Characterization of the complete mitochondrial genome of Rhododendron simsii var. putuoense and its comparative evolutionary analysis.

Rhododendron simsii var. putuoense is a coastal-endemic plant with significant ecological and genetic resource value. Although its chloroplast genome (cpDNA) was recently characterized, its mitochondrial genome (mtDNA) remains unexplored. Here, we report the first complete assembly and annotation of the R. simsii var. putuoense mtDNA using PacBio HiFi long-read sequencing. The genome sequence is 658,161 bp in length, organized as two subgenomic linear molecules (a major one of 633,105 bp and a minor one of 25,056 bp), and encodes 33 protein-coding genes (PCGs), 26 tRNAs, three rRNAs and 323 OR

Association of artificial sweeteners intake and risk of CKD: a prospective cohort study.

BACKGROUND: Artificial sweeteners (AS) are widely used in foods and beverages as low-calorie alternative, but their safety remains controversial. Robust epidemiological evidence regarding the impact of AS on kidney health, especially the risk of chronic kidney disease (CKD) is currently lacking. METHODS: The study included 156,000 participants from the UK Biobank cohort with a median follow-up of 13.3 years. The intake of AS from various sources was evaluated through a 24-h dietary recall. Incident CKD was defined as a newly diagnosed by the ICD codes and follow-up eGFR from study records and

The role of epigenetic modifications in cancer-associated fibroblasts.

Cancer-associated fibroblasts (CAFs) constitute a heterogeneous population of stromal cells whose tumor-modulating activities are highly context-dependent. Unlike cancer cells, CAFs rarely harbor stable genetic mutations; instead, their phenotypic plasticity is largely driven by reversible epigenetic reprogramming. Within the tumor microenvironment (TME), CAFs function as key regulators by secreting extracellular matrix (ECM) components, paracrine growth factors, cytokines, and metabolic intermediates that collectively promote tumor growth, invasion, and therapeutic resistance. Recent advances

Biomolecular condensates as dynamic regulators of musculoskeletal homeostasis, disease, and therapeutic challenges.

Biomolecular condensates are membraneless assemblies that concentrate proteins, nucleic acids, and other biomolecules into dynamic cellular compartments. Liquid-liquid phase separation (LLPS) is one important route by which such condensates form, particularly when multivalent interactions generate liquid-like, reversible assemblies. However, not every condensate or disease-associated assembly should be explained solely by LLPS. In the musculoskeletal system, condensates have been linked to transcription, signal transduction, RNA metabolism, stress responses, tissue development, homeostasis, an

From Misdiagnosis to Discovery: Expanding the Spectrum of Spliceosomopathies.

BACKGROUND: Defining a genetic diagnosis is a challenging and stepwise process, often limited by current knowledge perspectives. Alterations in non-coding RNAs and in their modulating mechanisms are now emerging as one of the main possible explanations underpinning many unsolved cases of neurodevelopmental disorders. The latest description concerns the PUS7 gene, which encodes a pseudouridine synthase (PUS) enzyme, which has been causatively associated with an extremely rare recessive disease characterized by postnatal microcephaly, neurodevelopmental impairment, and minor dysmorphisms. Here,

First detection of a recombinant enterovirus G carrying a torovirus papain-like cysteine protease gene from India.

Enterovirus G (EV-G) comprises 20 genotypes (G1-G20) associated with various disease conditions in pig populations. Recombination among homologous EV-G drives virus evolution; however, recombination with a virus from a heterologous family is a rare event. Currently, two types of recombinant EV-G strains containing torovirus papain-like cysteine protease (PLCP) at the 2C-3A junction (Type I) and in place of structural proteins (Type II) are reported. The present study was conducted to reveal the RNA virome in diarrheic piglet feces, retrieving the genome coding for a complete polyprotein of EV-

Antibody Responses to the Conserved Plasmodium falciparum Vacuolar Sorting Protein 29 in the Brazilian Amazon.

Vacuolar Protein Sorting 29 (VPS29) is a highly conserved subunit of the retromer complex, which mediates retrograde transport from endosomes to the Golgi apparatus and plays a critical role in membrane trafficking, protein recycling, and organelle biogenesis. In Plasmodium falciparum, the retromer has been implicated in the formation of apical organelles essential for parasite invasion and replication. In this study, we investigated naturally acquired antibody responses to P. falciparum VPS29 (PfVPS29) and the genetic diversity of the vps29 gene in isolates from three malaria-endemic areas of

Epigenetic control of transposable elements in brain and brain disorders.

Once considered somewhere between inert ('junk DNA') and parasitic ('selfish genetic elements'), transposable elements (TEs) are increasingly appreciated as inseparable from the healthy functioning of eukaryotic genomes. The ubiquitous TE sequences that make up about half of the human genome are sources of the physical genetic material necessary for establishing distinct domains of chromatin architecture, generating non-coding RNAs (ncRNAs), expanding transcription factor regulatory networks, and providing cis-regulatory elements to enable complex gene expression patterns. Given the critical r

A high-penetrance intergenic variant at 9p21 confers melanoma susceptibility.

Approximately 10% of cutaneous malignant melanoma cases are familial. Variants in CDKN2A account for up to 40% of melanoma-prone families, with an additional ~10% explained by other genes. Many CDKN2A mutation-negative families show linkage to chromosome-band 9p21, which harbors CDKN2A, suggesting non-coding variants may contribute to familial risk. Here, whole-genome sequencing revealed a novel 100 kb deletion mapping to 9p21 in a gene-desert region, 205 kb from CDKN2A, cosegregating in a four-case family from Genoa, Italy. The deletion overlaps melanocyte enhancers that interact with the pro

Pharmacoepigenetic Biomarkers in Inflammatory Bowel Diseases: A Narrative Review.

Inflammatory bowel disease (IBD), encompassing Crohn's disease (CD) and ulcerative colitis (UC), is a chronic, autoimmune disorder characterized by inflammation along the gastrointestinal tract. Global prevalence of the disease is increasing and patients often experience delays in diagnosis accessing effective therapy, highlighting an urgent need to develop a predictive biomarker for therapeutic response to reduce healthcare costs and disease burdens. Despite the advances to identifying genetic biomarkers for prediction of disease remission in IBD, patient responses vary widely, suggesting tha

Long-term globus pallidus internus deep brain stimulation in a young patient with spinocerebellar ataxia type 3 initially presenting with levodopa-responsive parkinsonism: a 6-year follow-up case report and literature review.

Spinocerebellar ataxia type 3 (SCA3) is an inherited neurodegenerative disorder. Some of its clinical features resemble those of primary Parkinson's disease (PD), which can easily lead to misdiagnosis. There is currently no disease-modifying therapy available for SCA3, treatment is mainly symptomatic. Herein, we report a case of a young female patient with SCA3 who presented with Parkinsonian as the main manifestation and underwent globus pallidus internus (GPi) deep brain stimulation (DBS). This is a 36-year-old female patient. Her first symptoms occurred at the age of 28 in 2009, manifesting

The Landscape of Structural Variation in Aye-Ayes (Daubentonia madagascariensis).

Aye-ayes (Daubentonia madagascariensis) are one of the 25 most critically endangered primate species in the world. Endemic to Madagascar, their small and highly fragmented populations make them particularly vulnerable to both genetic disease and anthropogenic environmental changes. Over the past decade, conservation genomic efforts have largely focused on inferring and monitoring population structure based on single nucleotide variants to identify and protect critical areas of genetic diversity. However, the recent release of a highly contiguous genome assembly allows, for the first time, for

WT1 pathogenic variant in azoospermic infertile men with an isolated undescended testis.

OBJECTIVE: An undescended testis (UDT) is a testicle that has not moved into the scrotum before birth. UDTs are linked to reduced fertility, primarily due to compromised semen quality and potential dysfunction in Sertoli and Leydig cells. Additionally, the Wilms tumor 1 (WT1) gene is crucial for spermatogenesis, as it regulates the polarity of Sertoli cells and the steroidogenesis in Leydig cells. Our study aimed to identify novel UDT-causing WT1 variants within a cohort of 60 unrelated men with infertile hypergonadotropic hypogonadism. METHODS: In this case-control study, the coding regions

The exposomal imprint on rosacea: More than skin deep.

Rosacea is a chronic, inflammatory dermatosis driven by a complex interplay of genetic, environmental and lifestyle factors, collectively known as the exposome. This review explores how intrinsic contributors such as genetic susceptibility, immune dysregulation, microbiome alterations, hormonal influences and psychosocial stress intersect with extrinsic triggers like ultraviolet radiation (UVR), air pollution, dietary factors, and climate variability to shape rosacea pathogenesis. Recent advances in single-cell transcriptomics have identified fibroblasts as key components of inflammatory and v

Unravelling CYP4V2: Clinical features, genetic insights, pathogenic mechanisms and therapeutic strategies in Bietti crystalline corneoretinal dystrophy.

Inherited retinal dystrophies (IRDs) comprise a spectrum of disease phenotypes with genetic heterogeneity and clinical phenotypic diversity. Bietti crystalline corneoretinal dystrophy (BCD) represents a distinct IRD subtype characterized by crystalline deposits in the retina. Although rare in Western populations, BCD ranks among the most prevalent IRDs in East Asia, affecting an estimated 124,000 to 377,000 individuals worldwide. As a severe type of IRD, BCD demonstrates accelerated progression and currently lacks effective treatment. The BCD disease is caused by a biallelic mutation in the CY
