Screening of microalgal strains with elevated α-tocopherol levels and antioxidant capacity for biotechnological applications.

One of the world's most important scientific challenges is addressing the global food crisis. Products offering potential health benefits, including various supplements with protective properties, have become a priority. Vitamin E is among the compounds gaining increasing attention from consumers. It is a group of compounds, with α-tocopherol of natural origin being the most bioavailable and valuable. This study investigated variations in α-tocopherol content, productivity, and antioxidant status at different growth stages in 15 microalgae strains from the Chlorophyta and Heterokontophyta. Res

A novel delins mutation in the alpha-TTP gene in a family segregating ataxia with isolated vitamin E deficiency.

Ataxia with isolated vitamin E deficiency is a rare autosomal recessive neurodegenerative disease due to mutations in the alpha-tocopherol transfer protein gene. In ataxia with isolated vitamin E deficiency, the biochemical hallmark is the low plasmatic levels of vitamin E and, in most of the patients, vitamin E supplementation allows a stabilization of the neurologic conditions. We have investigated the genetic cause of ataxia and reduced levels of vitamin E, and apolipoproteins A1 and B in a 16-y-old patient. Results revealed that our propositus is a compound heterozygote for the c.227_229de…

[Oxidative DNA damage in mothers of children with isolated orofacial clefts].

Recently, there is increasing evidence suggesting that oxidative stress may contribute to birth defects. 8-Hydroxy-2'-deoxyguanosine (8-OHdG) is considered to be a sensitive biomarker of oxidative DNA damage. It would seem important to establish whether levels of 8-OHdG and antioxidants are different in mothers of children with orofacial clefts comparing to women who gave birth to healthy children. The present study was carried out to investigate this point. Total plasma antioxidant status (TAS), serum 8-OHdG, plasma alpha-tocopherol and retinol were determined in 27 healthy mothers of childre…

Oxidative stress-independent depletion of epidermal vitamin A by UVA.

In hairless mice, epidermal vitamin A (retinol and retinyl esters) is strongly decreased following a single exposure to UVB. Here, using the same mouse model, we studied the effects of UVA on epidermal vitamin A content, lipid peroxidation, and CRBP-I expression, as well as the putative prevention of vitamin A depletion or lipid peroxidation by topical alpha-tocopherol. An acute exposure to UVA completely depleted epidermal vitamin A with EC50 of 0.25 and 0.5 J per cm2 for retinyl esters and retinol, respectively; these values were 0.1 J per cm2 for both retinoids under UVB exposure. CRBP-I ex…

Effects of phytanic acid on the vitamin E status, lipid composition and physical properties of retinal cell membranes: implications for adult Refsum disease.

Adult Refsum disease is an inherited disorder in which phytanic acid accumulates in tissues and serum. Two hypotheses have been proposed to explain the pathogenesis of this condition. The molecular distortion hypothesis suggests that phytanic acid may alter membrane composition and structure, thereby affecting membrane function(s). The anti-metabolite hypothesis suggests that an accumulation of phytanic acid in membranes may interfere with vitamin E function. These two hypotheses were investigated by studying the effects of modulating phytanic acid and alpha-tocopherol concentrations on the fa…

Mechanism causing vitamin E deficiency during chronic childhood cholestasis.

In order to characterize the mechanism(s) causing vitamin E deficiency during chronic childhood cholestasis, we studied 6 vitamin E-deficient cholestatic children with clinical evidence of neurologic dysfunction (group A), 4 vitamin E-sufficient cholestatic children and young adults with normal neurologic status (group B), and 6 vitamin E-sufficient noncholestatic children (group C). Intestinal absorption of dl-alpha-tocopherol (assessed by an oral tolerance test) was markedly impaired (p less than 0.001) in group A compared with groups B and C, which did not differ from each other. Intralumin…

Lipoproteins: Friend and foe for ferroptosis?

A recent study in Nature reports a mechanism for cancer cells to defend against ferroptosis. Calhoon et al.1 showed that glycosaminoglycans on the cell surface promote lipoprotein uptake in cancer cells and deliver anti-ferroptotic metabolites such as α-tocopherol, providing ferroptosis resistance in tumor growth.

A Simple ¹³C NMR Method for the Discrimination of Complex Mixtures of Stereoisomers: All Eight Stereoisomers of α-Tocopherol Resolved.

A simple one-dimensional (13)C NMR method is presented to discriminate between stereoisomers of organic compounds with more than one chiral center. By means of this method it is possible to discriminate between all eight stereoisomers of α-tocopherol. To achieve this the chiral solvating agent (S)-(+)-1-(9-anthryl)-2,2,2-trifluoroethanol and the compound of interest were dissolved in high concentrations in chloroform-d, and the nuclear magnetic resonance (NMR) spectrum was recorded at a low temperature. The individual stereoisomers of α-tocopherol were assigned by spikes of the reference compo

Antioxidant and UV-protective evaluation of tocochromanols in nanoformulations.

Tocochromanols, a group of compounds widely present in plant oils and commonly used in cosmetic formulations, exhibit varying photostability under UV irradiation. In this study, we investigated the photostability and potential UV-protective properties of several tocochromanol homologues (α-tocopherol, γ-tocopherol, γ-tocotrienol and plastochromanol-8) both in free form and encapsulated within liposomal and niosomal carriers. Our findings demonstrated that α-tocopherol, despite its widespread use in sunscreens, was the least stable under UV exposure. In contrast, γ-tocopherol, γ-tocotrienol and

Secondary carnitine deficiency and impaired docosahexaenoic (22:6n-3) acid synthesis: a common denominator in the pathophysiology of diseases of oxidative phosphorylation and beta-oxidation.

A critical analysis of the literature of mitochondrial disorders reveals that genetic diseases of oxidative phosphorylation are often associated with impaired beta-oxidation, and vice versa, and preferentially affect brain, retina, heart and skeletal muscle, tissues which depend on docosahexaenoic (22:6n-3)-containing phospholipids for functionality. Evidence suggests that an increased NADH/NAD(+) ratio generated by reduced flux through the respiratory chain inhibits beta-oxidation, producing secondary carnitine deficiency while increasing reactive oxygen species and depleting alpha-tocopherol…

Effects of graded dietary levels of Spirulina maxima on vitamins A and E in male rats.

The effects of ingesting the alga Spirulina maxima on the storage and utilization of vitamins A and E were investigated by feeding diets con … the feces. A low level of 2.7% S. maxima caused a significant reduction in plasma, liver and heart alpha-tocopherol levels. The concentrations of alpha-tocopherol in these tissues showed a marked decline with 10.7% S. maxima in the diet, followed by a lesser decline at higher levels. Liver retinoid levels of rats increased when S. maxima was added to the diet, su …

Formulation strategies to minimize oxidative damage in lyophilized lipid/DNA complexes during storage.

It has been shown that degradation of lipid/DNA complexes (lipoplexes) continues in the dried state during storage. The goal of this study was to evaluate the ability of various strategies to minimize the formation of reactive oxygen species (ROS) in lyophilized lipoplexes during storage, including metal removal from reagents, air displacement, and fortification with chelator/antioxidant agents. Formulations containing individual chelator (DTPA) and antioxidants (L-methionine or alpha-tocopherol), or in combination, were subjected to lyophilization. Accelerated storage conditions were investig…

Phospholipid transfer protein deficiency reduces sperm motility and impairs fertility of mouse males.

Vitamin E was discovered for its implication in reproductive biology, and its transport in mammalian plasma and brain was shown to be governed by plasma phospholipid transfer protein (PLTP). We show that PLTP deficiency is associated with hypofertility of mouse males but not mouse females, and it accounts for a significant decrease in total number of pups produced over a 2-month breeding period of PLTP knocked out mice (-32%, P<0.03). PLTP is highly expressed in epididymis of mouse males, and alpha-tocopherol, the main vitamin E isomer in vivo, was significantly less abundant in cauda and capu…

Genetic vitamin E deficiency does not affect MPTP susceptibility in the mouse brain.

Oxidative stress is involved in the degeneration of the nigrostriatal dopaminergic system in Parkinson's disease (PD). Vitamin E (alpha-tocopherol) is a potent antioxidant in the cell membrane that can trap free radicals and prohibit lipid peroxidation. The retention and secretion of vitamin E are regulated by alpha-tocopherol transfer protein (TTP) in the brain and liver. Dysfunction of TTP results in systemic deficiency of vitamin E in humans and mice, and increased oxidative stress in mouse brain. In this study, we investigated the effect of vitamin E deficiency in PD development by generat…
