Structure and immunogenicity of a peptide vaccine, including the complete HIV-1 gp41 2F5 epitope: implications for antibody recognition mechanism and immunogen design.

The membrane-proximal external region (MPER) of gp41 harbors the epitope recognized by the broadly neutralizing anti-HIV 2F5 antibody, a research focus in HIV-1 vaccine development. In this work, we analyze the structure and immunogenic properties of MPERp, a peptide vaccine that includes the following: (i) the complete sequence protected from proteolysis by the 2F5 paratope; (ii) downstream residues postulated to establish weak contacts with the CDR-H3 loop of the antibody, which are believed to be crucial for neutralization; and (iii) an aromatic rich anchor to the membrane interface. MPERp

Expression of truncated CTB::VP60 in tobacco exhibited no immunogenicity in rabbits.

Despite several optimizations, the production of CTB::VP60 antigen fusion proteins in tobacco is still very low. This might be due to the size of the fusion partner VP60 (579 aa). Hence, two different N-terminal truncations of VP60 were fused to CTB, either with or without an ER retention signal. CTB::VP60 expression levels, in vitro and in vivo antigenicity and immunogenicity were analyzed in plants carrying one of four different transgenes. Only one of the truncated CTB::VP60 fusions (365 aa) directed to the endoplasmic reticulum led to similar but not enhanced expression levels as compared

An ESAT-6:CFP10 DNA vaccine administered in conjunction with Mycobacterium bovis BCG confers protection to cattle challenged with virulent M. bovis.

The potency of genetic immunization observed in the mouse has demonstrated the utility of DNA vaccines to induce cell-mediated and humoral immune responses. However, it has been relatively difficult to generate comparable responses in non-rodent species. The use of molecular adjuvants may increase the magnitude of these suboptimal responses. In this study, we demonstrate that the co-administration of plasmid-encoded GM-CSF and CD80/CD86 with a novel ESAT-6:CFP10 DNA vaccine against bovine tuberculosis enhances antigen-specific cell-mediated immune responses. Antigens derived from melanocyte differentiation proteins: self-tolerance, autoimmunity, and use for cancer immunotherapy.

A large set of peptide antigens presented by class I major histocompatibility complex (MHC) molecules on human and murine melanomas and recognized by CD8+ T cells have been defined. These peptides represent attractive candidates for the development of therapeutic and/or prophylactic approaches to treat this cancer. However, the majority of the peptides that are presented by multiple tumors and recognized by T cells from multiple patients arise from proteins that are also expressed in normal melanocytes. It is expected that immune responses to such peptides will be compromised by self-tolerance

Phenotypic and functional analysis of LCMV gp33-41-specific CD8 T cells elicited by multiple peptide immunization in mice revealed the up-regulation of PD-1 expression on antigen-specific CD8 T cells.

The phenotype and function of antigen-specific CD8 T cells are closely associated with the efficacy of a therapeutic vaccination. Here we showed that multiple immunizations with LCMV gp33-41 peptide (KAV) in Freund's adjuvant could induce KAV-specific CD8 T cells with low expression of CD127 and CD62L molecules. The inhibitory receptor PD-1 was also expressed on a substantial part of KAV-specific CD8 T cells, and its expression level on KAV-specific CD8 T cells in spleen and lymph nodes was much higher when compared to those in peripheral blood. Furthermore, KAV-specific CD8 T cells could spec

Clinical disease caused by Klebsiella in 2 unrelated patients with interleukin 12 receptor beta1 deficiency.

Patients with interleukin 12 (IL-12)p40 or IL-12 receptor β1 (IL12Rβ1) deficiencies are prone to develop infections caused by mycobacteria and salmonella; other infections have only been rarely observed. In this report we describe 2 unrelated patients with complete autosomal recessive IL12Rβ1 deficiency who suffered from sepsis attributable to Klebsiella pneumoniae. A Mexican boy suffered from disseminated bacille Calmette-Guérin disease and infections caused by K pneumoniae and Candida albicans and had a fatal outcome. A Turkish girl living in France suffered from disseminated Nocardia nova i

Enhanced Antiviral Activity of Novel Umifenovir Derivatives against SARS-CoV-2: Insights from an International Collaborative Study.

Despite the availability of vaccines and treatments for COVID-19, the emergence of new SARS-CoV-2 variants continues to challenge vaccine-induced immunity, emphasizing the need to develop antiviral therapies to combat these variants and other viruses. Umifenovir (UMF), marketed as Arbidol, is a broad-spectrum antiviral drug approved in Russia and China for influenza viruses A and B. Recent studies have suggested its potential against SARS-CoV-2, demonstrating its ability to inhibit viral replication and obstruct viral entry by targeting the spike protein, despite having low oral bioavailabilit

Polysaccharide microparticles as an oral vaccine platform: proof-of-concept in male rats using a GnRH-targeting fusion antigen.

BACKGROUND: Immunocastration through vaccination against gonadotropin-releasing hormone (GnRH) represents a humane alternative to surgical castration in animal production. However, oral delivery of protein antigens remains challenging due to gastrointestinal degradation and poor mucosal immunogenicity. Polysaccharide microparticles (PSMs), particularly alginate-chitosan systems, offer protection against gastric degradation and enhanced mucosal uptake. OBJECTIVE: To evaluate whether alginate-chitosan microparticles entrapping a recombinant GnRH-fusion protein (GF45) can elicit an immunocastrat

Construction and Immunogenicity Evaluation of a Digestive Protein-Based Chimeric Antigen Composed of Cathepsin L1, Cathepsin B1, and Saposin-like Protein 2 from Fasciola gigantica.

Fasciolosis, caused by the liver fluke Fasciola gigantica, remains a major parasitic disease affecting livestock in tropical regions and results in substantial economic losses. Although anthelmintic drugs are widely used for disease control, increasing reports of drug resistance highlight the need for alternative strategies such as vaccination. In this study, a recombinant digestive protein-based chimeric antigen (rFgCHI-DP) composed of three F. gigantica antigens-cathepsin L1 (FgCL1), cathepsin B1 (FgCB1), and saposin-like protein 2 (FgSAP2)-was designed and expressed in Escherichia coli. The Human Cytomegalovirus UL116 Gene Encodes an Envelope Glycoprotein Forming a Complex with gH Independently from gL.

UNLABELLED: Human cytomegalovirus (HCMV) is a major cause of morbidity and mortality in transplant patients and is the leading viral cause of birth defects after congenital infection. HCMV infection relies on the recognition of cell-specific receptors by one of the viral envelope glycoprotein complexes. Either the gH/gL/gO or the gH/gL/UL128/UL130/UL131A (Pentamer) complex has been found to fulfill this role, accounting for HCMV entry into almost all cell types. We have studied the UL116 gene product, a putative open reading frame identified by in silico analysis and predicted to code for a se

A nanoemulsion-adjuvanted trivalent neuraminidase vaccine confers cross-protection against divergent influenza viruses.

Developing universal influenza vaccines (UIVs) capable of eliciting broad protection remains a critical public health priority. Neuraminidase (NA), a relatively conserved surface antigen of influenza viruses, is a promising target for UIV design. Here, we produced full-length trivalent NA proteins via a baculovirus-insect cell expression system, matching the WHO-recommended strains (H1N1, H3N2, and B/Victoria) for the 2024-2025 season. Immunogenicity evaluation of N2NA (derived from H3N2) in mice demonstrated that nanoemulsion (NE) adjuvant significantly enhanced NA-inhibiting (NI) antibody ti

Conserved HLA binding peptides from five non-structural proteins of SARS-CoV-2-An in silico glance.

Coronavirus Disease 2019 (COVID-19) is a dangerous global threat that has no clinically approved treatment yet. Bioinformatics represent an outstanding approach to reveal key immunogenic regions in viral proteins. Here, five severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) non-structural proteins (NSPs) (NSP7, NSP8, NSP9, NSP12, and NSP13) were screened to identify potential human leukocyte antigen (HLA) binding peptides. These peptides showed robust viral antigenicity, immunogenicity, and a marked interaction with HLA alleles. Interestingly, several peptides showed affinity by HLA

A synthetic peptide vaccine directed against the 2ß2-2ß3 loop of domain 2 of protective antigen protects rabbits from inhalation anthrax.

The current vaccines for anthrax in the United States and United Kingdom are efficacious in the two most accepted animal models of inhalation anthrax, nonhuman primates and rabbits, but require extensive immunization protocols. We previously demonstrated that a linear determinant in domain 2 of Bacillus anthracis protective Ag (PA) is a potentially important target for an epitope-specific vaccine for anthrax, as Abs specific for this site, referred to as the loop-neutralizing determinant (LND), neutralize lethal toxin in vitro, yet are virtually absent in PA-immunized rabbits. Chimeric virus-like particles of nodavirus displaying human and avian influenza A M2e induce conserved anamnestic protective immunity against human and avian influenza A viruses.

The conserved matrix 2 ectodomain (M2e) peptides of influenza A viruses (IAVs) have been employed in developing universal vaccines. However, interspecies M2e variations between human and avian strains, including those associated with host adaptation during zoonotic and reverse-zoonotic transmission, result in notable antigenic and immunogenic differences. Therefore, we fused two copies each of M2e peptides from human and avian IAVs at the C-terminus of Macrobrachium rosenbergii nodavirus capsid protein (NvC), which self-assembled into virus-like particles (VLPs). This chimeric VLP provided 100

Intranasal Delivery of MVA Vector Vaccine Induces Effective Pulmonary Immunity Against SARS-CoV-2 in Rodents.

Antigen-specific tissue-resident memory T cells (Trms) and neutralizing IgA antibodies provide the most effective protection of the lungs from viral infections. To induce those essential components of lung immunity against SARS-CoV-2, we tested various immunization protocols involving intranasal delivery of a novel Modified Vaccinia virus Ankara (MVA)-SARS-2-spike vaccine candidate. We show that a single intranasal MVA-SARS-CoV-2-S application in mice strongly induced pulmonary spike-specific CD8+ T cells, albeit restricted production of neutralizing antibodies. In prime-boost protocols, intra

A SARS-CoV-2 variant-induced NTD-targeting antibody enhances viral infection via a distinctive binding mode.

SARS-CoV-2 infection elicits both neutralizing and non-neutralizing monoclonal antibodies (mAbs), primarily targeting to the N-terminal domain (NTD), receptor-binding domain (RBD), and S2 subunit of the spike protein. Notably, a unique subset of NTD-targeting mAbs isolated from prototype Wuhan-Hu-1 strain infected donors displayed a capacity of facilitating the viral infection independent of the fragment crystallizable (Fc) region in vitro. However, the rapid evolution of SARS-CoV-2 variants, particularly with NTD mutations, has led to widespread immune evasion. Whether SARS-CoV-2 variants cou

Gel-Based Drug Delivery Platforms: A Critical, Mechanistic Review of Design, Cross-Linking, and Disease-Specific Translation (2010-2026).

Gel-based novel drug delivery systems (NDDS) occupy a mechanistically distinct niche among controlled-release platforms because they decouple three design variablesnetwork cross-link density, continuous-phase polarity, and stimulus sensitivitythat in particulate carriers (liposomes, polymeric nanoparticles) are often interdependent. This critical review synthesizes 102 primary and secondary sources published predominantly between 2010 and 2026 to interrogate, rather than merely catalog, how hydrogels, organogels, aerogels, nanogels, in situ gelling systems, and hydrogel-forming microneedles ha

Exploring the Intrinsic Structural Plasticity and Conformational Dynamics of Human Beta Coronavirus Spike Glycoproteins.

The spike (S) glycoprotein of human beta coronaviruses (HCoVs) is central to viral entry, receptor engagement, and immune evasion. Here, we present an in-depth computational analysis of spike conformational dynamics across HCoVs, with a focus on SARS-CoV-2 and its variants. Leveraging a large cryo-EM structural ensemble and integrative modeling approaches, we dissect the intrinsic plasticity and variant-specific motions of the spike protein. Our results show that, despite substantial sequence divergence, HCoV spikes retain the ability to sample open and closed receptor-binding domain (RBD) sta

Comprehensive analysis of the succinylome in Vero cells infected with peste des petits ruminants virus Nigeria 75/1 vaccine strain.

BACKGROUND: Peste des petits ruminants virus (PPRV) is currently the only member of the Morbillivirus caprinae species within the genus Morbillivirus of the family Paramyoxviridae. PPRV causes a highly contagious disease in small ruminants, especially goats and sheep. Succinylation is a newly identified and conserved modification and plays an important role in host cell response to pathogen infection. However, the extent and function of succinylation in Vero cells during PPRV infection remains unknown. RESULTS: In this study, a global profile of the succinylome in Vero cells infected with PPR
