Chitosan/indole-3-acetic acid/silver nanoparticle composite films: Preparation, characterization and efficacy in inhibiting postharvest potato dry rot.

This study aimed to prepare and characterize a chitosan (CS)-based composite film incorporating indole-3-acetic acid (IAA) and silver nanoparticles (AgNPs) via a solution casting method, and to evaluate its efficacy in controlling Fusarium oxysporum-induced dry rot in postharvest potatoes. Microstructural analysis revealed uniformly dispersed AgNPs and a compact network, with FTIR and XRD confirming that hydrogen bonding dominates CS and IAA interactions while coordination interactions govern CS and AgNP associations. This compact network significantly enhanced tensile strength (40.09% increas…

The study of intestinal toxicity induced by gossypol acetic acid and the mechanism using zebrafish model and network toxicology.

Gossypol acetic acid (GAA), a natural product obtained from the cottonseed, has various bioactivities, including antiviral, anti-tumor and anti-inflammatory properties, among others. However, the potential intestinal toxicity of GAA is poorly studied. Here, the intestinal toxicity and the underlying mechanism were investigated in vivo using zebrafish and network toxicology. After exposure from 3 days post fertilization (dpf) to 6 dpf, GAA could notably affect the zebrafish gut barrier by reducing the microvilli and goblet cells, and subsequently affected the intestinal function, including decr…

Ash-forming elements of poplar and hybrid pennisetum negatively affect selective delignification ability of hydrogen peroxide-acetic acid pretreatment.

Hydrogen peroxide-acetic acid (HPAA) is widely used in bio-refinery by delignification but exhibits different delignification ability and xylan loss toward different biomass materials. This work discovered that the synergistic cyclic catalysis of ash-forming elements (Fe, Cu, and Mn) in biomass was responsible for the differences in lignin and xylan removal in HPAA pretreatment. After the addition of 10 mg/mL ash in HPAA at 60 °C for 2 h, the concentrations of H2O2 and peracetic acid were reduced to 50.2 and 12.8 mg/mL, respectively. The corresponding reduction in the redox potential of HPAA f…

Engineered indoleamine 2,3-dioxygenase variant: Oxidative activation of prodrug indole-3-acetic acid against cancer cells for targeted therapy.

Indoleamine 2,3-dioxygenase (IDO) is a rate-limiting enzyme involved in the kynurenine pathway of tryptophan metabolism and has been implicated in immune regulation and tumor immune evasion. Beyond its role in tryptophan metabolism, we found that IDO can catalyze the oxidative activation of indole-3-acetic acid (IAA). This reaction generates reactive oxidative intermediates and triggers potent cytotoxicity in cancer cells. Leveraging human-derived enzymes as "suicide effectors" confers a critical advantage by mitigating the risk of immunogenic rejection. However, the intrinsic kynurenine-pathw…

Opposite regulatory effects of Blautia massiliensis and Blautia faecis on cognitive function, microglia and metabolite acetic acid in mice.

Cognitive impairment is a significant health problem worldwide, closely associated with the status of gut microbiota. Our recent research has revealed the Blautia faecis and Blautia massiliensis exhibit opposing associations with cognitive function in children with Down syndrome clinically characterized by cognitive dysfunction. However, the role and mechanisms of Blautia faecis and Blautia massiliensis in cognitive function remain unknown. Therefore, we gavaged C57BL/6 male mice with commercially available Blautia faecis and Blautia massiliensis for 3 weeks and assessed cognitive function usi

Effects of paraprobiotic Lactobacillus acidophilus LA-5 on PANoptosis, oxidative stress and inflammation in acetic acid-induced ulcerative colitis.

BACKGROUND: Ulcerative colitis (UC) is a relapsing inflammatory bowel disease characterised by oxidative stress, inflammation and regulated cell death of the colonic mucosa. Because live probiotics may cause bacteraemia and sepsis in immunocompromised hosts, non-viable (paraprobiotic) alternatives have attracted interest. METHODS: This study evaluated the antioxidant, anti-inflammatory and PANoptosis-regulating effects of a heat-inactivated paraprobiotic derived from Lactobacillus acidophilus LA-5 in acetic acid (AA)-induced experimental UC, compared to the live probiotic. Forty female Balb/c…

[Medium optimization and fermentation regulation strategies for enhancing CO2 conversion to acetic acid coupled with H2 by Clostridium ljungdahlii].

Clostridium ljungdahlii can reduce CO2 to acetic acid via the Wood-Ljungdahl pathway using H2 as an energy source and CO2 as a carbon source, and it is a key chassis for realizing resource utilization of CO2. Acetic acid, as a bulk basic organic chemical raw material, has a large market demand and wide application scenarios. Moreover, the syngas fermentation route is green and low-carbon, with mild reaction conditions, making it an ideal direction for replacing traditional petroleum-based processes. However, during the fermentation process, it encountered problems such as slow growth, low biom…

Enhancing HAP3 mediated xylose metabolism and acetic acid metabolism promotes lipid production from lignocellulosic hydrolysate fermented by Candida glycerinogenes.

Enhanced Candida glycerinogenes xylose metabolism as well as acetic acid metabolism for lipid production from undetoxified lignocellulosic hydrolysate. The C. glycerinogenes after overexpression of HAP3 increased the xylose assimilation capacity and the xylitol production decreased by 66.7%. HAP3 could bind to the upstream activation sequence of XYL2 promoter and promote the expression of XYL2. The transcription factor HAP3 enhanced xylose metabolism and increased lipid production in undetoxified lignocellulosic hydrolysate. By enhancing the metabolism of acetic acid, the ability of acetic aci…

High-energy irradiation of matrix isolated acetic acid yields a ·CH3⋯CO2 complex: A spectroscopic and ab initio study.

The mechanism of the chemical transformations of isolated small organic molecules induced by high-energy radiation is of basic interest for astrophysics and astrochemistry. In this work, we first applied a combination of electron paramagnetic resonance (EPR) and Fourier-transform infrared (FTIR) spectroscopy to identify the products of the radiation-induced transformations of isolated CH3COOH and CD3COOH molecules. As revealed by EPR, ·CH3 (or ·CD3) is the principal primary radical generated from acetic acid in solid argon and xenon, while the FTIR results suggest that this radical is trapped …

Gut microbiota-derived indole-3-Acetic Acid attenuates skeletal fluorosis via AHR-mediated suppression of Wnt/β-Catenin signaling.

Chronic fluoride exposure causes skeletal fluorosis (SF), a debilitating bone disease, but the lack of a comprehensive understanding of its pathogenesis has hindered the development of effective therapies. Although fluoride is known to stimulate osteoblasts, the role of gut-bone crosstalk in SF remains uninvestigated. In this study, using a rat SF model and in vitro osteoblast studies, we found that fluoride disrupted gut microbial tryptophan metabolism, resulting in decreased serum levels of indole-3-acetic acid (3-IAA) in both SF model rats and human SF patients. This deficiency in 3-IAA imp…

Effects of Acetic Acid and Morphine in Shore Crabs, Carcinus maenas: Implications for the Possibility of Pain in Decapods.

Noxious chemicals, coupled with morphine treatment, are often used in studies on pain in vertebrates. Here we show that injection of morphine caused several behavioural changes in the crab, Carcinus maenas, including reduced pressing against the sides of the enclosure and more rubbing and picking at the mouth parts and, at least for a short time, more defensive displays. Subsequent injection of acetic acid into one rear leg caused rubbing of the injected leg and the injected leg was held vertically off the ground. These activities directed at or involving the specific leg are consistent with p…
