District-representative data are rarely collected in the surveys for identifying localised disparities in Bangladesh, and so district-level estimates of undernutrition indicators - stunting, wasting and underweight - have remained largely unexplored. This study aims to estimate district-level prevalence of these indicators by employing a multivariate Fay-Herriot (MFH) model which accounts for the underlying correlation among the undernutrition indicators. Direct estimates (DIR) of the target indicators and their variance-covariance matrices calculated from the 2019 Bangladesh Multiple Indicato

Dichotomization is often used on clinical and diagnostic settings to simplify interpretation. For example, a person with systolic and diastolic blood pressure above 140 over 90 may be prescribed medication. Blood pressure as well as other factors such as age and cholesterol and their interactions may lead to increased risk of certain diseases. When using a dichotomized variable to determine a diagnosis, if the interactions with other variables are not considered, then an incorrect threshold for the continuous variable may be selected. In this paper, we compare single dichotomization with joint

Functional connectivity is defined as the undirected association between two or more functional magnetic resonance imaging (fMRI) time series. Increasingly, subject-level functional connectivity data have been used to predict and classify clinical outcomes and subject attributes. We propose a single-index model wherein response variables and sparse functional connectivity network valued predictors are linked by an unspecified smooth function in order to accommodate potentially nonlinear relationships. We exploit the network structure of functional connectivity by imposing meaningful sparsity c

Determining the effect of vaccine-induced immune response on disease risk is an important goal of vaccinology. Typically, immune correlates analyses are conducted prospectively with immune response measured shortly after vaccination and subsequent disease status regressed on immune response. In outbreaks and rare disease settings, collecting samples from all vaccinees is not feasible. The test negative design is a retrospective design used to measure vaccine efficacy where symptomatic individuals who present at a clinic are assessed for relevant disease (cases) or some other disease (controls)

In kin-cohort studies, clinicians want to provide their patients with the most current cumulative risk of death arising from a rare deleterious mutation. Estimating the cumulative risk is difficult when the genetic mutation status is unknown and only estimated probabilities of a patient having the mutation are available. We estimate the cumulative risk for this scenario using a novel nonparametric estimator that incorporates covariate information and dynamic landmark prediction. Our estimator has improved prediction accuracy over existing estimators that ignore covariate information. It is bui

Family studies involve the selection of affected individuals from a disease registry who provide right-truncated ages of disease onset. Coarsened disease histories are then obtained from consenting family members, either through examining medical records, retrospective reporting, or clinical examination. Methods for dealing with such biased sampling schemes are available for continuous, binary, and failure time responses, but methods for more complex life history processes are less developed. We consider a simple joint model for clustered illness-death processes which we formulate to study cov

Measuring a biomarker in pooled samples from multiple cases or controls can lead to cost-effective estimation of a covariate-adjusted odds ratio, particularly for expensive assays. But pooled measurements may be affected by assay-related measurement error (ME) and/or pooling-related processing error (PE), which can induce bias if ignored. Building on recently developed methods for a normal biomarker subject to additive errors, we present two related estimators for a right-skewed biomarker subject to multiplicative errors: one based on logistic regression and the other based on a Gamma discrimi

Potential disease-modifying therapies for neurodegenerative disorders need to be introduced prior to the symptomatic stage in order to be effective. However, current diagnosis of neurological disorders mostly rely on measurements of clinical symptoms and thus only identify symptomatic subjects in their late disease course. Thus, it is of interest to select and integrate biomarkers that may reflect early disease-related pathological changes for earlier diagnosis and recruiting pre-sypmtomatic subjects in a prevention clinical trial. Two sources of biological information are relevant to the cons

We propose a Bayesian approach to Mendelian randomization (MR), where instruments are allowed to exert pleiotropic (i.e. not mediated by the exposure) effects on the outcome. By having these effects represented in the model by unknown parameters, and by imposing a shrinkage prior distribution that assumes an unspecified subset of the effects to be zero, we obtain a proper posterior distribution for the causal effect of interest. This posterior can be sampled via Markov chain Monte Carlo methods of inference to obtain point and interval estimates. The model priors require a minimal input from t

Low-density lipoprotein cholesterol (LDL-C) has been identified as a causative factor for atherosclerosis and related coronary heart disease, and as the main target for cholesterol- and lipid-lowering therapy. Statin drugs inhibit cholesterol synthesis in the liver and are typically the first line of therapy to lower elevated levels of LDL-C. On the other hand, a different drug, Ezetimibe, inhibits the absorption of cholesterol by the small intestine and provides a different mechanism of action. Many clinical trials have been carried out on safety and efficacy evaluation of cholesterol lowerin

In this paper, we further extend the recently proposed Poisson-Tweedie regression models to include a linear predictor for the dispersion as well as for the expectation of the count response variable. The family of the considered models is specified using only second-moments assumptions, where the variance of the count response has the form μ+ϕμp $\mu + \phi \mu^p$, where µ is the expectation, ϕ and p are the dispersion and power parameters, respectively. Parameter estimations are carried out using an estimating function approach obtained by combining the quasi-score and Pearson estimating fun

Epidemiological research supports an association between maternal exposure to air pollution during pregnancy and adverse children's health outcomes. Advances in exposure assessment and statistics allow for estimation of both critical windows of vulnerability and exposure effect heterogeneity. Simultaneous estimation of windows of vulnerability and effect heterogeneity can be accomplished by fitting a distributed lag model (DLM) stratified by subgroup. However, this can provide an incomplete picture of how effects vary across subgroups because it does not allow for subgroups to have the same wi

Recurrent event data arise frequently in many longitudinal follow-up studies. Hence, evaluating covariate effects on the rates of occurrence of such events is commonly of interest. Examples include repeated hospitalizations, recurrent infections of HIV, and tumor recurrences. In this article, we consider semiparametric regression methods for the occurrence rate function of recurrent events when the covariates may be measured with errors. In contrast to the existing works, in our case the conventional assumption of independent censoring is violated since the recurrent event process is interrupt

In longitudinal data analyses, the observation times are often assumed to be independent of the outcomes. In applications in which this assumption is violated, the standard inferential approach of using the generalized estimating equations may lead to biased inference. Current methods require the correct specification of either the observation time process or the repeated measure process with a correct covariance structure. In this article, we construct a novel pairwise likelihood method for longitudinal data that allows for dependence between observation times and outcomes. This method invest

Recurrent event data are commonly encountered in clinical and epidemiological studies. A major complication arises when recurrent events are terminated by death. To assess the overall effects of covariates on the two types of events, we define a weighted composite endpoint as the cumulative number of recurrent and terminal events properly weighted by the relative severity of each event. We propose a semiparametric proportional rates model which specifies that the (possibly time-varying) covariates have multiplicative effects on the rate function of the weighted composite endpoint while leaving

We address the statistical challenge of classifying subjects as hemiplegic, vestibular or normal based on complex trajectories obtained through two experimental protocols designed to evaluate potential deficits in postural control. The classification procedure involves a dimension reduction step where the complex trajectories are summarized by finite-dimensional summary measures based on a stochastic process model for a real-valued trajectory. This allows us to retrieve from the trajectories information relative to their temporal dynamic. A leave-one-out evaluation yields a 79% performance of

Multiple outcomes multivariate meta-analysis (MOMA) is gaining in popularity as a tool for jointly synthesizing evidence coming from studies that report effect estimates for multiple correlated outcomes. Models for MOMA are available for the case of the pairwise meta-analysis of two treatments for multiple outcomes. Network meta-analysis (NMA) can be used for handling studies that compare more than two treatments; however, there is currently little guidance on how to perform an MOMA for the case of a network of interventions with multiple outcomes. The aim of this paper is to address this issu

The recent availability of survey data on social contact patterns has made possible important advances in the understanding of the social determinants of the spread of close-contact infections, and of the importance of long-lasting contacts for effective transmission to occur. Still, little is known about the relationship between two of the most critical identified factors (frequency of contacts and duration of exposure) and how this relationship applies to different types of infections. By integrating data from two independently collected social surveys (Polymod and time use), we propose a mo

Malignant brain tumors rank second in both incidence and mortality by cancer in children, and they are the leading cause of cancer death in children. Relatively little is known about the etiology of childhood brain tumor (CBT). While there are several studies which link pesticide exposure to increased risk of CBT, findings have been inconsistent. We performed a meta-analysis on 15 published epidemiological studies to test that in utero exposure to pesticides may be involved in the development of brain cancer in children. Meta-analysis was performed using the general variance-based method and h

The detection of differentially expressed (DE) genes, that is, genes whose expression levels vary between two or more classes representing different experimental conditions (say, diseases), is one of the most commonly studied problems in bioinformatics. For example, the identification of DE genes between distinct disease phenotypes is an important first step in understanding and developing treatment drugs for the disease. We present a novel approach to the problem of detecting DE genes that is based on a test statistic formed as a weighted (normalized) cluster-specific contrast in the mixed ef
