Sitting time is negatively related to microvascular endothelium-dependent function in rheumatoid arthritis.

BACKGROUND: Sedentary behaviour is linked to increased cardiovascular disease risk in Rheumatoid Arthritis (RA), but the biological processes underlying this relationship are not understood. OBJECTIVES: To investigate the cross-sectional associations of habitual sedentary behaviour, with endothelial function in RA. METHODS: Sixty-eight RA patients (Mage = 55 ± 12 years) underwent Laser Doppler Imaging with iontophoresis, to assess microvascular endothelium-dependent (acetylcholine, ACh) and endothelium-independent (sodium nitroprusside, SNP) function. Large-vessel endothelium-dependent and e

Inhibition of Endoplasmic Reticulum Stress Preserves the Integrity of Blood-Spinal Cord Barrier in Diabetic Rats Subjected to Spinal Cord Injury.

The blood-spinal cord barrier (BSCB) plays significance roles in recovery following spinal cord injury (SCI), and diabetes mellitus (DM) impairs endothelial cell function and integrity of BSCS. Endoplasmic reticulum (ER) stress occurs in the early stages of SCI and affects prognosis and cell survival. However, the relationship between ER stress and the integrity of BSCB in diabetic rats after SCI remains unclear. Here we observed that diabetic rats showed increased extravasation of Evans Blue (EB) dye, and loss of endothelial cells and pericytes 1 day after SCI compared to non-diabetic rats. D

Inducible overexpression of endothelial proNGF as a mouse model to study microvascular dysfunction.

Impaired maturation of nerve growth factor precursor (proNGF) and its accumulation has been reported in several neurodegenerative diseases, myocardial infarction and diabetes. To elucidate the direct impact of proNGF accumulation identified the need to create a transgenic model that can express fully mutated cleavage-resistant proNGF. Using Cre-Lox technology, we developed an inducible endothelial-specific proNGF transgenic mouse (proNGFLoxp) that overexpresses GFP-conjugated cleavage-resistant proNGF123 when crossed with VE-cadherin-CreERT2 (Cre). Expression of proNGF, inflammatory mediators,

Identification of HIF-2α-regulated genes that play a role in human microvascular endothelial sprouting during prolonged hypoxia in vitro.

During prolonged hypoxic conditions, endothelial cells change their gene expression to adjust to the low oxygen environment. This process is mainly regulated by the hypoxia-inducible factors, HIF-1α and HIF-2α. Although endothelial cells do not form sprouts during prolonged hypoxic culturing, silencing of HIF-2α partially restores sprout formation. The present study identifies novel HIF-2α-target genes that may regulate endothelial sprouting during prolonged hypoxia. The gene expression profile of primary human microvascular endothelial cells (hMVECs) that were cultured at 20 % oxygen was comp

Synthesis and Biological Evaluations of Cytotoxic and Antiangiogenic Triterpenoids-Jacaranone Conjugates.

BACKGROUND: The development of antiangiogenic agents arises as a more effective and selective therapeutic approach for the treatment of cancer. In addition to reduced acute toxicity, the efficacy of chemotherapy could be improved when administered in combination specific antiangiogenic with cytotoxic agents. The conjugation or hybridization of bifunctional molecules is one of the alternative rational design strategies for co-administration of anticancer drugs. OBJECTIVE AND METHODS: The goal of this work is to prepare the conjugates of an antiangiogenic triterpene, 3-oxo oleanolic acid, and s

Vessel microport technique for applications in cerebrovascular research.

Cerebrovascular research suffers from a lack of reliable methods with which to deliver exogenous substances effectively into the central nervous system (CNS) of small experimental animals. Here we describe a novel vessel microport surgical technique for a variety of cerebrovascular applications that is reproducible and well tolerated in mice. The procedure is based on the insertion of a vessel microport into the external carotid artery for substance delivery into the CNS via the internal carotid artery. The method results in selective substance delivery into the ipsilateral hemisphere. Other n

Collagen and microvascular alterations contribute to neuromuscular degeneration and disease progression in chronic intestinal pseudo-obstruction.

BACKGROUND: Chronic intestinal pseudo-obstruction (CIPO) is a severe gastrointestinal motility disorder that may be idiopathic or associated with systemic disease. In idiopathic cases, the pathophysiological mechanisms remain poorly defined. Although mutations in angiogenic factors have been reported in mitochondrial forms of CIPO, their role in non-mitochondrial cases is still unclear. OBJECTIVE: To investigate genetic and molecular contributors to CIPO, with a specific focus on intestinal microvasculature. METHODS: Jejunal samples from patients with CIPO were analysed by whole exome sequen

Microvascular remodeling strategy for skin rejuvenation through microenvironmental reprogramming and appendage restoration.

Skin photoaging, induced by excessive ultraviolet exposure, leads to microvascular and appendage degeneration, extracellular matrix degradation, and cellular senescence. The limited efficacy of current treatments for photoaging is partly due to underlying microvascular dysfunction. This study introduces a microneedle patch incorporating decellularized adipose-derived matrix (DAM) to enhance microvascular remodeling and mitigate photoaging. In vitro studies demonstrate that DAM enhances the function of photoaged endothelial cells via the VEGFA/PI3K/Akt pathway while simultaneously alleviating s

Association of circulating miR-29b with microvascular dysfunction and digital ulcers in patients with systemic sclerosis: a single-centre pilot study.

OBJECTIVES: Systemic sclerosis is a rare autoimmune disorder marked by vasculopathy, immune dysregulation, and progressive fibrosis. Digital ulcers (DUs) reflect severe microvascular dysfunction. This pilot, single-centre, observational study investigated whether circulating miR-29b is associated with DU occurrence, recurrence, and microvascular impairment. METHODS: Twenty-nine patients with systemic sclerosis and 17 healthy controls (HC) were evaluated. Clinical data, including DU history and recurrence, were collected. Cutaneous blood perfusion was assessed by laser speckle contrast analysi

DCIS-like invasive carcinoma of the breast with tumour-associated microvasculature mimicking myoepithelium: a diagnostic pitfall.

AIMS: Ductal carcinoma in-situ (DCIS)-like invasive carcinoma of the breast is an underdiagnosed pattern of invasive carcinoma that is challenging to separate from DCIS. We have observed some cases of DCIS-like invasive carcinoma in which smooth muscle myosin heavy chain (SMMHC) or calponin are strongly expressed in vessels tightly encircling the periphery of the tumour nodules, closely mimicking myoepithelium and potentially leading to misinterpretation as in-situ disease. We aimed to assemble and characterise a series of DCIS-like invasive carcinoma, with particular attention to presence and

An increase in micro-vessels beneath the pleural surface on computed tomography as a preoperative predictor of pleural adhesions: a prospective study.

PURPOSE: Pleural adhesions, which may be present in varying degrees and involve blood vessels, often complicate thoracic surgery. The blood flow within pleural adhesions can be identified on computed tomography (CT) as micro-vessels beneath the pleural surface (MVBP). We conducted a prospective study to assess if MVBP can be detected on preoperative CT to predict intraoperative pleural adhesions. METHODS: This prospective study investigated the preoperative CT images of patients scheduled to undergo surgery for lung tumors. MVBP was defined as positive when lung vessels were identified below

Genetic deletion of TRPM8 channels restores microvascular function and mitigates chronic kidney disease progression.

Chronic kidney disease (CKD) is a key public health issue. It initiates an inflammatory response and fibrotic processes, resulting in a gradual decline in kidney function. Yet, the detailed mechanism underlying the pathogenesis of chronic kidney disease remains unclear. Transient receptor potential melastatin 8 (TRPM8), a calcium (Ca2+) permeable, non-selective cation channel, is predominantly expressed in primary sensory neurons and plays a critical role in activating sensory neurons and regulating the inflammatory response upon stimulation. However, the interplay between TRPM8 and CKD is lar

Citrulline Supplementation Improves Microvascular Function and Muscle Strength in Middle-Aged and Older Adults with Type 2 Diabetes.

BACKGROUND: Patients with type 2 diabetes (T2D) develop vascular complications due to arginine deficiency-induced microvascular endothelial dysfunction, which is related to the loss of muscle strength (MS) associated with aging. Thus, increased nitric oxide (NO)-mediated vasodilation may improve MS. We investigated the impact of the NO precursor citrulline on microvascular function (endothelial and muscle reactivity) and MS in T2D patients. METHODS: Sixteen participants with T2D (53-72 years, nine females) were randomized to citrulline supplementation (CITS, 6 g/day) or placebo for 4 weeks pr

Endothelial TERT drives microvascular phenotype associated with coronary artery disease.

Microvascular endothelial dysfunction is a powerful predictor of future atherosclerotic cardiac events. Our previous studies revealed that under pathological states, such as coronary artery disease (CAD), the dilation mechanism switches from nitric oxide (NO)-mediated [determined by NO synthase (NOS) inhibitor] to mitochondria-derived H2O2 (determined by H2O2 scavenger). Telomerase reverse transcriptase (TERT), the catalytic subunit of telomerase, plays a noncanonical role in preventing the increase of mitochondrial reactive oxygen species in arterioles from subjects with CAD. Activation of TE

Matrine alleviates coronary microvascular dysfunction in ischemia with non-obstructive coronary artery disease mice induced by advanced glycation end products inhibition of the reactive oxygen species-mediated endoplasmic reticulum stress in cardiac microvascular endothelial cells.

OBJECTIVE: To investigate the protective effect of matrine on coronary microvascular dysfunction (CMD) induced by advanced glycation end products (AGEs) in a mouse model of ischemia with non-obstructive coronary artery disease (INOCA), with a focus on the underlying mechanisms, particularly the endoplasmic reticulum (ER) stress protein kinase R-like ER kinase (PERK)/ nuclear factor of activated T-cells (NFAT) signaling pathway. METHODS: An INOCA model was established in mice, and CMD was induced by peritoneal injections of AGEs. Matrine was administered daily via intraperitoneal injections. C

Retinal Microvascular Correlates of Cerebral Small Vessel Disease in Older Age.

BACKGROUND AND OBJECTIVES: Cerebral small vessel disease (cSVD), characterized by pathologic changes in the structure and function of small brain vessels, is detectable on brain MRI in the absence of clinical symptoms. However, imaging cerebral small vessels themselves in vivo remains costly and challenging. There is growing interest in investigating whether retinal microvascular imaging features could be proxies for changes in the brain microvasculature. Using a multipronged approach, we explored the relation of retinal microvascular characteristics with MRI markers of cSVD (MRI-cSVD). METHO

3D Imaging Reveals Complex Microvascular Remodeling in the Right Ventricle in Pulmonary Hypertension.

BACKGROUND: Pathogenic concepts of right ventricular (RV) failure in pulmonary arterial hypertension focus on a critical loss of microvasculature. However, the methods underpinning prior studies did not take into account the 3-dimensional (3D) aspects of cardiac tissue, making accurate quantification difficult. We applied deep-tissue imaging to the pressure-overloaded RV to uncover the 3D properties of the microvascular network and determine whether deficient microvascular adaptation contributes to RV failure. METHODS: Heart sections measuring 250-µm-thick were obtained from mice after pulmon
