Control of lumbar sympathetic nerve traffic by vagal cardiopulmonary baroreflexes in renal hypertension.

Renal nerve traffic is inhibited during volume expansion mainly because of stimulation of vagal cardiopulmonary baroreflexes. These responses are impaired in rabbits with renal hypertension caused by impaired cardiopulmonary baroreflexes. Previous studies have suggested that there may be differences in the extent to which renal hypertension alters reflex control of lumbar as compared with renal sympathetic nerve activity. The goals of this study were to determine if the responses to volume expansion of lumbar sympathetic nerve traffic also are impaired in renal hypertension and if this abnorma

Ocular effects of a relatively selective alpha 2 agonist (UK-14, 304-18) in cats, rabbits and monkeys.

UK-14, 304-18 (UK), a relatively selective alpha 2-agonist, was examined for its effects on intraocular pressure (IOP) and pupil diameter (PD) in rabbits, cats and monkeys and on noradrenergic function in the cat nictitating membrane (CNM) preparation. Topical, unilateral administration of UK (0.0005-0.5 mg) produced dose-dependent decreases in IOP and pupil size in normal, unanesthetized rabbits, cats and monkeys. The ocular hypotensive effect of UK in the ipsilateral eye was delayed relative to the contralateral eye in all three species; UK produced an initial transient ocular hypertension i

The inotropic actions of N-acetylprocainamide: blockade and reversal by propranolol.

The inotropic actions of N-acetylprocainamide (NAPA) were studied in chloralose-urethane anesthetized dogs. Myocardial contractile force was measured with a Walton-Brodie strain gauge sutured to the right ventricle. A low-dose NAPA infusion (12 mg/kg i.v.) increased myocardial force by a maximum of 11.6 +/- 2.4% (mean +/- SEM), whereas a high dose of NAPA (60 mg/kg i.v.) increased myocardial force by 33.3 +/- 2.6% at the peak of the effect. The high-dose NAPA infusion also caused significant reductions in heart rate and blood pressure, while the low-dose NAPA infusion lacked significant chrono

Respiratory effects of brief baroreceptor stimuli in the anesthetized dog.

To quantify the immediate isocapnic respiratory response to baroreceptor stimulation, pressure in the isolated externally perfused carotid sinuses (CS) of 24 vagotomized alpha-chloralose-anesthetized dogs was increased selectively during either inspiration or expiration as a step (from time of onset to end of respiratory phase) or a pulse (500 ms). The rise time (150 ms), base-line pressure (80 mmHg), and stimulus magnitude (40 mmHg) were similar for the two stimuli. The time of stimulus onset (delay), expressed as a percent of control time of inspiration (TI) or expiration (TE), was varied. T

Kidney function during common carotid artery occlusion in anaesthetized cats: influence of vagotomy, constant ventilation, blood pressure stabilization, and carotid body chemoreceptor inactivation.

The reactions of kidney function elicited by bilateral common carotid artery occlusion were studied in six groups of chloralosed cats in which the Nn. vagi, the breathing reaction, the increase of the mean systemic arterial blood pressure, and the carotid body chemoreceptors were excluded successively. Carotid occlusion in the control animals caused a rise of the mean systemic arterial blood pressure, hyperventilation, and an increase in renal sodium and water excretion, resulting from an inhibition of tubular reabsorption. Bilateral cervical vagotomy, relaxation and constant artificial ventil

The effects of naloxone on the peripheral sympathetics in cat endotoxin shock.

The effects of naloxone on sympathetic nerve activity during E coli endotoxin hypotension were studied in alpha-chloralose anesthetized cats. Blood pressure and left preganglionic splanchnic nerve (PSN) activity were measured. Naloxone (2 mg/kg) was injected intravenously 30 min after endotoxin. In control group, 30 min after endotoxin, both mean blood pressure (MBP) and PSN activity decreased by 68 +/- 7% and 47 +/- 10%, respectively, and were followed by a gradual decline, so that after 60 min MBP and PSN activity were 54 +/- 8% and 48 +/- 7% of the pre-endotoxin level. The reduced blood pre

Release of opioid peptides in anaesthetized cats?

1 The effect on arterial blood pressure of intravenous injections of naloxone (200 mug) was examined in cats anaesthetized with chloralose. Usually these injections have no effect on blood pressure unless morphine or opioid peptides have been injected, when they produce a pressor response with tachycardia.2 It was found that these injections produced a pressor response with tachycardia after a combination of two or more of the following surgical procedures: (1) tying sinus nerves, (2) removing stellate ganglia, (3) cutting vagi, (4) evisceration.3 The pressor responses obtained in these condit

[Mode of action of clonidine, a central hypotensive agent, analyzed by the interaction with various sympathomimetic agents].

Injection of a low dose of clonidine showed the biphasic blood pressure response which comprises a delayed onset of long lasting hypotension and a quick onset of hypertension of short duration followed by the former, in the alpha-chloralose-urethanized rat. A second injection of low dose of clonidine administered 90 min after the first showed only two peaks of pressor response, no longer a depressor one, and tachyphylaxis occurred with repeated injections of the same dose of clonidine, together with a gradual elevation of blood pressure level. Furthermore, the depressor action was inhibited by

Exposure of Domestic Cats (Felis catus) to Rodenticidal Compounds.

Anticoagulant rodenticides (ARs) are highly effective, but can be of environmental concern due to primary and secondary non-target exposure, with the latter possible being relevant to domestic cats. Therefore, liver residues of ARs and an alternative rodenticide, α-chloralose, were systematically monitored in domestic cats for the first time in the current study. In 2021 and 2022, the carcasses of 99 cats were collected in Slovenia and liver residues were measured by using solid supported liquid-liquid extraction and LC-MS/MS. The results show that 65% of cats carried at least one rodenticide.

The Effects of Mutual Interaction of Orexin-A and Glucagon-Like Peptide-1 on Reflex Swallowing Induced by SLN Afferents in Rats.

(1) Background: Our previous studies revealed that orexin-A, an appetite-increasing peptide, suppressed reflex swallowing via the commissural part of the nucleus tractus solitarius (cNTS), and that glucagon-like peptide-1 (GLP-1), an appetite-reducing peptide, also suppressed reflex swallowing via the medial nucleus of the NTS (mNTS). In this study, we examined the mutual interaction between orexin-A and GLP-1 in reflex swallowing. (2) Methods: Sprague-Dawley rats under urethane-chloralose anesthesia were used. Swallowing was induced by electrical stimulation of the superior laryngeal nerve (S

The impact of dihydropyridine derivatives on the cerebral blood flow response to somatosensory stimulation and spreading depolarization.

BACKGROUND AND PURPOSE: A new class of dihydropyridine derivatives, which act as co-inducers of heat shock protein but are devoid of calcium channel antagonist and vasodilator effects, has recently been developed with the purpose of selectively targeting neurodegeneration. Here, we evaluated the action of one of these novel compounds LA1011 on neurovascular coupling in the ischaemic rat cerebral cortex. As a reference, we applied nimodipine, a vasodilator dihydropyridine and well-known calcium channel antagonist. EXPERIMENTAL APPROACH: Rats were treated with LA1011 or nimodipine, either by ch

Phasic activation of the external urethral sphincter increases voiding efficiency in the rat and the cat.

OBJECTIVE: Electrical stimulation of the pudendal nerve (PN) is a potential therapy for bladder dysfunction, but voiding efficiency (VE) produced by PN stimulation appears limited to 60-70%. We conducted experiments in rats and cats to investigate the hypothesis that introduction of artificial phasic bursting activity of the external urethral sphincter (EUS) would enhance VE under conditions where such activity was absent. MATERIALS AND METHODS: Cystometry experiments were conducted in 17 urethane anesthetized female Sprague-Dawley rats and 4 α-chloralose anesthetized male cats. The effects o

Pudendal but not tibial nerve stimulation inhibits bladder contractions induced by stimulation of pontine micturition center in cats.

This study examined the possibility that pudendal nerve stimulation (PNS) or tibial nerve stimulation (TNS) inhibits the excitatory pathway from the pontine micturition center (PMC) to the urinary bladder. In decerebrate cats under α-chloralose anesthesia, electrical stimulation of the PMC (40 Hz frequency, 0.2-ms pulse width, 10-25 s duration) using a microelectrode induced bladder contractions >20 cmH2O amplitude when the bladder was filled to 60-70% capacity. PNS or TNS (5 Hz, 0.2 ms) at two and four times the threshold (2T and 4T) to induce anal or toe twitch was applied to inhibit the PMC

Temporal pattern of stimulation modulates reflex bladder activation by pudendal nerve stimulation.

AIMS: Reflex bladder activation and inhibition by electrical stimulation of pudendal nerve (PN) afferents is a promising approach to restore control of bladder function in persons with lower urinary tract dysfunction caused by disease or injury. The objective of this work was to determine whether bladder activation evoked by pudendal afferent stimulation was dependent on the temporal pattern of stimulation, and whether specific temporal patterns of stimulation produced larger bladder contractions than constant frequency stimulation. METHODS: The mean and maximum contraction pressures evoked b

Effects of intravenous metamizole on ongoing and evoked activity of dura-sensitive thalamic neurons in rats.

Migraine and tension-type headache (TTH) are the most common forms of primary headaches. A general key mechanism underlying development of both the diseases is the trigeminal system activation associated with the ascending nociceptive transmission via the trigemino-thalamo-cortical pathway. The ventroposteromedial (VPM) nucleus is a key thalamic structure, receiving afferent inflow from the craniofacial region; it holds the third-order neurons responsible for conveying sensory information from the extra- and intracranial nociceptors to the cortex. The VPM is currently seen as a therapeutic tar

Thermoregulatory responses elicited by microinjection of L-glutamate and its interaction with thermogenic effects of GABA and prostaglandin E2 in the preoptic area.

The aim of the present study was to investigate the thermoregulatory effects of neuronal activation with sodium L-glutamate (glutamate) in the preoptic area (POA) of the hypothalamus and to examine its possible interaction with the thermogenic effects of GABA and prostaglandin E(2) (PGE(2)). Unilateral microinjection of glutamate (5 nmol) into the lateral POA or its vicinity elicited a prompt increase in tail skin temperature and simultaneous decreases in the O(2) consumption rate (VO(2)), heart rate, and colonic temperature in urethane-chloralose-anesthetized rats. Suppression of bladder overactivity by activation of somatic afferent nerves in the foot.

OBJECTIVE: To investigate the possibility of suppressing bladder overactivity by electrical activation of somatic afferent nerves in the foot. MATERIALS AND METHODS: Cats with an intact spinal cord were studied under α-chloralose anaesthesia. Bladder pressure was recorded via a urethral catheter. Foot stimulation was applied via surface pad electrodes attached to the skin of the front or hind foot. RESULTS: Reflex micturition was inhibited by electrical stimulation of the hind foot at either low (5 Hz) or high (20 Hz) frequencies, but stimulation of the front foot was ineffective. The mean (

Suppression of reflex urethral responses by sacral dermatome stimulation in an acute spinalized feline model.

AIMS: Reflex contractions of the external urethral sphincter (EUS) are a major component of voiding dysfunction after neurological injury or disease. Aberrant urethral reflexes can prevent voiding and cause serious medical complications. Characterizing these urethral reflexes during genitourinary studies is necessary for evaluating novel pharmacological or neuroprosthetic approaches. The objectives of the present study were to generate urethral reflexes in the acute spinal feline, to quantify these reflexes, and to suppress them with electrical stimulation of the sacral dermatomes. METHODS: T

Gating of the micturition reflex by tonic activation of bladder cold receptors in the cat.

AIMS: To determine whether C afferents can modify the gating of the Adelta micturition reflex in order to identify the neuronal site of interaction of the two afferent systems. METHODS: Adult female cats, anaesthetized with alpha-chloralose, had their bladder and urethra catherized through a slit in the proximal urethra. Micturition threshold volume was assessed by cystometry and bladder efferent activity recorded simultaneously. The bladder was filled at a slow rate (1.2-3.5 ml/min) with either body-warm saline (control) or menthol solution (0.06 mM) or by cold saline (4 degrees C). RESULTS

Hypoxic activation of arterial chemoreceptors inhibits sympathetic outflow to brown adipose tissue in rats.

In urethane-chloralose anaesthetized, neuromuscularly blocked, artificially ventilated rats, we demonstrated that activation of carotid chemoreceptors inhibits the elevated levels of brown adipose tissue (BAT) sympathetic nerve activity (SNA) evoked by hypothermia, by microinjection of prostaglandin E2 into the medial preoptic area or by disinhibition of neurones in the raphe pallidus area (RPa). Peripheral chemoreceptor stimulation with systemic administration of NaCN (50 microg in 0.1 ml) or with hypoxic ventilation (8% O2-92% N2, 30 s) completely inhibited BAT SNA.
