Interstitial trophoblastic cell fusion and E-cadherin immunostaining in the placental bed of normal and hypertensive pregnancies.

During their invasion of the placental bed, interstitial trophoblasts fuse to multinuclear giant cells which are thought to have lost their invasive properties. Trophoblast fusion is associated with downregulation of E-cadherin, and persistent E-cadherin expression has been linked to defective placentation in preeclampsia. Since a previous study suggested 'premature' giant cell formation in preeclampsia, we started with the working hypotheses that fusion is increased in hypertensive pregnancies, and that the intensity of fusion correlates with the severity of disease. Using double immunostaini

Hematopoietic mobilization in mice increases the presence of bone marrow-derived hepatocytes via in vivo cell fusion.

The mechanisms for in vivo production of bone marrow-derived hepatocytes (BMDHs) remain largely unclear. We investigated whether granulocyte colony-stimulating factor (G-CSF)-mediated mobilization of hematopoietic cells increases the phenomenon. Recurrent liver injury in mice expressing green fluorescent protein (EGFP) in all hematopoietic-derived cells was produced by 3 months of carbon tetrachloride (CCL4) injections. Histologically, there were necrotic foci with histiocyte-rich infiltrates, but little oval cell proliferation. Subsequently, some animals were mobilized with G-CSF for 1, 2, or

The API2/MALT1 fusion product may lead to germinal center B cell lymphomas by suppression of apoptosis.

Low-grade B cell lymphomas of mucosa-associated lymphoid tissue (MALT) represent a distinct clinicopathological entity that arises in a wide variety of extranodal sites. Genetically, MALT lymphomas are characterized by the t(11;18)(q21;q21). The genes involved in this translocation have been identified to be API2 on chromosome 11, which encodes an apoptotic inhibitor, and MALT1, a novel gene on chromosome 18. We identified the t(11;18)(q21;q21) by Southern blot analysis and reverse transcriptase PCR in 42% of a panel of extranodal MALT lymphomas. We also identified the breakpoints within the A

Molecular mechanisms affecting HTLV type 1-dependent fusion at the cell membrane: implications for inhibiting viral transmission.

Infection with human T cell leukemia virus type 1 is detected by screening programs and contact follow-up procedures. Where chronic infection results in overt pathology, this is treated largely symptomatically and control of transmission relies on physical and educational constraints. The poor infectious transmission rate of HTLV-1 has long been described but to date has not been exploited in preventative measures to combat the spread of the virus. We undertook to investigate some of the molecular steps involved in HTLV-1 cell-cell fusion, the main mechanism of transmission. We showed that poo…

Frequent expression of the NPM-ALK chimeric fusion protein in anaplastic large-cell lymphoma, lympho-histiocytic type.

The revised European-American lymphoma classification recognizes a subtype of anaplastic large-cell lymphoma (ALCL), termed lympho-histiocytic because of its peculiar cytological composition. As in the case of classical ALCL, this tumor usually occurs in young patients and shows an excellent response to chemotherapy, but some authors have suggested that in reality this is a nonanaplastic T-cell lymphoma rich in histiocytes. In this paper, we show that three of five cases of lympho-histiocytic ALCL stain with anti-ALK antibodies and can therefore be presumed to express the chimeric NPM/ALK prot

Occurrence of TEL-AML1 fusion resulting from (12;21) translocation in human early B-lineage leukemia cell lines.

The recurrent (12;21)(p13;q22) translocation fuses the two genes TEL and AML1 that have previously been cloned from translocation breakpoints in myeloid leukemias. Using mainly reverse transcriptase-polymerase chain reaction (RT-PCR), the TEL-AML1 chimeric transcript has been observed in 22-27% of pediatric patients with acute lymphoblastic leukemia (ALL), in particular in the early B-lineage ALL subtype, making it the most common genetic lesion in these patients. The vast majority of acute myeloid leukemias, other ALL subtypes and even adults with early B-lineage ALL were TEL-AML1-negative. W

Renal Cell Carcinoma in Crossed Fused Renal Ectopia: A Case Report of a Rare Congenital Renal Fusion.

Crossed fused renal ectopia (CFRE) is a rare congenital renal fusion anomaly in which both kidneys are located on the same side of the body and are usually fused. Simultaneous occurrence of primary renal cell carcinoma (RCC) in CFRE represents a rare entity. Although CFRE may remain asymptomatic, it can be associated with complications such as urinary tract infections, obstruction, calculi, rarely malignancy, and tumors within the fused kidneys, which are exceptionally rare. Although the literature reports only a very small number of cases of CFRE coexisting with RCC, we present a case of a 46

Identification of a Novel, Oncogenic and Targetable TPR::ABL2 Fusion Gene in T-Cell Acute Lymphoblastic Leukaemia.

ABL2 rearrangements represent a subtype of acute lymphoblastic leukaemia (ALL) associated with poor prognosis and survival. This study reports a high-risk T-cell ALL (T-ALL) case with a novel TPR::ABL2 gene fusion resulting from a chromosomal deletion. Overexpression of TPR::ABL2 in Ba/F3 cells promoted cytokine-independent growth, demonstrating its oncogenic nature. Both primary patient and Ba/F3 cells carrying TPR::ABL2 exhibited kinase activation and sensitivity to tyrosine kinase inhibitors (TKIs). This study expands the repertoire of ABL2 fusions identified in ALL and supports the incorpo

Seminiferous Tubules-Targeted Resveratrol Carbon Dots With Sertoli Cell Biomimetic Vesicles Fusion for Improving Spermatogenesis of Cryptorchidism.

Impaired blood-testis barrier (BTB) integrity, induced by oxidative stress, is one of the primary factors leading to spermatogenic disorders in cryptorchidism. Therefore, the targeted elimination of testicular oxidative stress and repair of the BTB are promising strategies for promoting spermatogenesis in patients with cryptorchidism. This study aimed to demonstrate the potential of Sertoli cell membrane-camouflaged biomimetic vesicles loaded with resveratrol carbon dots (RCD@PCy@EVs) as a therapeutic nanoagent for cryptorchidism treatment in vivo. The prepared RCD@PCy@EVs are actively targete

Characterization of a PRKCE::ETV6 fusion as a potential oncogenic driver in T-cell acute lymphoblastic leukemia.

BACKGROUND: T-cell acute lymphoblastic leukemia (T-ALL) is an aggressive hematologic malignancy caused by mutation accumulation during hematopoiesis. The characterization of chromosomal abnormalities may provide significant insights into genetic mechanisms of malignant transformation in hematopoietic cells. However, T-ALL is genetically very heterogenous and driving mutations as well as clonal markers for the assessment of minimal residual disease are not always identifiable. Hence, there is a clinical need to further refine the genetic landscape of T-ALL including previously unrecognized fusi

scUCAF: An uncertainty-aware cross-omics alignment and fusion network for single-cell multi-omics data clustering.

The development of single-cell multi-omics sequencing technologies provides new insights into cell heterogeneity. Cell clustering is a crucial step in the analysis of multi-omics data. However, existing methods often overlook variations in data quality across omics, leading to unreliable feature representations. To address this issue, we propose scUCAF, an uncertainty-aware network for multi-omics clustering. Specifically, to mitigate the impact of noise on cell feature extraction, we introduce a variational autoencoder with a negative binomial distribution. After extracting each omics feature

TPM3-NTRK1 fusion confers resistance to osimertinib in lung adenocarcinoma: a model in a continuous cell line.

A 62-year-old woman with EGFR exon 19-mutant lung adenocarcinoma developed resistance to osimertinib after 13 months of treatment. At progression, genetic analysis revealed persistence of the EGFR mutation and acquisition of a TPM3-NTRK1 fusion. Cells from the pleural effusion at this stage were cultured and established as a continuous cell line, LUNK1. LUNK1 cells displayed epithelial characteristics, with a population doubling time of 57.66 h in vitro and 100% tumorigenicity in vivo. Xenograft histopathology showed poorly differentiated lung adenocarcinoma exhibiting solid, acinar, and micro

Selpercatinib-induced Acalculous Cholecystitis in an Elderly Patient with RET Fusion-positive Non-small Cell Lung Cancer.

The effectiveness of selpercatinib against RET-fusion-positive non-small cell lung cancer (NSCLC) in geriatric patients is underreported. We report the case of an 84-year-old woman in whom reduced-dose selpercatinib elicited a significant tumor response in KIF5B-RET fusion-positive NSCLC. However, she developed acalculous cholecystitis, which led to the discontinuation of the drug. This is the first reported case of selpercatinib-induced acalculous cholecystitis in a patient. The report highlighted the potential efficacy of selpercatinib in elderly patients while underscoring the risk of sever

Enhancement of polyethylene glycol-cell fusion efficiency by novel application of transient pressure using a jet injector.

Cell-cell fusion involves the fusion of somatic cells into a single hybrid cell. It is not only a physiological process but also an important cell engineering technology which can be applied to various fields, such as regenerative medicine, antibody engineering, genetic engineering, and cancer therapy. There are three major methods of cell fusion: electrical cell fusion, polyethylene glycol (PEG) cell fusion, and virus-mediated cell fusion. Although PEG cell fusion is the most economical approach and does not require expensive instrumentation, it has a poor fusion rate and induces a high rate …

SARS-CoV-2 spike protein-induced cell fusion activates the cGAS-STING pathway and the interferon response.

Severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) caused the unprecedented coronavirus disease 2019 (COVID-19) pandemic. Critical cases of COVID-19 are characterized by the production of excessive amounts of cytokines and extensive lung damage, which is partially caused by the fusion of SARS-CoV-2-infected pneumocytes. Here, we found that cell fusion caused by the SARS-CoV-2 spike (S) protein induced a type I interferon (IFN) response. This function of the S protein required its cleavage by proteases at the S1/S2 and the S2' sites. We further showed that cell fusion damaged nuclei a…

SEPT6_TRIM33 Gene Fusion and Mutated TP53 Pathway Associate With Unfavorable Prognosis in Patients With B-Cell Lymphomas.

BACKGROUND: Mounting studies have sought to identify novel mutation biomarkers having diagnostic and prognostic potentials. Nevertheless, the understanding of the mutated pathways related to development and prognosis of B-cell lymphoma is still lacking. We aimed to comprehensively analyze the mutation alterations in genes of canonical signaling pathways and their impacts on the clinic outcomes of patients with B-cell lymphoma. METHODS: Circulating cell-free DNA (cfDNA) samples from 79 patients with B-cell lymphomas were used for targeted sequencing with a 560-gene panel for depicting mutation

Inhibition of tissue factor-dependent and -independent coagulation by cell surface expression of novel anticoagulant fusion proteins.

BACKGROUND: Thrombotic vascular occlusion occurs in disorders of diverse etiology, including atherosclerosis, vasculitis, and disseminated intravascular coagulation. The same process results in hyperacute rejection of renal allografts transplanted into sensitized patients and remains a major problem in experimental xenotransplantation. METHODS: We have previously described the design and expression of several genetic constructs encoding novel fusion proteins with anticoagulant properties. They are based on two naturally occurring soluble anticoagulant proteins, human tissue factor pathway inh

Proteolysis and fusion of low density lipoprotein particles independently strengthen their binding to exocytosed mast cell granules.

Contact between low density lipoproteins (LDL) and exocytosed mast cell granules, the "granule remnants," leads to binding of LDL to the granule remnants via ionic interactions between the apolipoprotein B-100 (apoB-100) component of LDL and the heparin proteoglycan component of the granule remnants. Upon incubation at 37 degrees C, the heparin proteoglycan-bound apoB-100 is progressively proteolyzed by remnant chymase and carboxypeptidase A, which are also bound to the heparin proteoglycans. Thereupon, the LDL particles fuse, and their binding to the granule remnants strengthens, as defined b

A Cell-Penetrating Peptide and Signal Sequence Fusion DNA Vaccine Enhances CD8+ T Cell-Mediated Anti-Tumor Immunity Targeting HPV-16 Cervical Cancer Antigens.

BACKGROUND: Cervical cancer is the fourth most common cancer affecting women, with almost all cases caused by infection with Human Papillomavirus (HPV). Previous research has explored the potential of cell-penetrating peptides to improve the efficacy of peptide vaccines and signal peptide sequences to increase protein secretion in DNA vaccines. In this study, we evaluated the in vivo efficacy of DNA vaccines targeting HPV-16 E6 and E7 viral antigens that are constructed with both a cell-penetrating peptide and IL-2 signal peptide to improve immunogenicity. RESULTS: We generated DNA vaccine co
