Sleep architecture in infants with spinal muscular atrophy type 1.

OBJECTIVE: Few reports on sleep patterns of patients with spinal muscular atrophy type 1 (SMA1) have been published and none on sleep microstructure. The aim of this study was to analyze sleep architecture and microstructure in a group of infants with SMA1, compared with age- and sex-matched controls. METHODS: Twelve SMA1 patients (six males, mean age 5.9 months) and 10 controls (five males, mean age 4.8 months) underwent full polysomnography to evaluate their sleep architecture and microstructure by means of the cyclic alternating pattern (CAP). RESULTS: Compared with control children, SMA1…

The effect of age on rat rotator cuff muscle architecture.

BACKGROUND: Understanding rotator cuff muscle function during disease development and after repair is necessary for preventing degeneration … y 27%, but sarcomere lengths are nearly constant. Therefore, these normal growth-induced changes in architecture must be considered when muscle atrophy or fiber shortening is measured after rotator cuff tears in this model.

Does joint architecture influence the nature of intra-articular fractures?

INTRODUCTION: The architecture of joints has potentially the greatest influence on the nature of intra-articular fractures. We analysed a large number of intra-articular fractures with two aims: (1) to determine if the pattern of injuries observed supports our conjecture that the local skeletal architecture is an important factor and (2) to investigate whether associated dislocations further affect the fracture pattern. METHODS: A retrospective study of intra-articular fractures over a 3.5-year period; 1003 joints met inclusion criteria and were analysed. Three independent investigators deter…

Architecture of the human regulatory network derived from ENCODE data.

Transcription factors bind in a combinatorial fashion to specify the on-and-off states of genes; the ensemble of these binding events forms a regulatory network, constituting the wiring diagram for a cell. To examine the principles of the human transcriptional regulatory network, we determined the genomic binding information of 119 transcription-related factors in over 450 distinct experiments. We found the combinatorial, co-association of transcription factors to be highly context specific: distinct combinations of factors bind at specific genomic locations. In particular, there are significa

Cellular and tissue architecture of conjunctival membranes in epidemic keratoconjunctivitis.

PURPOSE: Conjunctival membranes are differentiated from pseudomembranes by bleeding on removal. The authors sought to study the cellular constituents and extracellular structure of formed conjunctival exudates in epidemic keratoconjunctivitis (EKC), and to determine whether these represent true conjunctival membranes. METHODS: Formed conjunctival exudates harvested from 2 patients with EKC were studied by immunohistochemistry. Adenoviral infection was evaluated by PCR. RESULTS: Each patient's tears were PCR positive for adenovirus, but not HSV-I. Both conjunctival specimens contained a fibri

Periodic leg movements in narcolepsy patients: impact on sleep architecture.

INTRODUCTION: We assessed the effect of periodic leg movements (PLMs) on sleep architecture in narcolepsy patients by comparing sleep patterns in narcoleptics of comparable age with and without PLMs. METHODS: Patients diagnosed with narcolepsy between January 1999 and December 2005 were divided into those with PLM index >or= 5/hr and <5/hr. Each patient underwent polysomnographic recording followed by a multiple sleep latency test (MSLT). RESULTS: There were 47 patients, of mean age 28.9 +/- 1.8 years, 31 (66%) with PLM index >or= 5/hr and 16 (34%) with PLM index <5/hr. Arousal index, PLM ar…

Three-dimensional realisation of muscle morphology and architecture using ultrasound.

Two-dimensional B-mode ultrasound imaging and motion tracking were combined to generate three-dimensional reconstructions of the medial gastrocnemius. Architectural and morphological features of this muscle could be visualised. The length of the gastrocnemius belly was measured in normally (ND) developing children and in children with spastic diplegic cerebral palsy (SDCP) who had plantarflexion contractures. Using a random effects linear model we demonstrated that the gastrocnemius muscle bellies of children with SDCP were shorter than those of ND children (P = 0.001) even when corrected for

Primary dystonia: is abnormal functional brain architecture linked to genotype?

The DYT1 dystonia mutation is associated with an abnormal metabolic brain network characterized by hypermetabolism of the basal ganglia, supplementary motor area, and the cerebellum. In this study, we quantified the activity of this network in carriers of other dystonia mutations to determine whether this functional abnormality is linked to genotype. The findings suggest that the DYT1 metabolic topography is not genotype specific and may be present in carriers of other dystonia mutations.

Methyl CpG binding proteins: coupling chromatin architecture to gene regulation.

A correlation between DNA methylation and transcriptional silencing has existed for many years. Recently, substantial progress has been reported in the search for proteins that interpret the regulatory information inherent in DNA methylation and translate this information into functional states, resulting in the identification of a family of highly conserved proteins, the MBD family. Direct connections between these proteins and histone modification enzymes have emerged as a common theme, implying that DNA methylation exerts its effects primarily through repressive chromatin architecture. Rece…

Architecture of the Streptomyces lividans DnaA protein-replication origin complexes.

The Streptomyces oriC region contains two clusters of 19 DnaA boxes separated by a spacer (134 bp). The Streptomyces DnaA protein consists, like all other DnaA proteins, of four domains: domain III and the carboxyterminal part (domain IV) are responsible for binding of ATP and DNA, respectively. Binding of the DnaA protein to the entire oriC region analysed by electron microscopy showed that the DnaA protein forms separate complexes at each of the clusters of DnaA boxes, but not at the spacer separating them. In vivo mutational analysis revealed that the number of DnaA boxes and the presence o

Abnormal neuritic architecture identified by Di-I in Pick's disease.

The fluorescent stain Di-I combined with confocal microscopy identified an abnormal neuritic pattern in the cortices of frontal and anterior … r neurites compared to controls. The occipital cortex, which is unaffected in PD, showed a neuritic architecture comparable to normal controls as did affected brain tissues from patients with Alzheimer's disease or remote, ischemic infarction. This neuritic pattern is, thus far, unique to PD and may reflect the loss of specific subpopulations of cortical neuro …

Sleep architecture and sleep apnea in patients with Cushing's disease.

Patients with Cushing's syndrome (CS) frequently have sleep complaints. We evaluated sleep polysomnographically in 22 patients, including 17 … subjects. Nonapneic CD patients differed strikingly from healthy volunteers in sleep continuity and architecture, demonstrating lighter, fragmented sleep. Rapid eye movement (REM) sleep in CD patients bore many similarities to the sleep of patients with major depression, with REM latency being significantly shortened and REM density significantly increased. Duodenal mucosal architecture in non-specific and ulcer-associated duodenitis.

This study was done to determine the severity and extent of abnormalities of duodenal mucosal architecture in non-specific (non-ulcerative) and ulcer-associated duodenitis. The effect of successful treatment with cimetidine on these changes has also been assessed. A method of microdissection and measurement of villus height, crypt depth, and mitotic figure count per crypt was applied to endoscopic biopsies from the duodenum. Five groups of patients were studied: untreated ulcer-associated duodenitis, untreated non-specific duodenitis, healed ulcer-associated and non-specific duodenitis after c…

Human stapes crura: surface bone architecture: scanning electron microscopic findings.

Human stapes crura obtained at the time of stapedectomy for clinical otosclerosis have been studied after removal of the mucosal and periosteal layers by treatment with hydrogen peroxide and ultrasonification. With the scanning electron microscope, four basic architectural patterns are evident: honeycomb, fibrillar, compact and pitted. The significance of these patterns is as yet unclear, but they do appear to be in intimate relationship one to another and perhaps are varying stages in a single process.

Architectural patterns of growth of colon adenocarcinoma - comparative study.

BACKGROUND AND OBJECTIVES: Colorectal adenocarcinoma (CR ADK) may show either a uniform architectural pattern or intratumoral architectural heterogeneity, but the clinicopathological relevance of these growth patterns is still insufficiently characterized. AIM: The study's aim was to compare the clinical and morphological profiles of tumors with a single dominant architectural pattern and tumors with two coexisting architectural patterns. PATIENTS, MATERIALS AND METHODS: We analyzed 104 CR ADKs, divided into two groups: one group of 78 patients with a single architectural pattern all over th

Programmable Anisotropy: From Mesophase Order to Functional Architectures.

Across biology, structural hierarchy converts bottom-up self-assembly into macroscopic function, yielding emergent mechanical, transport, and optical behaviors. Liquid crystals (LCs) are uniquely positioned in this realm by coupling fluidity with long-range orientational order, providing a physics-grounded toolkit: director fields, elasticity, chirality, and topological defects for sculpting free-energy landscapes that organize matter with molecular precision. Crucially, these soft blueprints can be transduced into solids, preserving order and programming function, which offers a route to nano

Unseen, Ungrieved: Idiocy and the Architecture of Exclusion in Eugenic Institutions.

This essay reexamines institutional segregation during the eugenics era by centering the experiences of individuals labeled "low-grade" or "unimprovable," who have been largely excluded from both public life and historical memory. While scholarship on eugenics has emphasized sterilization, immigration restriction, and the politics of heredity, it has paid comparatively little attention to how significant intellectual disability was discursively and materially constructed within custodial institutions governed by medical and administrative authority. The essay argues that the figure of the "idi
