Ultrasensitive Protease Sensors Using Selective Affinity Binding, Selective Proteolytic Reaction, and Proximity-Dependent Electrochemical Reaction.

The development of a fast and ultrasensitive protease detection method is a challenging task. This paper reports ultrasensitive protease sensors exploiting (i) selective affinity binding, (ii) selective proteolytic reaction, and (iii) proximity-dependent electrochemical reaction. The selective affinity binding to capture IgG increases the concentration of the target protease (trypsin as a model protease) near the electrode, and the selective proteolytic reaction by trypsin increases the concentration of the redox-active species near the electrode. The electrochemical reaction, which is more se

Therapeutic efficacy of sodium nitrite and 4-dimethylaminophenol or hydroxylamine co-administration against cyanide poisoning in rats.

1. The therapeutic efficacy of combined treatment of sodium nitrite (NaNO2) and 4-dimethylaminophenol (DMAP) or hydroxylamine (H2NOH) was investigated in potassium cyanide (KCN) intoxication in male rats. 2. Therapy with NaNO2 (0.27 mmol kg-1) + DMAP (0.09 mmol kg-1) or NaNO2 + H2NOH (0.09 mmol kg-1, produced a protection index (ratio of LD50 of KCN in rats receiving therapy to an LD50 of KCN in rats given only 0.9% saline) of 2.5 and 2.0 respectively. 3. Both the regimens exhibited a beneficial effect in terms of improving the survival time and postural defects in rats exposed to 2 LD50 KCN.

Fetal Transplacental Therapy to Mitigate Consequences of Cystic Fibrosis.

Highly effective modulator therapy (HEMT) has been life-changing for many living with cystic fibrosis (CF), a genetic disease affecting all races and ethnicities. With diagnoses now occurring in utero for many patients, prenatal treatment of the pregnant person with HEMT to mitigate the effects of CF on the growing fetus is a hot topic of discussion. This review summarizes recent literature for fetal transplacental therapy with HEMT for providers caring for parent/fetal dyads and offers expert opinion regarding monitoring and future areas of research.

When rash challenges treatment: reintroducing cystic fibrosis modulator therapy safely.

Various adverse reactions to elexacaftor/tezacaftor/ivacaftor (ETI) therapy have been documented in the literature. Here, we present the first reported case in Croatia of severe rash following ETI therapy in a 19-year-old female cystic fibrosis (CF) patient. There were two attempts to introduce ETI therapy, spaced two months apart, both of which resulted in diffuse maculopapular rash, in the second attempt, the rash appeared on the same day as re-initiation. Given the clinical benefits of ETI treatment, andinspired by several case reports on drug desensitization, in July 2023, we started the p

Lumacaftor inhibits channel activity of rescued F508del cystic fibrosis transmembrane conductance regulator.

Lumacaftor, the corrector of Orkambi, enhances the processing of F508del cystic fibrosis transmembrane conductance regulator (CFTR), but its impact on the channel activity of rescued F508del CFTR (rF508del) is unclear. Using an electrode-based, real-time iodide efflux assay performed at room temperature, acute exposure to lumacaftor was shown to increase the processing of F508del CFTR without a proportional increase in channel activity in a CFBE41o-cell line stably expressing F508del CFTR (CFBE-DF). A similar effect was not observed on wild-type CFTR in a HEK293 cell line. At 37°C, rF508del ch

Advanced Cystic Fibrosis Lung Disease and Lung Transplantation in the Era of Cystic Fibrosis Transmembrane Conductance Regulator Modulators.

Cystic fibrosis transmembrane conductance regulator (CFTR) modulators have changed the clinical landscape of cystic fibrosis (CF) by improving clinically significant outcome measures and quality of life of people with CF (pwCF). There are now long-term data showing improved 5-year survival with the use of ivacaftor, and the field continues to evolve at a rapid pace with the continued development of highly effective CFTR modulators. While the randomized controlled trials of CFTR modulators excluded patients with severe lung disease (forced expiratory volume in 1 second <40% predicted), observat

Effect of elexacaftor/tezacaftor/ivacaftor on airway and systemic inflammation in cystic fibrosis.

Treatment with elexacaftor/tezacaftor/ivacaftor (ETI) has been shown to improve lung function in people with cystic fibrosis (PWCF). However, its biological effects remain incompletely understood. Here we describe alterations in pulmonary and systemic inflammation in PWCF following initiation of ETI. To address this, we collected spontaneously expectorated sputum and matching plasma from PWCF (n=30) immediately prior to ETI therapy, then again at 3 and 12 months. Within 3 months, PWCF demonstrated reduced activity of neutrophil elastase, proteinase three and cathepsin G, and decreased concentr

Demonstration of putative Ca-binding domains in dentin matrix of rat incisors after daily injections of 1-hydroxyethylidene-1,1-bisphosphonate (HEBP).

In order to clarify the initial process of dentin mineralization, the inhibitory effect of 1-hydroxyethylidene-1,1-bisphosphonate (HEBP) on dentin mineralization was investigated. Rats (100 g) were subcutaneously injected with HEBP (8 mg P/kg) for 7 or 14 d, and the incisors were processed for Ca histochemistry and/or electron microscopy. HEBP-treated incisors demonstrated ladder-like alternate rows of mineralized and non-mineralized dentin at the apical end. GBHA revealed moderate Ca reactions in the non-mineralized circumpulpal dentin matrix where electron microscopy revealed rich distributi

Therapeutic problems in cyanide poisoning.

In three patients with severe acute cyanide poisoning, a cyanosis was observed instead of the bright pink skin coloration often mentioned as a sign in textbooks. Treatment of cardiopulmonary insufficiency is as essential as antidotal therapy and the use of sodium nitrite and 4-DMAP is not without risk as, in practice, the methemoglobin-level induced is difficult to control.

Epinephrine-mediated sensitization of thioredoxin 1 enhances chlorpromazine protection against acute hepatotoxicity.

Acute liver injury (ALI) is characterised by rapid hepatic necrosis and compromised liver functions. Drug-induced ALI, in particular, is associated with glutathione depletion and overproduction of reactive oxygen species (ROS). These alterations highlight the contribution of the thioredoxin system (Trx/TrxR/NADPH) in preserving hepatic redox homeostasis. Previously, we demonstrated that thioredoxin-1 (Trx1) overexpression exerts protective effects against ischemic heart and peripheral vascular disease. Also, the neuroprotective role of thioredoxin reductase (TrxR1) has been reported in Parkins

Distinct cadmium detoxicity response between rice cultivars carrying functional or loss-of-function OsHMA3 allele.

BACKGROUND AND AIMS: Plants encounter various abiotic stress during their growth, making it essential to understand their responses to such challenges. Though function of OsHMA3 in cadmium (Cd) sequestration into rice root vacuoles has been well characterized, little is known about how different OsHMA3 alleles influence plant responses to high Cd exposure. METHODS: In this study, we utilized rice cultivars carrying a functional OsHMA3 allele (9311) and a loss-of-function OsHMA3 allele (W4) to investigate their differences in Cd detoxification responses. Morphological, physiological and subcel

Photochemically Engineered, DNA-Functionalized AgCl@AuAg Nanoprobes for Colorimetric Detection of Target DNA via In Situ Catalytic Amplification.

Colorimetric DNA detection is attractive for point-of-care testing, yet sensitivity often remains insufficient for clinically relevant applications/targets. We report a signal-amplification strategy that uses DNA-functionalized AgCl@AuAg nanoparticles (DNA-AgCl@AuAgNPs) as chemically transformable catalytic nanoprobes. To overcome the inherent challenge of functionalizing ionic AgCl surfaces, we employed a photochemical reduction method to decorate cubic AgCl nanoparticles (AgClNPs) with Au/Ag metallic domains (AgCl@AuAgNPs). These metallic "bumps" serve as stable anchoring sites for thiol-mod

Preventive and therapeutic effects of cepharanthine on acetaminophen-induced acute liver and kidney injury in male laboratory mice.

AIM: This study aimed to evaluate the preventive and therapeutic effects of cepharanthine on APAP (N-acetyl-para-aminophenol )-induced liver and kidney injury in an experimental model. BACKGROUND: Acetaminophen (APAP) is widely used as an analgesic and antipyretic drug; however, its overdose can cause acute liver and kidney injury. Cepharanthine, a natural alkaloid with antioxidant properties, has been proposed as a potential protective agent. METHODS: In this experimental study 40 Male NMRI mice were divided into five groups: Control, Sham, APAP-treated, APAP + preventive cepharanthine, and
