Topographical distribution of lactate dehydrogenase activity in human clear eye lenses and in lenses with different types of senile cataract: a histochemical investigation.

BACKGROUND: Homogenates of human clear lenses show an age-dependent reduction of enzyme activities. Topographical patterns of enzymes in clear and cataractous lenses can be visualized by histochemistry. MATERIAL AND METHODS: Human lenses were characterized by slit-lamp investigations as bearing different types of senile cataracts. Subsequently, lenses were removed by intracapsular extraction. Clear human lenses served as controls. Bovine lenses served to standardize freeze-cutting and incubation for lactate dehydrogenase histochemistry. RESULTS: Bovine lenses show a sharp demarcation between

Keratotomy model of pseudomonas keratitis: gentamicin chemotherapy.

BACKGROUND: Chemotherapy of bacterial keratitis requires frequent application of antibiotic drops. Collagen shields containing antibiotics could reduce the need for frequent antibiotic application. To determine the effect of gentamicin-containing collagen shields and gentamicin drops on Pseudomonas keratitis, a new keratotomy model of infection was employed. METHODS: Model--contact lenses (58% water content) presoaked in 1% bovine serum albumin and exposed to 10(8) colony forming units per mL of Pseudomonas aeruginosa strain 27853, were found to reproducibly retain 5.9 (log base 10) colony-fo

Morphological studies of an ion-dependent perinuclear cataract model.

The perinuclear region of the rabbit lens is susceptible to alterations in the ionic composition of incubation medium. Rabbit lenses and a comparable cell type, red blood cells, were stressed during ex vivo incubations in isotonic modified Earle's medium with 131 mM NaCl replaced by either 232 mM sucrose or 131 mM choline chloride at pH 7.2 (normal) or 9.2. Our parallel NMR study revealed that these experimental media maintain normal intracellular pH and phosphorus metabolite levels. The present study demonstrates that lens transparency, normal fiber cell ultrastructure and volume were maintai

Direct Ink Writing of an Orthogonal Collagen-Based Membrane for Sutureless Corneal Repair.

The native corneal stroma is composed of highly organized collagen lamellae with alternating orientations. However, reconstructing this complex architecture while avoiding suture-associated complications remains a major challenge in corneal repair. Herein, building on the previously established pseudo-polyrotaxane and collagen (PPR-COL) system, an orthogonally structured PPR-COL membrane fabricated via direct ink writing (DIW) was integrated with a bioadhesive layer composed of gelatin methacryloyl and oxidized hyaluronic acid for sutureless fixation. The concentration-dependent rheological be

Comparison of Peripapillary and Submacular Choroidal Changes After Trabeculectomy and Phaco-Trabeculectomy in Open-Angle Glaucoma.

PURPOSE: Choroidal vascular and structural alterations are increasingly recognized as relevant to intraocular pressure (IOP) regulation and glaucoma pathophysiology. Surgical IOP reduction induces hemodynamic and biomechanical changes that may affect choroidal thickness and perfusion, particularly in the peripapillary and submacular regions. Although trabeculectomy is known to cause early postoperative choroidal expansion, the additional impact of concomitant phacoemulsification remains unclear. We compared early regional choroidal responses after trabeculectomy (Trab) alone and phaco-trabecul

Integrative analytical and statistical framework for optimization of multiplex qPCR detection of TGFBI mutations in refractive surgery candidates.

Refractive surgery can unmask or accelerate transforming growth factor-β-induced (TGFBI)-related corneal dystrophies that are undetectable by routine slit-lamp examination, creating a clear need for a rapid, standardized, preoperative genetic screening. We developed a multiplex, allele-specific real-time quantitative polymerase chain reaction (qPCR) panel targeting five high-frequency TGFBI hotspots (R124C/L/H, R555W/Q) and built a statistics-driven analytical framework to optimize assay decisions. Receiver operating characteristic (ROC) analysis defined locus-specific cycle threshold (CT) cut

[Anterior segment changes in mucopolysaccharidoses].

BACKGROUND AND AIM: Mucopolysaccharidoses (MPS) are rare lysosomal storage disorders in which the accumulation of undegraded glycosaminoglycans leads to progressive multiorgan and ocular changes. The anterior segment of the eye is particularly affected, with type-specific, distinct corneal and chamber angle alterations. The aim of this study was to characterize anterior segment changes in different MPS types based on structural and functional examinations using clinical and imaging parameters. METHODS: In a clinical cross-sectional study, patients with MPS (types I, II, III, IV, VI) were comp

Explantation of Implantable Collamer Lenses due to high intraocular pressure in a highly hyperopic patient: a case report.

BACKGROUND: Implantable Collamer Lens (ICL) implantation is an effective treatment for correcting high hyperopia. However, late anterior segment complications remain insufficiently characterized. The simultaneous occurrence of angle narrowing, pigment dispersion, and intraocular pressure (IOP) elevation nearly two decades after implantation of a non-central-port ICL is exceptionally rare. This case illustrates the diagnostic and surgical challenges of late-onset angle narrowing in hyperopic eyes and outlines strategies to minimize iris injury during ICL explantation.
