Scorpion (Odontobuthus doriae) venom induces apoptosis and inhibits DNA synthesis in human neuroblastoma cells.

Scorpion and its organs have been used to cure epilepsy, rheumatism, and male impotency since medieval times. Scorpion venom which contains different compounds like enzyme and non-enzyme proteins, ions, free amino acids, and other organic inorganic substances have been reported to posses antiproliferative, cytotoxic, apoptogenic, and immunosuppressive properties. We for the first time report the apoptotic and antiproliferative effects of scorpion venom (Odontobuthus doriae) in human neuroblastoma cells. After exposure of cells to medium containing varying concentrations of venom (10, 25, 50, 1

The type III neurofilament peripherin is expressed in the tuberomammillary neurons of the mouse.

BACKGROUND: Peripherin, a type III neuronal intermediate filament, is widely expressed in neurons of the peripheral nervous system and in selected central nervous system hindbrain areas with projections towards peripheral structures, such as cranial nerves and spinal cord neurons. Peripherin appears to play a role in neurite elongation during development and axonal regeneration, but its exact function is not known. We noticed high peripherin expression in the posterior hypothalamus of mice, and decided to investigate further the exact location of expression and function of peripherin in the mo

Oxidative stress and transcriptional regulation in Alzheimer disease.

Alzheimer disease (AD) is defined by progressive impairments in memory and cognition and by the presence of extracellular neuritic plaques and intracellular neurofibrillary tangles. However, oxidative stress and impaired mitochondrial function always accompany AD. Mitochondria are a major site of production of free radicals [ie, reactive oxygen species (ROS)] and primary targets of ROS. ROS are cytotoxic, and evidence of ROS-induced damage to cell membranes, proteins, and DNA in AD is overwhelming. Nevertheless, therapies based on antioxidants have been disappointing. Thus, alternative strateg

Early downregulation of IGF-I decides the fate of rat retinal ganglion cells after optic nerve injury.

Retinal ganglion cells (RGCs) die by apoptosis after optic nerve injury. A number of reports have separately shown changes in pro-apoptotic proteins such as the Bcl-2 family members following optic nerve injury. However, induction time of these apoptotic signals has not been identified due to different treatments of the optic nerve, and insufficient time intervals for measurements. Therefore, the stream of cell death signals is not well understood. In the present study, we systematically reinvestigated a detailed time course of these cell death/survival signals in the rat retina after optic ne

The prolyl isomerase Pin1 regulates amyloid precursor protein processing and amyloid-beta production.

Neuropathological hallmarks of Alzheimer's disease are neurofibrillary tangles composed of tau and neuritic plaques comprising amyloid-beta peptides (Abeta) derived from amyloid precursor protein (APP), but their exact relationship remains elusive. Phosphorylation of tau and APP on certain serine or threonine residues preceding proline affects tangle formation and Abeta production in vitro. Phosphorylated Ser/Thr-Pro motifs in peptides can exist in cis or trans conformations, the conversion of which is catalysed by the Pin1 prolyl isomerase. Pin1 has been proposed to regulate protein function

Methylmercury induces activation of Notch signaling.

Methylmercury (MeHg) toxicity in humans manifests deficits in neurological function. Cases of prenatal exposure to mercury have established that the developing nervous system is most highly susceptible to perturbation by MeHg. At a cellular level, MeHg-induced defects result from altered neuronal proliferation, migration and pathfinding. However, the molecular targets of MeHg that give rise to these outcomes are not fully understood. In an overall effort to identify the fundamental molecular targets of MeHg in neural development, we have explored the effects of MeHg on cell surface receptor fu

Neonatal loss of gamma-aminobutyric acid pathway expression after human perinatal brain injury.

OBJECT: Perinatal brain injury leads to chronic neurological deficits in children. Damage to the premature brain produces white matter lesions (WMLs), but the impact on cortical development is less well defined. Gamma-aminobutyric acid(GABA)ergic neurons destined for the cerebral cortex migrate through the developing white matter and form the subplate during late gestation. The authors hypothesized that GABAergic neurons are vulnerable to perinatal systemic insults in premature infants, and that damage to these neurons contributes to impaired cortical development. METHODS: An immunohistochemi

Identification of PINCH in Schwann cells and DRG neurons: shuttling and signaling after nerve injury.

Particularly interesting new cysteine-histidine rich protein (PINCH) is a double zinc finger domain (LIM)-only adapter protein that functions to recruit the integrin-linked kinase (ILK) to sites of integrin activation. Genetic studies have shown that PINCH and ILK are required for integrin signaling. Since integrin activation is associated with Schwann cell migration, neurite outgrowth and regeneration, this study examined PINCH in the normal peripheral nervous system and after chronic constriction injury (CCI) in adult Sprague-Dawley rats. Immunohistochemistry identified PINCH immunoreactivit

Effects of prolonged exposure to nanomolar concentrations of methylmercury on voltage-sensitive sodium and calcium currents in PC12 cells.

The neurotoxicant methylmercury (CH(3)Hg(+)) inhibits voltage-sensitive Na(+) and Ca(2+) currents in neuronal preparations following acute, in vitro, exposure. In the present study, effects on voltage-sensitive Na(+) (I(Na)) and Ca(2+) (I(Ca)) currents in pheochromocytoma (PC12) cells were examined following prolonged exposure to CH(3)Hg(+). When PC12 cells cultured in the presence of nerve growth factor (NGF) for 7 days ('primed') were replated in the presence of NGF and 30 nM CH(3)Hg(+), I(Ca), but not I(Na), amplitude was reduced (29%) significantly approximately 24 h later. Pathological correlates of late-onset dementia in a multicentre, community-based population in England and Wales. Neuropathology Group of the Medical Research Council Cognitive Function and Ageing Study (MRC CFAS).

BACKGROUND: We have undertaken a large unselected, community-based neuropathology study in an elderly (70-103 years) UK population in relation to prospectively evaluated dementia status. The study tests the assumption that dementing disorders as defined by current diagnostic protocols underlie this syndrome in the community at large. METHODS: Respondents in the Medical Research Council Cognitive Function and Ageing Study were approached for consent to examine the brain at necropsy. Dementia status was assigned by use of the automated geriatric examination for computer-assisted taxonomy algori

The apolipoprotein E epsilon4 allele frequency is normal in fronto-temporal dementia, but correlates with age at onset of disease.

The apolipoprotein (apoE) epsilon4 allele was studied in fronto-temporal dementia (FTD), a diagnostic category including the specific disorders Pick's disease and frontal lobe degeneration of non-Alzheimer type (FLD). These dementing diseases have neuronal and synaptic degeneration in common with Alzheimer's disease (AD), for which the presence of the apoE epsilon4 allele is a known risk factor, and lowers the age of onset of disease. Previous studies on the apoE epsilon4 allele frequency in FTD have been inconclusive. The structural hallmarks of AD, allegedly linked to apoE presentation, neur

Loss of Selenot in POMC Neurons Disrupts ER Homeostasis and Induces A Senescent-like Phenotype.

AIMS: Selenoprotein T (SELENOT) is an endoplasmic reticulum (ER)-resident thioredoxin-like oxidoreductase implicated in redox and calcium (Ca2+) homeostasis and protein quality control. Although SELENOT is highly expressed in the brain, its role in hypothalamic pro-opiomelanocortin (POMC) neurons, which regulate energy balance, remains poorly understood. We investigated whether SELENOT is required for maintaining ER homeostasis and neuronal integrity in POMC neurons. RESULTS: Using CRISPR-edited and siRNA-silenced POMC neuronal models together with a POMC-specific SELENOT knockout mouse, we d

Integrated electrophysiological and payload-release assessment of neuronal responses to antibody-drug conjugates in human iPSC-derived sensory neurons: implications for in vitro neurotoxicity evaluation.

Antibody-drug conjugates (ADCs) enable tumor-specific drug delivery; however, target antigen-independent adverse events, including peripheral neuropathy, remain a significant clinical concern. The mechanisms underlying ADC-induced neurotoxicity, particularly the relationship between payload release and alterations in neuronal function, are not yet fully understood. In this study, we established an analytical platform that integrates microelectrode array (MEA) recordings using human induced pluripotent stem cell (iPSC)-derived sensory neurons with quantitative measurement of payload levels dete

A bilayer nerve conduit integrating aligned guidance with an RGI peptide/retinoic acid-loaded hydrogel for peripheral nerve regeneration.

The repair of long-gap peripheral nerve defects remains challenging, as regeneration depends on temporally coordinated bioactive regulation and spatially organized guidance. To address this, we developed a spatiotemporally coordinated nerve conduit integrating an injectable thermosensitive poly(amino acid) hydrogel with an aligned-random bilayer poly(ε-caprolactone) (PCL) wall. A brain-derived neurotrophic factor (BDNF)-mimetic RGI peptide was covalently tethered to mPEG-PMet-PLysMA (P), and retinoic acid (RA) was physically incorporated to form P-RGI@RA. Aligned luminal fibers promoted Schwan

Histomorphological and ultrastructural analysis of vibrissal follicle-sinus complexes in the dog (Canis lupus familiaris).

Trimming of vibrissae in dogs (Canis lupus familiaris) for cosmetic reasons remains a common practice, particularly among poodle breeds and show dogs. In discussions surrounding this practice, it has been claimed that domestic dogs would have fewer and structurally less developed sinus hairs than other mammals, and that, especially in poodles, vibrissae would lack functional relevance, although such narratives represent popularized assumptions rather than systematically supported peer-reviewed evidence. The follicle-sinus complexes (FSCs) of various species, including cats (Felis catus), rats

Diselenide-bridged mesoporous silica nanoplatform for baicalin delivery facilitates spinal cord injury repair via CHCHD2-mediated mitochondrial homeostasis restoration.

During the secondary phase of spinal cord injury (SCI), excessive accumulation of reactive oxygen species (ROS), mitochondrial dysfunction and amplified inflammatory cascades reinforce one another, forming a vicious cycle that drives progressive neuronal damage and limits functional recovery. To enhance lesion-site drug exposure and microenvironment-responsive delivery, we engineered a biomimetic nanoplatform (Ba@Se-MSN&BV2) by loading baicalin into diselenide-bridged mesoporous silica nanoparticles (MSNs) and cloaking them with BV2 microglial membranes, enabling lesion-targeted accumulation a

Phenotyping iPSC-derived neurons from Angelman patients and partial rescue via JNK pathway inactivation.

BACKGROUND: Angelman Syndrome (AS, MIM #105,830) is a rare neurodevelopmental disorder caused by imprinting defects, and it is marked by motor impairment, intellectual disability, speech limitations, and seizures. The condition arises due to the loss of function in neurons of the maternally-inherited UBE3A gene, mapping in the imprinted chromosomal region 15q11.2-q13, through a variety of genetic and epigenetic mechanisms. UBE3A encodes a ubiquitin E3 ligase, which is essential for synaptic plasticity. Most studies of AS have been performed on animal models, while induced pluripotent stem cell

Visualizing Specific Tubulin Isotypes and Pathogenic Variants in Cellular Microtubule Arrays.

Eukaryotic cells depend on dynamic microtubule arrays to execute a wide range of functions vital for life. These microtubule filaments are formed through the polymerization of α/β-tubulin proteins, which can be generated from numerous tubulin genes, or isotypes. In the neuron, microtubule networks are responsible for a myriad of specialized functions, including neuronal migration, neurite extension, and long-range cargo transport. Pathogenic mutations in tubulin genes can result in a wide range of neurodevelopmental defects, collectively termed tubulinopathies. How distinct mutations within di
