Synthesis of hapten, production of monoclonal antibody, and development of immunoassay for ribavirin detection in chicken.

Ribavirin (RBV) is an effective antiviral drug, whose use is prohibited in animal husbandry worldwide. In this work, a novel immunizing hapten of RBV, named Hapten 4, was designed by comparing the conformational and electronic properties of RBV and haptens based on computational chemistry. Hapten 4 was synthesized and conjugated with carrier proteins to produce monoclonal antibody (mAb). The obtained mAb 4C3 for RBV exhibited an IC50 value of 6.24 ng/ml in an indirect competitive enzyme-linked immunosorbent assay (icELISA) and displayed no cross-reaction with five other antiviral drugs, includ…

Chalcogen-mercury bond formation and disruption in model Rabenstein's reactions: A computational analysis.

Methylmercury is a highly toxic compound and human exposure is mainly related to consumption of polluted fish and seafood. The inactivation of thiol-based enzymes, promoted by the strong affinity binding of electrophilic mercuric ions to thiol and selenol groups of proteins, is likely an important factor explaining its toxicity. A key role is played by the chemistry and reactivity of the mercury-chalcogens bond, particularly HgS and HgSe, which is the focus of this computational work (level of theory: (COSMO)-ZORA-BLYP-D3(BJ)/TZ2P). We analyze nine ligand-exchange model reactions (the so-cal

Leaving Group Effects in a Series of Electrosprayed CcHhN1 Anthracene Derivatives.

We investigate the gas-phase structures and fragmentation pathways of model compounds of anthracene derivatives of the general formula CcHhN1 utilizing tandem mass spectrometry and computational methods. We vary the substituent alkyl chain length, composition, and degree of branching. We find substantial experimental and theoretical differences between the linear and branched congeners in terms of fragmentation thresholds, available pathways, and distribution of products. Our calculations predict that the linear substituents initially isomerize to form lower energy branched isomers prior to lo

Monodentate phosphine substitution in [Pd(κ3-dppf)(PR3)][BF4]2 (dppf = 1,1'-bis(diphenylphosphino)ferrocene) compounds.

The ligand 1,1'-bis(diphenylphosphino)ferrocene (dppf) is commonly employed in a variety of catalytic systems. There are a variety of coordination modes known for dppf, the least studied being the κ3 coordination mode, in which both phosphorus atoms and the iron atom of dppf interact with another metal center. One such compound is the previously reported [Pd(κ3-dppf)(PPh3)]2+. A series of related compounds, [Pd(κ3-dppf)(P(p-C6H4R)3)]2+ (R = OCH3, CH3, F and CF3), has been synthesized and characterized. The X-ray crystal structure of [Pd(dppf)(P(p-C6H4F)3)][BF4]2 was determined. Electrochemical

Structure and evaluation of antibacterial and antitubercular properties of new basic and heterocyclic 3-formylrifamycin SV derivatives obtained via 'click chemistry' approach.

Thirty four novel derivatives of 3-formylrifamycin SV were synthesized via reductive alkylation and copper(I)-catalysed azide-alkyne cycloaddition. According to the obtained results, 'click chemistry' can be successfully applied for modification of structurally complex antibiotics such as rifamycins, with the formation of desired 1,2,3-triazole products. However, when azide-alkyne cycloaddition on 3-formylrifamycin SV derivatives demanded higher amount of catalyst, lower temperature and longer reaction time because of the high volatility of substrates, an unexpected intramolecular condensation

Structure-based design of a heptavalent anthrax toxin inhibitor.

The design of polyvalent molecules, consisting of multiple copies of a biospecific ligand attached to a suitable scaffold, represents a promising approach to inhibit pathogens and oligomeric microbial toxins. Despite the increasing interest in structure-based drug design, few polyvalent inhibitors based on this approach have shown efficacy in vivo. Here we demonstrate the structure-based design of potent biospecific heptavalent inhibitors of anthrax lethal toxin. Specifically, we illustrate the ability to design potent polyvalent ligands by matching the pattern of binding sites on the biologic

Fast and accurate simulation of Raman spectra of gold-organic systems.

Resolving the spectral Raman signature of molecules grafted onto a metallic support is often a difficult task, in which quantum chemistry methods allow for valuable additional rationalization and signal attributions, especially to probe the formation of a bond with the support. In the specific case of gold-organic architectures based on a Au-C bond, only a limited amount of experimental and theoretical reference data is available in the literature, and Raman simulations based on quantum mechanics quickly become unaffordable with the size of the system. In this work, we evaluate the precision o

On the Electronic Nature of Open-Shell Germylene Complexes of MI Cations (M = Fe, Co, Ni).

While the coordination chemistry of heavier tetrylene ligands is generally well established, their effect on the electronic nature of open-shell metal centers is underexplored. This work takes steps to define these effects in an analogous series of cationic germylene-MI complexes (M═Fe, Co, Ni). First, the electronic ground-states of toluene-coordinated MI synthons, [IPr·M(η6-tol)]+ (IPr = [{(H)CN(Dip)}2C:]; Dip = 2,6-C6H3) are elucidated using CAS-SCF calculations, defining a rare non-Aufbau for the Fe(I) derivative. Following this, a combination of experimental and computational investigatio

Investigation of Acoramidis-based ionic liquids as potential stabilizers of transthyretin using DFT calculations, molecular docking, and ADMET studies.

Transthyretin amyloidosis (ATTR) is a type of amyloidosis that characterize by abnormal deposition of the transthyretin protein in various organs. Acoramidis drug is considered a promising treatment for patients with ATTR. However, oral delivery of the insoluble Acoramidis is challenging due to its poor bioavailability and low solubility. In the present study, three Acoramidis-based ionic liquids (ILs) are reported to address this challenge. The biological activity and reactivity of these compounds evaluated using computational chemistry methods, including molecular DFT calculations, molecular…

Predicting the Reactivity of Acyclic Silylenes and Germylenes in Hydrogen Activation Using Machine Learning.

Heavier carbenes can mimic transition metals in activating small molecules and strong bonds. Incorporating this ability into catalytic cycles demands careful, experimentally challenging tuning of their molecular properties via substituent variation. This study employs computational chemistry and data science to investigate dihydrogen activation by heavier carbenes to aid future candidate selection. Analyzing around 600 acyclic silylenes and germylenes derived from 40 different substituents, we found that a simple linear model predicts the activation barrier and reaction energy of H2 activation…

Multi-Step Oxygen Redox Mechanism in the Polyanionic Lithium-Rich Cathode Li2FeSiO4.

Lithium-rich cathode materials offer significantly higher energy densities than conventional cathode materials, due to their ability to store charge beyond the formal transition-metal redox limit, via so-called "oxygen redox". In lithium-rich oxides, oxygen redox is accompanied by molecular O2 formation and transition-metal migration, associated with irreversible structural change and poor cycling stability. Lithium-rich silicates provide an alternative lithium-rich chemistry, and have been proposed to be resilient to O2 formation at the top of charge, due to their covalently bonded [SiO4]4- f

Multidimensional Protein Corona Analysis Toward Predictive Nano-Bio Interface Design.

Nanoparticles entering biological fluids are rapidly coated by proteins and other biomolecules, converting their synthetic surfaces into biologically active nano-bio interfaces. These coronas regulate colloidal stability, immune recognition, cellular uptake, biodistribution, pharmacokinetics, cargo delivery, and toxicity. Yet a protein list obtained by mass spectrometry captures only part of this interface. Corona identity and function are also shaped by protein organization, binding stability, exchange dynamics, conformational changes, and molecular accessibility. Here, we discuss recent prog

Purine-centred fused, conformationally constrained, and ring-expanded analogues in antitumor and antiviral drug discovery: A comprehensive review.

Purine-derived scaffolds remain important in anticancer and antiviral drug discovery because their electronic properties, hydrogen-bonding capacity, and conformational adaptability support interactions with kinases, polymerases, nucleotide-processing enzymes, and nucleic acids. This review critically examines three major purine-centred classes: fused tricyclic purines, conformationally constrained purine nucleosides bearing bridged sugar or carbocyclic systems, and ring-expanded purines. Relevant publications from 1986 to 2026 were evaluated with emphasis on scaffold design, synthetic strategi

Organosulfur scaffolds as quorum sensing and biofilm modulators in Gram-negative bacteria.

Quorum sensing (QS) is a cell-density-dependent communication system that coordinates bacterial virulence, biofilm formation, motility, and stress adaptation, making it a compelling target for antivirulence intervention. Due to the possibility of sulfonyl (-SO₂-) and sulfinyl (-S = O-) functional groups to impart specific electronic and steric properties that can modulate bacterial signaling and receptor interactions, organosulfur compounds are increasingly gaining attention. This review highlights recent advances in the development of sulfonyl- and sulfinyl-containing organosulfur compounds a

Engineering Altermagnetic States in Two-Dimensional Square Tessellations.

Altermagnetism, an emergent collinear magnetic phase with zero net magnetization and momentum-dependent spin splitting, promises to revolutionize spintronics by leveraging symmetry-driven effects without requiring spin-orbit coupling. Despite its potential, a comprehensive understanding of design principles and spin-splitting mechanisms remains elusive. Here, from a mathematical perspective, we exploit the intrinsic fourfold symmetry and structural versatility of 2D square tessellations to engineer altermagnetic states. By systematically screening the altermagnetic state in all 34 2D square te

The intramolecular stabilizing effects of O-benzoyl substituents as a driving force of the acid-promoted pyranoside-into-furanoside rearrangement.

Furanoside derivatives are broadly present in the antigenic structures of pathogenic microorganisms and play a key role in their recognition by the host immune system. Despite the high demand for vaccine and diagnostic development, their chemical synthesis remains challenging. During the development of a new methodology for the synthesis of galactofuranoside building blocks, we encountered an unexpected predominance of the furanoside form in the equilibrium mixture of benzoylated β-galactosides. Since the furanoside form is typically less stable and is usually present only in minor amounts, we

Computational and Experimental Study of the Conformational Variation of the Catalytic Residue His41 of the SARS-CoV-2 Main Protease.

The main protease (Mpro) is essential for the replication of SARS-CoV-2, making it one of the major therapeutic targets for COVID-19 treatment. Here, we explored the conformational dynamics and energetics of the catalytic residue His41 in Mpro, as revealed by a rare conformational shift observed in the cocrystal structures of Mpro bound by certain inhibitors. Using steered molecular dynamics combined with umbrella sampling, we demonstrated that π-cation interactions between these inhibitors and the ionized catalytic dyad significantly reduced the energy barrier for the conformational flip of t
