Indoprofen, a new-steroidal anti-inflammatory agent, binds to human serum albumin in an endothermic reaction at low degrees of saturation and with an exothermic reaction at higher drug to protein ratios. Although indoprofen appears to bind to the same primary site as diazepam, dialysis studies show an increased binding of both drugs in the presence of the other. This novel observation is probably due to the effect of the drugs on the N leads to B conformational change of human serum albumin.

Albumin metabolism was studied with the aid of 131 I- albumin in human blood serum in 5 healthy individuals and 40 patients with chronic cardiac insufficiency. It is shown that in patients with different degrees of circulatory insufficiency the loss of albumin with feces into gastro-intestinal lumen does not exceed the loss in the healthy individuals. The cardiac insufficiency is characterized by intracellular metabolic disorders of albumin of the hypercatabolic type.

Hetastarch, 6% hydroxyethyl starch solution, is an artificial colloid proposed for use as a volume expander. There is concern that hetastarch, like dextran, may adversely affect coagulation. Hyperoncotic albumin proved ineffective as a plasma volume expander in the resuscitation of four acutely burned children. Hypovolemia and pulmonary edema were apparently intensified by an obligatory fluid shift following tissue extravasation of protein. We conclude that, when used, hyperoncotic albumin should be suspended in a sufficient volume of saline to produce a solution isotonic and iso-oncotic relative to plasma.

The in vitro binding of warfarin by human serum albumin was studied at various temperatures and at pH 7.4 by a frontal gel filtration technique. The results can be best described in terms of a two class-of-binding site model, in which the numbers of primary and secondary sites are constrained to the average values for all experiments (n1 = 1.38 and n2 = 3.73). Analysis of the temperature dependence of the binding yielded the following thermodynamic parameters: deltaH1 =-2.55 kcal/mole, deltaS1=16.1 eu, and deltaF1=-7.34 kcal/mole for the primary binding and deltaH2=-5.08 kcal/mole, deltaS2=-1.

The rates of elimination of iodinated human serum albumin (ALBUMIN-125I), polyvinylpyrrolidone (PVP-125I); and tritiated dextran (DEXTRAN - METHOXY-3H) (mol masses of 69 000 - 72 000, 30 000 - 40 000 and 60 000 - 90 000 respectively) from the circulation of sheep were studied; albumin and PVP initially disappeared from the circulation rapidly having half-life times (t 1/2) of 10,5 +/- 3,7 and 43 +/- 45 hours respectively. This phase is regarded as being due to equilibration within the initial volume of distribution, the rate being determined primarily by the relative mol. mass of the molecule.

We report a fluorometric technique for determination of albumin-titratable bilirubin in the jaundiced neonate. Although bilirubin alone has very little native fluorescence, considerable emission is observed in the presence of albumin under acid conditions. Analysis of the plasma sample alone and in the presence of excess human serum albumin solution appears to reflect the bilirubin tightly bound to albumin and the total serum bilirubin, respectively. The antibody content of three reaginic sera from patients sensitive to highly purified horse serum albumin have been investigated by measuring the capacity to bind 131I-labelled albumin. No correlation was found between skin sensitizing activity and the content of either heat stable or heat labile antibody, but there was a correlation with the ratio of the concentration in the serum of the two kinds of antibody.
