Aspergillosis is one of the most common fungal infections that can threaten individuals with immune compromised condition. Due to the increasing resistance of pathogens to the existing antifungal drugs, it is difficult to tackle such disease conditions. Whereas, nikkomycin is an emerging safe and effective antifungal drug which causes fungal cell wall disruption by inhibiting chitin synthase. Hence, the study aims at the development of nikkomycin loaded PEG coated PLGA nanoparticles for its increased antifungal efficiency and inhibiting Aspergillus infections. The P-PLGA-Nik NPs were synthesiz…

Aspergillus species are a major cause of life-threatening infections in immunocompromised hosts, and the most common pathogen of invasive aspergillosis is Aspergillus fumigatus. Recently, the development of molecular identification has revealed cryptic Aspergillus species, and A. felis is one such species within the Aspergillus section Fumigati reported in 2013. We describe a case of invasive pulmonary aspergillosis caused by A. felis in a 41-year-old Japanese woman diagnosed with myelodysplastic syndrome. She presented with fever 19 days after undergoing autologous peripheral blood stem cell …

While the epidemiology and clinical differences of various Candida spp. has been relatively well-identified, data regarding invasive aspergillosis (IA) caused by different Aspergillus spp. are insufficient. We aimed to determine the epidemiology of culture-positive invasive pulmonary aspergillosis (IPA) and to compare the characteristics and outcomes of Aspergillus fumigatus IPA with those of non-fumigatus IPA in patients with hematologic diseases. All consecutive cases of IPA from 2011 to 2015 were reviewed retrospectively. There were 430 proven/probable IPA and 76 culture-positive proven/proba…

Maize is frequently contaminated with aflatoxins (AFs) and fumonisins (FBs), posing a significant global food safety threat. Understanding how environmental factors regulate fungal toxigenesis under global warming is crucial. This study systematically investigated the in vitro stress responses of Aspergillus flavus, Aspergillus parasiticus, and Fusarium verticillioides (strains isolated from contaminated maize) across broad gradients of temperature (15-40 °C) and water activity (0.93-0.995 aw). Phenotypically, primary and secondary metabolism exhibited striking divergence: A. flavus grew optim…

Aspergillus fumigatus has been included by the World Health Organization in the priority list of fungal pathogens because (i) it causes 90% of invasive aspergillosis cases, with a high mortality rate, and (ii) infections are becoming increasingly resistant to azole antifungals. A. nidulans is an opportunistic pathogen and a saprotroph which has served during the last 80 years as a reference system for filamentous fungi. Here, we characterized the role in morphogenesis and development of the putative transcriptional cyclin/kinase complex CTDK-1 in both aspergilli. The null mutants of the corres…

In most cases, invasive aspergillosis (IA) is caused by A. fumigatus, though infections with other Aspergillus spp. with lower susceptibilities to amphotericin B (AmB) gain ground. A. terreus, for instance, is the second leading cause of IA in humans and of serious concern because of its high propensity to disseminate and its in vitro and in vivo resistance to AmB. An early differentiation between A. fumigatus and non-A. fumigatus infections could swiftly recognize a potentially ineffective treatment with AmB and lead to the lifesaving change to a more appropriate drug regime in high-risk pati…

It is unclear whether differences exist in baseline serum galactomannan (sGM) in patients with hematologic malignancies and invasive pulmonary aspergillosis (IPA) caused by non-fumigatus Aspergillus species vs Aspergillus fumigatus. We found no differences in baseline sGM positivity rates, median sGM levels, and 42-day mortality in 72 such patients (Aspergillus fumigatus in 43 and non-fumigatus Aspergillus in 29).

An animal model of disseminated aspergillosis was used to test the in-vivo activity of itraconazole against four isolates of Aspergillus fumigatus. Two reference isolates of A. fumigatus known to be resistant to itraconazole in vitro and in vivo were used as control isolates, and two new isolates were tested under the same conditions. For each isolate MICs for itraconazole and amphotericin B were determined by an NCCLS-based method. Mice infected intravenously were treated either with itraconazole 100 mg/ kg/day or amphotericin B 4.5 mg/kg/day for 10 days. Amphotericin B showed good in-vivo ac…

The effects of illumination, near-ultraviolet, incubation temperature pH and some minor elements on the growth rate and production of aflatoxin B1 by A. flavus and ochratoxin A by A. ochraceus were investigated. Aflatoxin B1 and ochratoxin A production was considerably higher in the light than in the dark. The greatest aflatoxin B1 and ochratoxin A production was occurred after 11 days of fermentation with light- and dark-grown cultures at 25 degrees C. The mycelial dry weight was also greater in the light than in the dark for both A. flavus and A. ochraceus. Exposure of conidia to near-UV irr

Triazole antifungals inhibit the lanosterol C14-demethylase Cyp51A/B and are the first-line treatment for invasive aspergillosis caused by Aspergillus fumigatus . We previously proposed that triazoles impose a secondary constraint on ergosterol biosynthesis through negative feedback regulation of HMG-CoA reductase (Hmg1) through its sterol sensing domain (SSD), and SSD mutations compromise this regulation to produce resistance. The molecular mechanism underpinning this feedback remained unknown. In this study, we show that voriconazole drives accelerated protein degradation of Hmg1 and that SS…

Fungal peritonitis is a rare but serious complication of peritoneal dialysis that is associated with high morbidity and mortality. Candida species are the predominant pathogens, whereas Aspergillus peritonitis is less common but often fatal. We report a rare case of Aspergillus oryzae peritonitis in a 54-year-old man undergoing continuous ambulatory peritoneal dialysis for diabetic end-stage renal disease. The patient presented with turbid peritoneal dialysate containing fibrinous debris and had a prior history of Enterobacter cloacae peritonitis. Laboratory tests showed elevated C-reactive pr…

Glucoamylases are highly demanded by the industry for starch saccharification, which necessitates an increase in enzyme activity. To improve the activity of Aspergillus awamori VKPM F-1262 glucoamylase N181Q (AGAM), amino acid substitutions were made in the region of substrate binding. In the case of the variant D237G, there was a 1.3-1.4-fold increase in activity towards soluble starch for the individual enzyme and crude enzyme solution after cultivation in flasks. For the variants G57A and C319T, there was also a 1.1-1.4-fold increase in enzyme activity for the crude flask solution, while th…

UNLABELLED: Diagnosing pulmonary diseases caused by non-fumigatus Aspergillus species remains challenging. We conducted a single-center, retrospective observational study in a French Respiratory Medicine Department. Patients with at least one respiratory sample positive for a non-fumigatus Aspergillus species, without concurrent A. fumigatus isolation over a 12-month period, were included. The primary objective was to determine the prevalence of pulmonary events (colonization or pulmonary diseases) associated with non-fumigatus Aspergillus species. Secondary objectives included species charact…

INTRODUCTION: Aspergillus fumigatus is a major airborne fungal pathogen that causes invasive aspergillosis in immunocompromised individuals. This study aims to elucidate the autophagic mechanisms activated following the internalization of A. fumigatus conidia in human bronchial epithelial cells.
METHODS: Specifically, we investigated the role of ULK1 and the autophagic processes in Beas2B cells upon A. fumigatus conidia internalization. The Beas2B cell line was used to assess the protein expression of ULK1, phosphorylated ULK1, and LC3-I/II via Western blotting. Autophagosome structures were …

Azole-resistant Aspergillus, particularly azole-resistant Aspergillus fumigatus (ARAf), is an increasingly consequential problem in Japan because triazoles remain central to aspergillosis management and chronic pulmonary aspergillosis (CPA) often requires prolonged oral therapy. CPA provides a within-host evolutionary niche in which persistent fungal burden and long-term azole exposure select resistant subpopulations, contributing to non-response, relapse, and progressive lung destruction. This review synthesizes Japan-specific clinical and environmental epidemiology, resistance mechanisms, an…

INTRODUCTION: The global prevalence of non-tuberculous mycobacterial pulmonary disease (NTM-PD) is increasing. Individuals with NTM-PD frequently develop chronic pulmonary aspergillosis, which is associated with poor clinical outcomes. However, the biological mechanisms underlying the interaction between Aspergillus species and non-tuberculous mycobacteria (NTM) remain poorly understood. This study aimed to investigate the interaction between Aspergillus fumigatus and NTM and to identify mechanisms that may inform novel therapeutic strategies.
METHODS: A. fumigatus was incubated with NTM cult…

Immune suppression heightens the risk for fungal infections, but the mechanisms that result in clinical disease are poorly understood. Here we demonstrate that macrophage ferroptosis, an iron-dependent form of regulated cell death, inhibits Aspergillus fumigatus ( Af ) killing. In a mouse tracheal transplant model of Af infection, we observed an increase in macrophage lipid peroxidation, a decreased expression of negative ferroptosis regulators Gpx4 and Slc7a11 , and an increase in positive regulators Ptgs2 and Nox2 , relative to syntransplants. Depletion of macrophages in transplant recipient…

Aneuploidy causes genome plasticity and enables adaptive responses that confer drug resistance in eukaryotes ranging from fungal pathogens to human cancer cells. Aspergillus fumigatus is a soil-resident fungus and the most common cause of invasive fungal infections globally. Aspergillosis has an alarmingly high mortality rate due in part to treatment failure resulting from widespread antimicrobial resistance and an ability of the fungus to dynamically adapt to changing environmental and host conditions. Our understanding of these treatment failures is hindered by gaps in knowledge about how th…
