Seroprevalence of Toxoplasma gondii in ruminants by using latex agglutination test (LAT) and enzyme-linked immunosorbent assay (ELISA) in Assiut governorate.

Toxoplasmosis is an important zoonotic parasitic disease world-wide. However, data on Toxoplasma gondii infection in ruminants are scarce in Assiut governorate, Egypt. Thus we conducted a survey on the seroprevalence of T. gondii infection by using latex agglutination test (LAT) and enzyme-linked immunosorbent assay (ELISA). A total of 274 sera were tested (56 imported camels, 56 cattle, 55 buffaloes, 50 sheep and 57 goats) in the period from November 2014 - February 2016. The overall seroprevalence in ruminants was (35.8%) by LAT and (83.6%) by ELISA. The highest rate of T. gondii infection b

Performance of an indigenous β-mercaptoethanol-modified antigen in comparison with a commercial reference in direct agglutination test for detection of canine visceral leishmaniasis.

We compared the performance of a locally produced β-mercaptoethanol-modified promastigote antigen (β-ME-Ag) of an indigenous Leishmania infantum strain against that of a trypsinized Leishmania donovani reference (REF-Ag) in the direct agglutination test (DAT) for detection of canine visceral leishmaniasis (CVL). One hundred and fifty-one serum samples collected from dogs belonging to four groups with different conditions were included. At a DAT titre of 1 : 320, statistically determined as optimal cut-off value for β-ME-Ag, and 1 : 160 for REF-Ag, a sensitivity and a specificity of 100 % were

Development of a latex agglutination test for detecting antibodies against avian influenza virus based on matrix 1 protein expressed in vitro.

The matrix 1 (M1) gene, present in all subtypes of avian influenza virus (AIV), was cloned and expressed in Escherichia coli. Reactivity of the expressed protein was confirmed by western blot. Subsequently, the M1 gene expression product was purified and used as the antigen to develop a latex agglutination test (LAT) for detecting antibodies against these conventional subtypes of AIV including AIV H3, H5, H7, and H9 from chicken sera. The LAT is specific for AIV, and no cross-reaction was shown with chicken antisera against other avian viruses. Compared with the hemagglutination inhibition tes

Practical therapeutic application of the oxoid PBP2' latex agglutination test for the rapid identification of methicillin-resistant Staphylococcus aureus in blood cultures.

The Oxoid PBP2' latex agglutination test (OLA; Oxoid, Basingstoke, England) was evaluated in a controlled prospective study examining Staphylococcus aureus from 25 positive blood cultures. Subcultures of positive blood cultures with coagulase-positive, gram-positive cocci in clusters were batched, and the OLA was performed at the end of the working day, once growth was seen on the plate. Results were sent to the infectious disease pharmacist for therapy evaluation, and the 24-hour minimum inhibitory concentration (MIC) was confirmed the next day. Blood culture OLA results correlated 100% with

False-positive latex agglutination test for Neisseria meningitidis groups A and Y caused by povidone-iodine antiseptic contamination of cerebrospinal fluid.

The cerebrospinal fluid of a patient yielded a positive latex agglutination test for Neisseria meningitidis groups A and Y. The latex agglutination results were not consistent with clinical and other laboratory findings. An investigation determined that the positive agglutination test was caused by contamination of the cerebrospinal fluid with povidone-iodine during the lumbar puncture.

Development and diagnostic evaluation of a reverse passive latex agglutination test for rapid field detection of lumpy skin disease virus antigen.

Lumpy skin disease (LSD) is a vector-borne viral disease of cattle that causes significant production losses. Caused by Lumpy skin disease virus (LSDV), it has recently emerged as a Transboundary Animal Disease (TAD), creating a need for rapid and sensitive diagnostic assays to facilitate disease surveillance and control programs. In this study, a reverse passive latex agglutination test (RPLAT) was developed and validated for the rapid detection of lumpy skin disease virus (LSDV) antigen. Polystyrene Latex beads were coated with hyperimmune serum raised against LSDV in Guinea pig, and assay c

Detection of anti-leptospiral antibodies using recombinant ErpY-like lipoprotein based latex agglutination test for serodiagnosis of animal leptospirosis.

Precise and timely diagnosis is essential to prevent severe outcomes of leptospirosis in humans and animals. Existing diagnostic methods face challenges and limitations, underscoring the need for novel, field-applicable screening, and diagnostic tests/assays. This study evaluates the diagnostic utility of a recombinant ErpY-like lipoprotein (rErpY-LIC11966) in a latex agglutination test (LAT) for diagnosis of animal leptospirosis. The ErpY gene sequence from Leptospira interrogans serovar Pomona, excluding the signal peptide, was amplified, cloned into the pETite vector, and expressed in Esche

High specificity of the Salmonella Pullorum/Gallinarum rapid plate agglutination test despite vaccinations against Salmonella Enteritidis and Salmonella Typhimurium.

In Europe, monitoring of breeding stock for Salmonella Pullorum (SP) or Salmonella Gallinarum (SG) infections is compulsory at the point of lay. Vaccinations against Salmonella Enteritidis (SE) and Salmonella Typhimurium (ST) are increasingly administered in Europe. These vaccines might induce cross-reactions in the rapid plate agglutination (RPA) SP/SG test due to shared O-antigens, possibly resulting in a lower test specificity. The extent to which the specificity of SP/SG serological tests is influenced by SE and/or ST vaccinations in the field has not been reported. In this paper, we repor

A comparative study of Toxoplasma gondii seroprevalence in mink using a modified agglutination test, a Western blot, and enzyme-linked immunosorbent assays.

Toxoplasma gondii can infect almost all warm-blooded animals, and many serological methods have been developed to detect T. gondii infection in a variety of animal species. In the present study, the seroprevalence of T. gondii infection in farmed mink in northeast China was determined using the modified agglutination test (MAT), a Western blot (WB), and 3 enzyme-linked immunosorbent assays (ELISAs) with protein A/G conjugate, using either of 2 recombinant dense granule antigens, GRA1 and GRA7, or Toxoplasma soluble antigens (TSA). There was no significant difference between the detection resul

A new prepatory method of thermically inactivated Leptospira Patoc antigen for rapid slide agglutination used as serosurvey test for human leptospiroses.

830 sera from leptospirosis-suspect patients and 133 sera from investigations were analysed by the new test--rapid slide agglutination with Patoc antigen, inactivated at 60 degrees C for 15 minutes, in parallel with two standard tests--CFT with merthiolated Patoc antigen and MAR with 17-23 live pathogenic antigens. The new serotest proved efficient for selecting the cases of disease or leptospira infection, acute or recent form, not older than 2 years, both by its sensitivity and specificity and also by its rapid and economical character. In the present paper, the technology of preparation and

An efficient and convenient Fiber -2- based latex agglutination test for the detection of antibodies against fowl adenovirus serotype 4 in clinical samples.

To detect the antibody against fowl adenovirus serotype 4 (FAdV-4) in clinical practice, the latex agglutination test (LAT) was developed by using the Fiber-2 protein of FAdV-4 as an antigen bound to sensitized latex microspheres. The concentration, time, and temperature of sensitization latex microspheres by the Fiber-2 protein were studied and optimized; the specificity, sensitivity, and repeatability of LAT were tested; and the method developed in the study was applied. The results showed that the optimum sensitization concentration of Fiber-2 protein was 0.8 mg/mL, the time was 120 min, an

Comparison of a commercial ELISA and indirect hemagglutination assay with the modified agglutination test for detection of Toxoplasma gondii antibodies in giant panda (Ailuropoda melanoleuca).

Toxoplasma gondii is a worldwide-distributed zoonotic protozoan parasite which causes toxoplasmosis and has a significant effect on public health. In the giant panda (Ailuropoda melanoleuca), toxoplasmosis can cause asymptomatic infections, reproductive disorder and even death, which poses a serious threat to the conservation of this rare protected species. Therefore, serological investigation of T. gondii is essential to understanding its risk to giant pandas, however, there are no specific testing kits for giant pandas. Previous research has used MAT as the reference method for screening T.
