In the evolving landscape of cancer treatment, the strategic manipulation of regulated cell death (RCD) pathways has emerged as a crucial co … h modalities. We then discuss how these processes activate antigen-presenting cells, facilitate the cross-priming of CD8+ T cells, and trigger anti-tumor immune responses, highlighting the complex effects of novel forms of tumor cell death on TME and tumor biology. Furthermore, we summarize potential drugs and nanoparticles that can induce or inhibit these emer …

The Platform trial In COVID-19 priming and BOOsting (PICOBOO) is a multi-site, adaptive platform trial designed to generate evidence of the immunogenicity, reactogenicity, and cross-protection of different booster vaccination strategies against severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) and its variants, specific for the Australian context. The PICOBOO trial randomises participants to receive one of three COVID-19 booster vaccine brands (Pfizer, Moderna, Novavax) available for use in Australia, where the vaccine brand subtypes vary over time according to the national vaccine

The increased tropism for malignant cells of some viruses has been highlighted in recent studies, prompting their use as a strategy to modify the transcriptional profile of those cells, while sparing the healthy ones. Likewise, they have been recognized as players modulating microenvironmental immunity, namely through an increase in antigen-presenting, natural-killer, and T CD8+ cytotoxic cells by a cross-priming mechanism elicited by tumor-associated antigens. The immunomodulatory role of the oncolytic virus seems relevant in hematological malignancies, which may relapse as a result of a prol…

Hepatic metastases are a poor prognostic factor of colorectal carcinoma (CRC) and new strategies to reduce the risk of liver CRC colonizatio … ffer cells (KCs) and liver capsular macrophages (LCMs). Moreover, IFNα endowed LSECs with efficient cross-priming potential that, along with the early intravascular tumor burden reduction, supported the generation of antitumor CD8+ T cells and ultimately led to the establishment of a protective long-term memory T cell response. These findings provide a rational …

Childhood exposure to traumatic events, termed early life stress (ELS), is now widely recognized for causing long-term negative health effects that may not manifest until adulthood. Allostatic load (AL) describes the cumulative "wear-and-tear" effects of chronic stress on the body that may adversely affect human health by accelerating other disease processes. Recent epidemiological studies have reported higher stress levels in industrialized countries and trends of increasing prevalence in autoimmune diseases during recent decades. To elucidate mechanisms of stress-related immune dysregulation

Human T-cell responses to the stress protein alpha B-crystallin in multiple sclerosis (MS)-affected brain samples are dominant when compared to other myelin antigens. The establishment of the apparent autoimmune repertoire against this antigen has been suggested to involve cross-priming during viral infection. Yet, another possibility would be that determinant spreading during ocular inflammation could generate a response to alpha B-crystallin, since it is also a major component of the eye. In this study, we compared serum IgG, IgA and IgM repertoires against a range of eye lens-derived ocular…

The effect of a corticosteroid drug on type I reactions in the nose was studied in eighteen hay fever patients. They were treated with a daily dosage of 800 microng beclomethasone dipropionate intranasally for I week, in a double-blind, placebo-controlled cross-over trial. Allergen challenge on the last day of each treatment period showed that steroid treatment diminished sneezing and rhinorrhoea, but the effect was not statistically significant (P = 0-10). Immediate nasal blockage was unaffected. There is a marked discrepancy between the minor protection offered by beclomethasone dipropionate

Mechanical high-intensity focused ultrasound releases tumor antigens and danger signals. It provides a potential substrate for immune checkpoint blockade. However, antigen release does not necessarily generate productive dendritic cell priming. In colorectal cancer, this transition occurs within a mucosal microenvironment shaped by epithelial barrier integrity, microbial exposure and myeloid clearance. Neutrophils can support epithelial repair or reinforce tumor-associated suppression. Macrophages remove apoptotic granulocytes and regulate inflammatory resolution. They can also restrain dendri

Cancer nanovaccines offer great promise for eliciting antitumor immune responses, yet challenges persist in achieving efficient antigen delivery, robust immune activation, and formulation simplicity suitable for practical application. Here, we present a generalizable vaccine design strategy based on protein cross-linking combined with innate immune stimulation via the STING agonist MSA-2. This approach enables the construction of self-adjuvanted protein nanovaccines that unify tumor antigen stabilization, lymph node delivery, and immune activation within a single nanosystem. The representative

Protein corona formation is widely viewed as an inevitable but poorly controlled event when nanoparticles encounter biological environments. Converting this interfacial layer into a programmable functional entityremains a major challenge. Here, we report an adaptive protein corona nanoassembly strategy that integrates cytokine signaling with endogenous antigen transport. Coordination-engineered manganese nanoscaffolds were assembled with interleukin-12 (IL-12) and protected by an enzyme-responsive hyaluronic acid shell to form nanoshuttles (NSs) capable of dynamic protein recruitment in the tu

Oncolytic peptides (OLPs) constitute an emerging class of immunotherapeutics that combine direct cancer cell lysis with immune activation. Indeed, OLPs induce rapid immunogenic cell death by disrupting intracellular membranes, which culminates with the abundant release of malignant cell contents including immunostimulatory danger-associated molecular patterns. This results in the engagement of innate immune sensors that potently activate dendritic cells, leading to the cross-priming of tumor-specific T lymphocytes that mediate systemic anticancer immunity. Through such a dual mechanism, OLPs c…

Ambient ozone (O3) has been reported to be associated with influenza activity, but evidence remains inconsistent, particularly in subtropical settings. Here, we investigated this question in subtropical Hong Kong using weekly and daily data from 2005 to 2016 through a multi-method triangulation methodology integrating convergent cross mapping (CCM) from dynamical systems theory, Peter-Clark momentary conditional independence (PCMCI) algorithm from graphical causal modeling, and regression-based generalized linear model (GLM). We found a consistent inhibitory relationship of O3 with influenza a

The S2 subunit of the severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) spike is highly conserved across coronavirus strains and therefore is a potential pan-coronavirus vaccine target. However, antibodies targeting this region are typically non-neutralizing. We report herein that S2-targeting antibodies from patients who recovered from SARS-CoV-2 infection bound only closely related sarbecovirus subgenus strains and, like most known S2 antibodies, none of these were neutralizing. In contrast, first-exposure, severe acutely infected COVID-19 patients predominantly induced back-boost

INTRODUCTION: In patients with cirrhosis and ascites, failure of intestinal barriers and cirrhosis-associated immune dysfunction contribute to the translocation of bacteria and microbial products to the peritoneal cavity, which promotes infection, perpetuates inflammation and accelerates acute-on-chronic liver failure. Resident peritoneal immune cells are repeatedly exposed to bacterial products, and their activation status is linked to complications of spontaneous bacterial peritonitis.
AREAS COVERED: This narrative review summarizes the recent research on human peritoneal immunity in decomp

Boosting antitumor immunity has emerged as a powerful strategy in cancer treatment. While releasing T-cell brakes has received most attention, tumor recognition by T cells is a pre-requisite. Radiotherapy and certain cytotoxic drugs induce the release of damage-associated molecular patterns, which promote tumor antigen cross-presentation and T-cell priming. Antibodies against the "do not eat me" signal CD47 cause macrophage phagocytosis of live tumor cells and drive the emergence of antitumor T cells. Here we show that CXCR4 activation, so far associated only with tumor progression and metasta

Chronic diabetic glucolipotoxicity is associated with progressive skeletal muscle atrophy, bone loss, cellular senescence, and ferroptotic injury. Because ferroptosis inhibition alone may not restore compromised myogenic capacity, we developed a combined stem cell-conditioning strategy integrating fullerol priming with the stem cell screen-enrich-combine(-biomaterials) circulating system (SECCS). This integrated three-dimensional culture platform uses porous β-TCP granules and intermittent medium circulation to generate BMSCs-derived extracellular vesicles (EVs). Among 2D-EVs, F-2D-EVs, SECCS-

The outbreak of coronavirus disease 19 (COVID-19) has highlighted the demand for vaccines that are safe and effective in inducing systemic and airway mucosal immunity against the aerosol transmission of severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2). In this study, we developed a novel helper-dependent adenoviral vector-based COVID-19 mucosal vaccine encoding a full-length SARS-CoV-2 spike protein (HD-Ad-FS). Through intranasal immunization (single-dose and prime-boost regimens), we demonstrated that the HD-Ad-FS was immunogenic and elicited potent systemic and airway mucosal pro

BACKGROUND: Recently, it has been proposed that psychosocial concerns may motivate the demand for aesthetic rhinoplasty. Although successful operations often improve the quality of life and self-esteem symptoms in patients with sound mental health, they may actually result in unsatisfactory outcomes in those patients with significant depression, anxiety, or other severe psychological disorders. The purpose of this study was to assess the incidence of psychological disorders in patients seeking rhinoplasty.
METHODS: A prospective cross-sectional study of 298 random volunteers was conducted, wi

Microglial priming is caused by aging and neurodegenerative diseases, and is characterized by an exaggerated microglial inflammatory response to secondary and sub-threshold challenges. In the present study, we examined the effects of the matrix metalloproteinase-8 (MMP-8) inhibitor (M8I) on the brain of aged normal and leucine-rich repeat kinase 2 (LRRK2) G2019S Parkinson's disease (PD) model mice systemically stimulated with lipopolysaccharide (LPS). The results indicated that Iba-1 positive microglia and GFAP-positive astrocytes, which were increased by LPS, significantly decreased by M8I in
