A genetic approach was taken to test the function of yeast metallothionein in metal ion detoxification. A yeast strain was constructed in which the metallothionein locus was deleted (cup1 delta). The cup1 delta strain was complemented with normal or mutant metallothionein genes under normal or constitutive regulatory control on high copy episomal plasmids. Metal resistance of the cup1 delta strain with and without the metallothionein-expressing vectors was analyzed. The normally regulated metallothionein gene conferred resistance only to copper (1000-fold); constitutively expressed metallothio

Potential therapeutic effect of yeast killer toxin.

Experimental infections were produced in guinea pigs, rabbits and dogs with lesions similar to those seen in human seborrheic dermatitis and otitis externa by cutaneous application of cultures of Malassezia furfur and M. pachydermatis. Infected animals were treated by topical application of a concentrated yeast killer toxin (Hansenula anomala UCSC 25F). Clinical recovery as well as negative mycological test cultures of infected animals proved to the clearly associated with the treatment by the killer toxin.

Detection of yeast septicemia by biphasic and radiometric methods.

From January 1977 to April 1980 our microbiology laboratory used a commercial biphasic brain heart infusion vented culture method for fungal blood cultures and a commercial radiometric (BACTEC 460, Johnson Laboratories, Cockeysville, Md.) method for bacterial blood cultures. A total of 668 biphasic fungal blood cultures were processed, of which 30 grew yeasts from 19 patients. There were 38,324 BACTEC blood cultures processed for bacteria, of which 184 grew yeasts from 85 patients. The overall detection time for all yeasts averaged 8.3 days for the biphasic method and 2.4 days for the radiomet…

Respiration in the yeast and mycelial phases of Histoplasma capsulatum.

Respiration in the yeast and mycelial phases of Histoplasma capsulatum proceeds via a cytochrome system and an alternate oxidase, both present constitutively. The mycelial cytochrome system is distinguished by an additional partial shunt around the antimycin-sensitive site.

Stability of patulin to sulfur dioxide and to yeast fermentation.

The affinity of patulin for sulfur dioxide (SO2) is much less than was previously reported and is of little significance at the SO2 concentrations (below 200 ppm) used in the processing of apple juice and cider. However, at concentrations of 2000 ppm SO2 and 15 ppm patulin, combination was 90% complete in 2 days. Removal of SO2 liberated only part of the patulin, which suggests that 2 mechanisms are involved: one reversible (opening the hemiacetal ring) and one irreversible (SO2 addition at the double bond). Test with 2 yeasts used in English commercial cider making confirmed that patulin is e…

L-asparaginase by filamentous fungi and yeasts from lichens and soils across maritime Antarctic islands.

This study aimed to evaluate L-asparaginase (L-ASNase) production by fungi isolated from soils across different Antarctic islands. Out of 138 isolates, 33 filamentous fungi and 5 yeasts exhibited enzymatic activity in solid culture media. Filamentous fungi were identified by sequencing the Internal Transcribed Spacer (ITS) region, while yeasts were identified using the D1/D2 domains of the 26 S rDNA gene. The blue filamentous fungus Antarctomyces F13.CR.LASP exhibited a maximum L-ASNase productivity of 0.76 U/L. Regarding the yeasts, the isolate Vishniacozyma victoriae G.L11 reached a producti…

Differential Attraction of Summer and Winter Morphs of Spotted Wing Drosophila, Drosophila suzukii, to Yeasts.

Drosophila suzukii is a pest of soft and stone fruits that is attracted to yeast volatile metabolites. Drosophila suzukii has distinct summer and winter morphs which are found in different habitats. Complex communities of yeasts likely differ between habitats and thus we hypothesized morphs differ in their attraction to yeast volatiles from different species and combinations of species. We presented D. suzukii with yeast species in isolation and in combinations, as either post-culture mixes or co-cultured, and measured activity of the flies in laboratory choice tests with a Locomotor Activity …

Yeasts, arthropods, and environmental matrix: a triad to disentangle the multi-level definition of biodiversity.

Our understanding of the spread of yeasts in natural ecosystems remains somewhat limited. The recent momentum of yeast ecology research has unveiled novel habitats and vectors that, alongside human activities, impact yeast communities in their natural environments. Yeasts, as non-airborne microorganisms, rely on animal vectors, predominantly insects. However, the overlooked actor in this interplay is the environmental matrix, a player potentially influencing yeast populations and their vectors. This study aims to delve deeper into the intricate, multi-layered connections between yeast populati…
