Prepartum management factors associated with the development of calcium imbalance during the first days postpartum in grazing dairy cows in Uruguay.

To evaluate the association between calcium (Ca) imbalance postpartum, animal-associated data, and prepartum management practices in Holstein dairy cows, we conducted a cross-sectional study based on a convenience sample of 122 dairy farms (204 primiparous and 661 multiparous cows) in Uruguay. Cows were blood sampled once between days 1 and 3 DIM after parturition for metabolite analysis. At the time of sampling, data were collected on management, housing, and feeding. To evaluate the association between Ca imbalance and animal-associated data (parity, body condition, high NEFA (non-esterified

Low Prevalence of Hypomagnesemia in Long-term Recipients of Proton Pump Inhibitors in a Managed Care Cohort.

BACKGROUND & AIMS: Chronic intake of proton pump inhibitors (PPI) has been associated with hypomagnesemia, but prevalence of PPI-associated hypomagnesemia is not known. METHODS: We examined the prevalence of hypomagnesemia among long-term PPI recipients by using a large health maintenance organization database. We collected data on 10,167 participants eligible for chronic drug prescriptions from 2008 through 2013. Adult subjects receiving continuous PPI therapy for ≥ 6 months between 2008 and 2013 and ≥ 1 serum magnesium determination(s) were identified. Patients with any magnesium levels les

[Level of brain-derived neurotrophic factor in the microenvironment of the neuron-astrocyte co-culture system by Mg2+-free-induced seizure-like discharge].

OBJECTIVE: To study the level of brain-derived neurotrophic factor (BDNF) in the microenvironment of the neuron-astrocyte co-culture system by Mg2+-free-induced seizure-like discharge and analyze the source of BDNF. METHODS: Hippocampal neurons (N) of fetal rats and astrocytes (AST) of neonatal rats were purified and divided into four groups, included control N (Con N) group, Mg2+-free treated N (Mg2+-free N) group, control N+AST co-culture (Con N+AST) group and Mg2+-free treated N+AST co-culture (Mg2+-free N+AST) group. The Mg2+-free treated groups were exposed to Mg2+-free media for 3 hrs t

Nausea, emesis, and muscle weakness in a pregnant adolescent.

BACKGROUND: Gitelman syndrome is a rare autosomal recessive disorder that presents in early adulthood with fatigue, muscle cramps and electrolyte abnormalities. CASE: A 17-year-old African-American woman presented at 17 weeks of pregnancy with nausea, emesis, profound lower extremity proximal muscle weakness, hypokalemia, and hypomagnesemia. After a thorough evaluation, Gitelman syndrome was diagnosed. The patient was maintained on high levels of potassium and magnesium supplementation throughout the rest of her pregnancy and delivered a healthy infant. CONCLUSION: In pregnancy, nausea and e

[Atypical electrolytic disorders in patients with diseases of the blood system].

AIM: To describe and analyze rare iatrogenic disturbances of water-electrolyte and acid-base balances in systemic blood diseases. MATERIAL AND METHODS: Five cases of life-threatening electrolytic imbalance resultant from therapy in patients with hemoblastoses and a patient with acute intermittent porphyria (AIP). RESULTS: Hyperkalemia arose in 2 patients treated with cyclosporin A and trimethoprim-sulfamethoxasole. One patient developed severe arrhythmia because of hypomagnesemia following long-term therapy with aminoglycosides and amphotericin B. In one AIP patient hyponatremia ending in co

Effects of extracellular ATP on Ca2+ mobilization in Madin Darby canine kidney (MDCK) cells.

Previous studies have demonstrated that ATP activates calcium-dependent K+ channels thereby leading to plasma membrane hyperpolarization and chloride secretion in MDCK cells. However, the signaling pathway involved in regulating ATP-evoked Ca2+ mobilization in MDCK cells remains unclarified. The present study was performed to elucidate the mechanisms underlying ATP-evoked calcium mobilization and inositol phospholipid turnover in MDCK cells. Our results indicate that ATP exerted its effects by activating the P2u subtype purinoceptor because both ATP and UTP but not adenosine caused Ca2+ mobili

Genetic heterogeneity in tubular hypomagnesemia-hypokalemia with hypocalcuria (Gitelman's syndrome).

To better clarify the genetic inheritance of primary tubular hypomagnesemia-hypokalemia with hypocalciuria, or Gitelman's syndrome (GS), we studied eight families (10 patients aged 11 to 22 years; 16 parents; 9 siblings) in which at least one offspring had GS (plasma magnesium < 0.65 mmol/liter; plasma potassium < 3.6 mmol/liter; high magnesium and potassium fractional excretions; molar urinary calcium/creatinine < 0.10). Two families each had two offspring of different sex with GS, who all had tetanic episodes and/or marked weakness during childhood or adolescence, whereas in three other fami

Effects of urea on K-Cl cotransport in sheep red blood cells: evidence for two signals of swelling.

The activation proceeds with a delay, like activation by swelling. Swelling of cells in urea activates K uptake further, but with no delay. Inactivation after removal of urea also proceeds without delay. With cotransport partially activated by reducing intracellular Mg concentration ([Mg]i) or with staurosporine, urea did not activate cotransport further. However, swelling activated cotransport further in these two types of cells. In terms of the three-state process for swelling-activation of K-Cl cotransport (P. B. Dunham, J. Klimczak, and P. J. Logue, J. Gen. Physiol. 101: 733-765, 1993), th

In vitro studies on the broad spectrum anticonvulsant loreclezole in the hippocampus.

In hippocampal slices from guinea-pig a paired-pulse stimulation protocol was used to examine the effects of loreclezole, R-(+)-etomidate, phenobarbital and pentobarbital on orthodromic and antidromic GABAergic neuronal inhibition in the CA1 region. All four compounds increased orthodromic GABAergic inhibition, with R-(+)-etomidate and pentobarbital inducing a quantitatively larger effect than loreclezole and phenobarbital. Only R-(+)-etomidate and pentobarbital increased antidromic GABAergic inhibition. We propose that all four compounds are anticonvulsant by increasing feed-forward dendritic

The influence of dietary calcium on kidney calcification and renal function in rats fed high-phosphate diets.

This study was carried out to test the hypothesis that kidney calcification caused by high dietary phosphorus intake can be prevented by increasing the calcium content of the diet, because the latter reduces the intestinal P absorption. The investigation was conducted in rats fed semipurified diets with either a low (0.15%) or a high (1.20%) P content. Although dietary Ca supplementation (as CaCO3 or CaCl2) reduced the intestinal P absorption, it did not prevent kidney calcification induced by high dietary P. On the contrary, when CaCO3 was supplemented for 16 weeks, renal calcification increa

Restoration of normal appearance, growth behavior, and calcium content to transformed 3T3 cells by magnesium deprivation.

A spontaneously transformed clone was isolated from repeatedly passaged BALB/c 3T3 cells. The transformed cells were rounded or slender and elongated, were randomly arranged in an overlapping pattern, grew to high cell density, and had a low requirement for serum. The rates of multiplication and DNA synthesis of the nontransformed and the transformed lines were reduced for several days by drastic reduction in the Mg2+ concentration of the medium, but the rate of DNA synthesis in the Mg2+-deprived cultured increased after 6--8 days, suggesting an adaptation of the cells or a change in local env

Hypocalcemic, hypophosphatemic rickets in rat pups suckling vitamin D-deprived mothers.

The effects of maternal vitamin D deprivation on rat pups were examined in terms of serum levels of Ca, phosphorus (P), and 25-hydroxyvitamin D (250HD), ash content, and histological appearance of long bones. In each of 10 experiments, litters of 8--10 young were examined at 2 or 3 different ages within the range of 5--57 days. Pregnant mothers were fed a diet containing 0.4% Ca, and 0.4% P and either no vitamin D(--D) or 5 IU D3/g diet (+D). Mothers fed the --D diet developed marked hypocalcemia (4--5 mg/dl) during lactation, but no change in milk Ca or P. The earliest changes in pups suckled

[Enzymatic properties in muscle membranes].

A study in the enzymatic properties of muscle membranes established that sarcolemma of the rabbit skeletal muscles contains the Ca2+-ATPase system which does not require Mg2+ for manifestation of ions activity. By some kinetic properties it differs from ATPase of myosin. The complex Ca-ATP2+ is a substrate of Ca2+-ATPase. Ions of a series of bivalent metals inhibit the latter as well as the passive transport of Ca2+, that may evidence for a definite relation of Ca2+-ATPase with Ca+2 transport in skeletal muscles. Acetyl cholinesterase and AMP-aminohydrolase are strongly bound with the sarcolem
