Extracellular vesicles (EVs) are emitted from cells throughout the body and serve as signaling molecules that mediate disease development. Emerging evidence suggests that per- and polyfluoroalkyl substances (PFAS) impact EV release and content, influencing liver toxicity. Still, the upstream regulators of EV changes affected by PFAS exposure remain unclear. This study evaluated the hypothesis that PFAS exposures, individually and in a mixture, alter the expression of genes involved in EV regulation at concentrations comparable to genes involved in global biological response mechanisms. Perfluorooctanoic acid (PFOA) is a globally pervasive environmental contaminant characterized by chemical stability and bioaccumulation through the food chain, posing serious health risks to both humans and animals. Accumulating evidence has linked PFOA exposure to the development of various cancers, including prostate cancer (PCa), renal cancer, breast cancer, and ovarian cancer. However, the molecular mechanisms by which PFOA promotes PCa progression remain unclear. In this study, we demonstrate that low-dose PFOA exposure enhances the expression of mitogen-activated protein kinase 15 (MAPK1

The growing demand for phosphorus recovery from wastewater has increased interest in Enhanced Biological Phosphorus Removal (EBPR) processes. However, the fate of per- and polyfluoroalkyl substances (PFAS) in EBPR-derived biosolids remains poorly understood. This study investigated the adsorption behavior of four PFAS: perfluorooctanoic acid (PFOA), perfluorooctanesulfonic acid (PFOS), perfluorooctanesulfonamide (PFOSA), and perfluorotetradecanoic acid (PFTDA) in EBPR sludge and assessed the long-term soil risks following biosolid application. Results showed that PFAS adsorption was significan

While the occurrence of GenX, a novel alternative for perfluorooctanoic acid (PFOA), in the environment and its cytotoxicity at high concentrations to thyroid cells are well documented, limited information is available regarding its impact at low concentrations. GenX is detected to be as low as 0.001 ng/mL in drinking water and 0.04-1 ng/mL in human serum. In this study, we investigated the impact of GenX at levels similar to that from drinking water and human serum on thyroid function in vitro and in vivo. Our data showed that GenX from 0.001 to 0.1 ng/mL was sub-cytotoxic and did not exhibit

Perfluorooctanoic acid (PFOA) is a commonly used short-chain synthetic perfluoroalkyl agent. Immature Leydig cells (ILCs) are localized in the testis and responsible for androgen biosynthesis and metabolism. Although PFOA shows toxicity in the reproductive system, it is not clear if it disrupts the function of ILCs. In the present study, primary ILCs were isolated from 35-day-old rats and exposed to a range of PFOA concentrations (0, 0.01, 0.1, or 1 μM). It was determined that 0.1 or 1 μM PFOA reduced total androgen biosynthesis in ILCs. Specifically, 22R-hydroxycholesterol (22R), and pregneno

This research aimed to evaluate the adsorption behaviors and mechanisms of perfluorooctanoic acid (PFOA) onto polyethyleneimine modified graphene oxide (GO-PEI) from aqueous solutions. The adsorption capacity was significantly improved by doping polyethyleneimine (PEI) onto graphene oxide (GO). The Brunauer-Emmett-Teller (BET) isotherm model was considered as the best isotherm model in describing the PFOA adsorption onto GO-PEI3 (wPEI/wGO = 3). GO-PEI3 exhibited high adsorption capacity (qe = 368.2 mg/g, calculated from BET isotherm model) and excellent stability. The maximum monolayer amount

BACKGROUND: Per- and polyfluoroalkyl substances (PFAS) are shown to have neurotoxic effects on animals, but epidemiological evidence for associations between childhood PFAS exposure and neurodevelopment is inconclusive. We examined if childhood PFAS concentrations are associated with a diagnosis of autism spectrum disorder (ASD), developmental delay (DD), and other early concerns (OEC) in development.
METHODS: We included 551 children 2-5 years old from the CHildhood Autism Risks from Genetics and Environment (CHARGE) case-control study. Children were clinically diagnosed and classified as ha

OBJECTIVE: At high altitude (HA), acute mountain sickness (AMS) is accompanied by neurologic and upper gastrointestinal symptoms (UGS). The primary aim of this study was to test the hypothesis that delayed gastric emptying (GE), assessed by 13 C-octanoate breath testing (OBT), causes UGS in AMS. The secondary aim was to assess post-gastric mechanisms of OBT, which could confound results under these conditions, by determination of intermediary metabolites, gastrointestinal peptides, and basal metabolic rate.
METHODS: A prospective trial was performed in 25 healthy participants (15 male) at 455

BACKGROUND/AIMS: The aim of this study was to determine whether Billroth I pancreaticogastrostomy (PG-I) or Billroth II pancreaticojejunostomy (PJ-II) after pylorus-preserving pancreatoduodenectomy is associated with better postoperative fat absorption, based on residual pancreatic exocrine function. Several reconstructive operations have been employed after pylorus-preserving pancreatoduodenectomy to maximize postoperative nutrition. However, no single-institution study has been published comparing the reconstructive procedures with respect to digestion and absorption of fat.

Various bifunctional reagents by the free NH2 group of ristomycinic acid of ristomycin A were used for selective chemical modification of the antibiotic. The bifunctional reagents were the following: di-N-hydroxysuccinimide ether of suberic acid and 4,4'-difluoro-3,3'-dinitrodiphenylsulfone. Bis-N,N'-derivatives of ristomycin A were prepared using these reagents. The derivatives inhibited the growth of Bac. subtilis but the concentrations required for the inhibition were 2-4 times higher than those of ristomycin A. It was noted that the MIC of the bis-N,N'-derivatives depended on the length an

The authors describe percutaneous treatment of gallbladder or bile duct stones in 18 patients who were poor surgical candidates or in whom conventional therapy failed. Dissolution was performed in most cases with methyl tert-butyl ether (MTBE) because of its potent dissolution properties; other solvents used included monooctanoin or chelating solutions. Gallbladder stones were eliminated in 11 of 13 patients (six of seven with dissolution alone, four of four with dissolution and basket extraction, one with basket removal alone). In five patients with stones in the common bile duct (n = 3), cys

Per- and poly-fluoroalkyl substances (PFAS) have been associated with hypertension during pregnancy, although human studies are inconsistent. We examined this relationship in the Puerto Rico PROTECT cohort (N = 548). PFAS concentrations were measured in maternal serum at 16-20 weeks' gestation. Blood pressure (BP) was measured at two time points (16-20 and 24-28 weeks). We used multivariable linear regression to estimate associations between PFAS and continuous systolic and diastolic BP, pulse pressure (PP), and mean arterial pressure (MAP). We examined hypertension, high PP, and high MAP as b

Exposure to per- and polyfluoroalkyl substances (PFAS) has been linked to renal impairment, but evidence on mixed PFAS exposure in the general Chinese population and the underlying mechanisms remains limited. This study therefore investigated the associations between PFAS exposure and renal function and evaluated the potential mediating role of blood lipids. Serum levels of legacy and emerging PFAS, renal function parameters, and blood lipid profiles were measured in 1370 adult residents from Zhejiang Province, China; additionally, 452 lipid metabolites were quantified in a subset of 316 parti

BACKGROUND: The C8 Science Panel previously conducted studies of perfluorooctanoic acid (PFOA) exposure in Ohio Valley residents, finding probable links with kidney and testicular cancers, raised cholesterol, ulcerative colitis, thyroid disease, and pre-eclampsia. Findings were largely based on a cohort of 32,254 persons with high PFOA exposure followed through 2011 for disease incidence. We have now followed this cohort through 2021 for mortality.
METHODS: Deaths were ascertained via the National Death Index (NDI). We extended cumulative PFOA serum estimates from 2011 until 2021. We used int

Perfluorooctanoic acid (PFOA), a legacy perfluoroalkyl substance with immuno- and repro-toxicant effects, has poorly characterized bioaccumulation and distribution patterns in postpubertal female pigs. The potential for heat stress (HS) to influence PFOA partitioning, potentially through intestinal hyperpermeability and alterations in systemic blood flow, also warrants investigation. This study investigated PFOA uptake, accumulation, and distribution in thermal neutral (TN) and heat-stressed gilts. Pigs (n = 48) were estrus synchronized and experienced TN (20 °C) or HS (26.6 to 32.2 °C) condit

Perfluorooctanoic acid (PFOA) is a persistent environmental pollutant that has been associated with reproductive dysfunction, yet cellular mechanism underlying its ovarian toxicity remain poorly understood. From the perspective of cellular senescence, we conducted this study by establishing a murine (female ICR mice) PFOA exposure (10 mg/kg/day administered orally via gavage for 28 consecutive days), then examined changes in ovarian structure, cellular composition, and transcriptional profiles. Compared with the control group, we found significant reductions in follicle numbers across all deve

Perfluorooctanoic acid (PFOA) has raised public concern due to its widespread presence/use and toxic health effects including hepatotoxicity, neurotoxicity, and developmental toxicity. Because dietary intake is a major route of PFOA exposure, and milk is a primary source of nutrition in early life, the present systematic review discusses PFOA occurrence in milk samples and the employed determination methods. In the present article, 69 studies (published 2007-2024) reporting PFOA levels in infant formula, commercial milk, and human breast milk were included. The highest concentration of PFOA in

Medium-chain perfluoroalkyl carboxylic acids (MC-PFCAs) are frequently detected at elevated concentrations in various environmental matrices and human samples worldwide. MC-PFCA exposure is associated with a number of adverse health effects including male reproductive toxicity, but their toxicological mechanisms remain unclear. This study investigated the male reproductive toxicity of MC-PFCAs and their molecular mechanisms by taking a combined approach of in vivo, in vitro and in silico studies at environmental and human exposure levels.
