Enhancing the value of psychiatric mouse models; differential expression of developmental behavioral and cognitive profiles in four inbred strains of mice.

The behavioral characterization of animal models of psychiatric disorders is often based upon independent traits measured at adult age. To model the neurodevelopmental aspects of psychiatric pathogenesis, we introduce a novel approach for a developmental behavioral analysis in mice. C57BL/6J (C57) mice were used as a reference strain and compared with 129S1/SvImJ (129Sv), BTBR T+tf/J (BTBR) and A/J (AJ) strains as marker strains for aberrant development. Mice were assessed at pre-adolescence (4 weeks), adolescence (6 weeks), early adulthood (8 weeks) and in adulthood (10-12 weeks) on a series

Temporal-Targeted Accelerated 40 Hz tACS Enhances Cognitive Function and Network Integration in Alzheimer's Disease: A Randomized Clinical Trial.

The temporal lobes are key hubs for memory and cognition. Although 40 Hz transcranial alternating current stimulation (tACS) has shown potential cognitive benefits, evidence for accelerated bilateral temporal stimulation remains limited. This study aims to evaluate its cognitive and neural effects. In this randomized, double-blind, controlled trial, 100 Alzheimer's disease (AD) patients were randomized 1:1 to active or sham 40 Hz tACS over the bilateral temporal. Cognitive outcomes at three time points were analyzed using mixed repeated-measures analysis of variance (ANOVA). Resting-state func

High-concentration hydrogen mitigates cognitive impairment in a murine model of Sepsis-associated encephalopathy by enhancing oligodendrocyte maturation and myelination in the mPFC.

Sepsis-associated encephalopathy (SAE) commonly elicits long-lasting cognitive deterioration. Although hydrogen inhalation exerts protective effects in sepsis, its underlying mechanisms remain unclear. Dysregulated oligodendrocyte maturation and myelin architectural defects are pivotal pathological drivers of cognitive deficits across multiple neurological disorders. This study investigates whether high-concentration(67%) hydrogen gas protects against SAE by promoting oligodendrocyte maturation and myelination in the medial prefrontal cortex (mPFC). A CLP-induced mouse SAE model was establishe

Healing together: Results from a pilot trial of 3,4-Methylenedioxymethamphetamine-enhanced cognitive-behavioral conjoint therapy for posttraumatic stress disorder.

Posttraumatic stress disorder (PTSD) disrupts individual well-being and relationship health, particularly among veteran/military couples with heightened risks due to deployment, family reintegration, and traumatic stress. Although cognitive-behavioral conjoint therapy (CBCT) and brief CBCT (bCBCT) have demonstrated strong efficacy in reducing PTSD symptoms, their impact on relational outcomes is less compelling. Given the importance of relationship functioning for veterans' overall health, enhancing interpersonal outcomes of PTSD treatment remains a research priority. One promising approach is

Clostridium butyricum enhances cognitive function in APP/PS1 mice by modulating neuropathology and regulating acetic acid levels in the gut microbiota.

The amyloid deposition-to-cognitive impairment pathway is crucial in Alzheimer's disease (AD) pathogenesis, with gut microbes influencing its development. Our study evaluated Clostridium butyricum MIYAIRI 588 (CBM588) as a potential AD treatment. In APP/PS1 mice, CBM588 improved cognitive function and alleviated colonic tissue pathology by modulating tight junction proteins (upregulating Claudin 1, ZO-1, and Occludin) and reducing inflammatory cytokines (IL-6, IL-1β, and TNF-α mRNA). It also reduced Aβ plaque deposition in the brain, regulated the JNK/CDK5/GSK-3β pathway to inhibit Tau hyperph

Food-borne polystyrene microplastic exposure exacerbates cognitive deficiency via enhanced neuronal synaptic damage and neuroinflammation in Alzheimer's disease.

The impact of polystyrene microplastics (PS-MPs) on the nervous system has been documented, yet the potential role of PS-MPs exposure in exacerbating neuronal damage and neuroinflammation in Alzheimer's disease (AD) remains unclear. Our research demonstrated that oral exposure to PS-MPs caused hippocampal mitochondrial damage in APP/PS1 mice, reduced expression of hippocampal mitochondrial and synapse-associated proteins, inhibited ErbB4 signaling pathway, and damaged hippocampal neurons. Additionally, PS-MPs exposure induced overactivation of astrocytes and microglia with NLRP3 pathway activa

Dual Inhibition of sEH and COX-2 Improved Cognition in Alzheimer's Disease via Enhanced Myogenic Response and Cerebral Artery Distensibility.

Genetic studies have linked EPHX2 (encoding soluble epoxide hydrolase, sEH) and PTGS2 (encoding cyclooxygenase-2, COX-2) to Alzheimer's disease (AD). Elevated levels of sEH and COX-2 found in AD patients and animals suggest their involvement in neurodegeneration, glial activation, vascular dysfunction, and inflammation. This study evaluated the effects of a new dual sEH/COX-2 inhibitor, PTUPB, on cerebrovascular function and cognition in TgF344-AD rats. The rats received oral PTUPB (2 mg/kg/day) for 25 days. Body weight, plasma glucose, and HbA1c levels remained stable between PTUPB- and vehic

SDF-1(5-67) neutralizing antibody enhances hippocampal neurogenesis and rescues cognitive deficits in chronic temporal lobe epilepsy.

Cognitive impairment is a debilitating comorbidity in temporal lobe epilepsy (TLE), linked to suppressed hippocampal neurogenesis in the chronic phase. Building on evidence that links SDF-1 to epilepsy, emerging research, including our own preliminary data, suggests that its cleaved product, SDF-1(5-67), may play a critical role in this process. To investigate the therapeutic potential of SDF-1(5-67) neutralizing antibody (NAb) in a kainic acid (KA)-induced temporal lobe epilepsy (TLE) model. TLE was induced in male Wistar rats by intraventricular KA injection. Animals were divided into acute

IL-17A-mediated copper accumulation participates in chronic neuropathic pain-induced cognitive impairment by enhancing microglial synaptic pruning.

Chronic neuropathic pain is frequently accompanied by cognitive impairment, which seriously influence the quality of the patient's life. The stability of synapse is the basis for maintaining neural circuits. And overactive microglia can prune normal synapses through phagocytosis, leading to cognitive impairment. This study aims to investigate the role of microglial synaptic pruning in chronic neuropathic pain-induced cognitive impairment, and explore the mechanisms of microglial activation through Interleukin-17A (IL-17A) activation and copper accumulation. We found that chronic neuropathic pa

Curcumin-enhanced stem cell exosomes: A novel approach to modulating neuroinflammation and improving cognitive function in a rat model of Alzheimer's disease.

The effect of Curcumin-enhanced stem cell exosomes on the learning and memory impairment induced by streptozotocin (STZ) and neuro-inflammation in rats was evaluated. An animal model of Alzheimer's disease (AD) was established by intracerebroventricular (ICV) injection of STZ (3 mg/kg) in male Wistar rats (250 ± 50 g). ICV STZ injections chronically reduce cerebral glucose uptake and produce other effects similar to pathological, molecular and behavioral features of AD. Numerous studies confirmed the anti-inflammatory and antioxidant properties of curcumin (a natural polyphenol) against free r
