Effect of the support alkyl chain nature in the functional properties of the immobilized lipases.

Supports coated with amino-hexyl and amino octyl have been prepared from glyoxyl agarose beads and compared in their performance with octyl-agarose to immobilize lipases A and B from Candida antarctica (CALA and CALB). Immobilization courses were similar using all supports, but enzyme release was more difficult using the amino-alkyl supports suggesting a mixed interfacial activation/ionic exchange immobilization. The enzyme activity and specificity (using p-nitrophenyl propionate, triacetin and both isomers of methyl mandelate) greatly depended on the support. In many instances the enzymes imm

A General Metallaphotoredox Platform for N-Alkylated Sulfoximines as Bioisosteric Building Blocks.

The precise modulation of physicochemical and ADME properties is critical in drug discovery. We report a unified, mild, and broadly applicable metallaphotoredox-enabled protocol for the N-alkylation of sulfoximines, a versatile S(VI) motif gaining prominence as a bioisostere. This method overcomes limitations of previous N-substitution approaches by accommodating diverse alkyl sources, including alcohols, alkyl bromides, and carboxylic acids, under visible-light irradiation. The strategy features high functional group tolerance and offers efficient access to both primary and secondary N-alkyl …

Enantioselective Alkyl-Acyl Radical Cross-Coupling Enabled by Metallaphotoredox Catalysis.

Radical-radical cross-coupling (RCC) offers a promising approach for carbon-carbon bond formation in organic synthesis, particularly for creating complex, three-dimensional molecules. However, achieving both cross- and enantioselectivity in RCC reactions has remained a significant challenge. Here, we report a novel metallaphotoredox platform that enables highly enantioselective decarboxylative coupling of carboxylic acid derivatives with aldehydes. Our strategy leverages independent control over radical generation and subsequent enantioselective bond formation through fine-tuning of a common p

Superbasic Anionic Calcium Alkyl Complexes: Templated Twofold Deprotonation of Benzene.

Anionic calcium alkyl complexes have been synthesised through the reduction of ethylene by an anionic calcium hydride. The alkyl complexes are demonstrated to be superbasic, rapidly deprotonating ethers below room temperature, leading to C─O bond cleavage. In reactions with benzene, selective 1,4-metalation of benzene is observed, forming an inverse-crown areneide complex. This transformation, previously inaccessible to calcium, proceeds via a stepwise mechanism, which can also be facilitated by co-complexation with organometallic reagents such as tert-butyllithium and phenylpotassium.

Ligand-Promoted Copper-Catalyzed Radical Relay for Alkyl-Alkynylation of Alkenes.

The alkynylation of alkenes provides an efficient platform for synthesizing compounds containing C≡C bonds. However, the intermolecular alkynylation of alkenes has been largely restricted to precious metal palladium-catalyzed systems, limiting reaction versatility. Herein, we report a copper-catalyzed radical relay strategy for intermolecular alkyl-alkynylation of alkenes and construction of challenging C(sp3)-C(sp3) bonds and C(sp3)-C(sp) bonds in one pot. This method utilizes alkene-tethered α-carbonyl bromides as the alkyl source, which undergo single-electron transfer to generate alkyl rad

Reversible cyclodimerization of cyclic (alkyl)(amino)carbene-carbon disulfide adduct.

The cyclic (alkyl)(amino)carbene-carbon disulfide adduct undergoes thermally induced cyclodimerization to a 1,2,4,5-tetrathiane and quantitatively dissociates back to the monomer under light irradiation. This stimulus-driven, reagent-free, and reversible bond formation and cleavage contrasts with the conventional dynamic behavior of S-S bonds.

Trifluoromethylation of alkyl electrophiles with 11C- or 18F-labeled fluoroform for PET applications.

Continued development of positron emission tomography (PET) tracers is essential for advancing molecular imaging in biomedical research and clinical diagnostics. A long-standing limitation in radiochemistry for PET imaging has been the lack of general methods for radiolabeling trifluoromethyl (CF3) groups at C(sp3) sites, despite their growing prevalence in bioactive molecules and radiopharmaceuticals. Here, we present a general approach for late-stage installation of either a [18F]CF3 or [11C]CF3 group at a C(sp3) site. This method leverages unusual copper-mediated radiotrifluoromethylation o

Intracellular Generation of Alkyl Radicals Enabled by a Self-Catalytic ATRP Nanoinitiator.

Oxygen-independent alkyl radicals (R•) have demonstrated great promise in combating tumor hypoxia. Currently, Azo compounds have been the primary source of R•, suffering from external stimuli and decomposition during circulation. Herein, we developed a self-catalytic ATRP nanoinitiator that could generate R• via glutathione (GSH) reduction and thus selectively induce apoptosis of tumor cells. Specifically, a conjugation of laccase (possessing a copper(II) complex) and polymeric alkyl bromide, poly(iBBr), was fabricated to yield an ATRP nanoinitiator (Lac-P(iBBr)). After internalization by cell

Electrochemical Multicomponent Synthesis of Alkyl Alkenesulfonates using Styrenes, SO2 and Alcohols.

A novel electrochemical approach to access alkyl alkenesulfonates via a multicomponent reaction was developed. The metal-free method features easy-to-use SO2 stock solution forming monoalkylsulfites from alcohols with an auxiliary base in-situ. These intermediates serve a dual role as starting materials and as supporting electrolyte enabling conductivity. Anodic oxidation of the substrate styrene, radical addition of these monoalkylsulfites and consecutive second oxidation and deprotonation preserve the double bond and form alkyl β-styrenesulfonates in a highly regio- and stereoselective fashi

The synergy between alkylating agents and ERCC1-XPF inhibitors is p53 dependent.

BACKGROUND: DNA repair plays a major role in maintaining genomic stability, thus limiting the transformation of normal cells into cancer cells. However, in cancer patients treated with DNA-targeting drugs, DNA repair can decrease efficacy by removing the damage generated by such molecules that is needed to induce pharmacological activity. Inhibiting DNA repair thus represents an interesting approach to potentiating the activity of chemotherapy in this setting. OBJECTIVES: Here, we continue the characterization of an inhibitor of the interaction between Excision Repair Cross-Complementing Rrod

Reactivity of Yellow Arsenic towards Cyclic (Alkyl)(Amino) Carbenes (CAACs).

Different cyclic (alkyl)(amino)carbenes (CAACs) were reacted with yellow arsenic. Several products [(CAAC-n)2 (μ,η1:1 -As2 )] (n=1 (1), 4 (2)), [(CAAC-2)3 (μ3 ,η1:1:1 -As4 )] (3) and [(CAAC-3)4 (μ4 ,η1:1:1:1 - As8 )] (6) were isolated due to the differing steric properties of CAAC-1-4. The products contain As2 , As4 or As8 units and represent the first examples of CAACs-substituted products of yellow arsenic. The reactivity of As4 was compared with the reactivities of P4 and the interpnictogen compound AsP3 , which led to a series of phosphorus-containing derivatives such as ([(CAAC-3)3 (μ3 ,η

Nickel catalyzed three-component sulfonylation of non-activated alkyl chlorides.

We describe a nickel-catalyzed three-component sulfonylation of readily available non-activated alkyl chlorides. A wide range of alkyl aryl sulfones can be synthesized from alkyl chlorides, aryl boronic acids, and potassium metabisulfite, a cheap, efficient, and commercially available SO2 source under simple and easy-to-handle reaction conditions. High selectivity can be achieved with a slight excess of phenylboronic acid and a sulfur dioxide source.

Nickel-catalyzed cyanation reaction of aryl/alkenyl halides with alkyl isocyanides.

We herein describe a nickel-catalyzed cyanation reaction of aryl/alkenyl halides with alkyl isocyanides. Both aryl/alkenyl iodines and bromides were found to be competent electrophiles that reacted with alkyl isocyanides, affording nitrile compounds in moderate to good yields. A range of functional groups including halogens as well as hydroxyl, formyl, and acetamino groups were fairly compatible with the nickel catalysis. This protocol featured broad functional group tolerance, simple reaction conditions, and gram-scale synthesis.

A Selective Alkylating Agent for CTG Repeats in Myotonic Dystrophy Type 1.

Disease intervention at the DNA level generally has been avoided because of off-target effects. Recent advances in genome editing technologies using CRISPR-Cas9 have opened a new era in DNA-targeted therapeutic approaches. However, delivery of such systems remains a major challenge. Here, we report a selective DNA-modifying small molecule that targets a disease-specific structure and mismatches involved in myotonic dystrophy type 1 (DM1). This ligand alkylates T-T mismatch-containing hairpins formed in the expanded CTG repeats (d(CTG)exp) in DM1. Ligand alkylation of d(CTG)exp inhibits the tra…

Catalytic Asymmetric Homologation of Ketones with α-Alkyl α-Diazo Esters.

The homologation of ketones with diazo compounds is a useful strategy to synthesize one-carbon chain-extended acyclic ketones or ring-expanded cyclic ketones. However, the asymmetric homologation of acyclic ketones with α-diazo esters remains a challenge due to the lower reactivity and complicated selectivity. Herein, we report the enantioselective catalytic homologation of acetophenone and related derivatives with α-alkyl α-diazo esters utilizing a chiral scandium(III) N,N'-dioxide as the Lewis acid catalyst. This reaction supplies a highly chemo-, regio-, and enantioselective pathway for the

Monitoring of linear alkyl benzenes (LABs) in riverine and estuarine sediments in Malaysia.

The aim of this a pioneering research is to investigate linear alkylbenzenes (LABs) as biomarkers of sewage pollution in sediments collected from four rivers and estuaries of the south and east of Peninsular Malaysia. The sediment samples went through soxhlet extraction, two-step column chromatography purification, fractionation and gas chromatography-mass spectrometry (GC-MS) analysis. Principal component analysis (PCA) with multivariate linear regression (MLR) was used as well for source apportionment of LABs. The results of this study showed that total LAB concentration was 36-1196 ng g-1dw

Direct α-Tertiary Alkylations of Ketones in a Combined Copper-Organocatalyst System.

Herein, we report an efficient method for the tertiary alkylation of a ketone by using an α-bromocarbonyl compound as the tertiary alkyl source in a combined Cu-organocatalyst system. This dual catalyst system enables the addition of a tertiary alkyl radical to an enamine. Mechanistic studies revealed that the catalytically generated enamine is a key intermediate in the catalytic cycle. The developed method can be used to synthesize substituted 1,4-dicarbonyl compounds containing quaternary carbons bearing various alkyl chains.

Human METTL7B is an alkyl thiol methyltransferase that metabolizes hydrogen sulfide and captopril.

Methylation of alkyl thiols is a biotransformation pathway designed to reduce thiol reactivity and potential toxicity, yet the gene and protein responsible for human alkyl thiol methyltransferase (TMT) activity remain unknown. Here we demonstrate with a range of experimental approaches using cell lines, in vitro systems, and recombinantly expressed enzyme, that human methyltransferase-like protein 7B (METTL7B) catalyzes the transfer of a methyl group from S-adenosyl-L-methionine (AdoMet) to hydrogen sulfide (H2S) and other exogenous thiol small molecules. METTL7B gene modulation experiments, i

Nickel-Catalyzed Reductive Thiolation of Unactivated Alkyl Bromides and Arenesulfonyl Cyanides.

The cross-electrophile coupling between unactivated alkyl bromides with arenesulfonyl cyanides catalyzed by Ni(acac)2 under reductive conditions to form unsymmetrical sulfides is developed. This approach for sulfide synthesis is practical, relies on available, unfunctionalized materials such as alkyl (pseudo)halides, and is scalable. This catalytic strategy provides a complementary method for the preparation of unsymmetrical alkyl-aryl sulfides under mild conditions with good functional group tolerance.

Catalyst-Controlled 1,2- and 1,1-Arylboration of α-Alkyl Alkenyl Arenes.

Two methods are reported for the 1,2- and 1,1-arylboration of α-methyl vinyl arenes. In the case of 1,2-arylboration, the formation of a quaternary center occurred through a rare cross-coupling reaction of a tertiary organometallic complex. 1,1-Arylboration was enabled by catalyst optimization and occurred through a β-hydride elimination/reinsertion cascade. Enantioselective variants of both processes are presented as well as mechanistic investigations.
