It was demonstrated that biogenic elicitors, arachidonic acid and chitosan, locally and systemically stimulated wound healing in potato tuber tissues by increasing the number of wound periderm layers, accelerating the development of cork cambium (phellogen), and inducing proteinase inhibitors. The signal molecules, jasmonic and salicylic acids, had different effects on the development of wound periderm: jasmonic acid locally and systemically stimulated potato wound healing and elevated the level of proteinase inhibitors, whereas salicylic acid did not have any effect on wound healing and even

Groundwater contamination by carbon tetrachloride (CCl4) presents a health risk as a potential carcinogen and pollutant that is capable of depleting the ozone layer. Although use of poplar trees in a phytoremediation capacity has proven to be cost effective for cleaning contaminated sites, minimizing leaf emission of volatile contaminants remains a pressing issue. We hypothesized that recently fixed carbon plays a key role in CCl4 metabolism in planta yielding nonvolatile trichloroacetic acid (TCA) and that the extent of this metabolism can be altered by heightening plant defenses. The Arabidopsis enhanced disease susceptibility 4 (eds4) mutation causes enhanced susceptibility to infection by the bacterial pathogen Pseudomonas syringae pv. maculicola ES4326 (Psm ES4326). Gene-for-gene resistance to bacteria carrying the avirulence gene avrRpt2 is not significantly affected by eds4. Plants homozygous for eds4 exhibit reduced expression of the pathogenesis-related gene PR-1 after infection by Psm ES4326, weakened responses to treatment with the signal molecule salicylic acid (SA), impairment of the systemic acquired resistance response, and reduced accumulation of SA after

A fungal metabolite brefeldin A (BFA) was used to study virus-host interaction in glycoprotein processing in West Nile virus-infected Vero and C6/36 cells. The results indicated that as little as 1 microgram/ml of BFA resulted in complete breakdown in the Golgi organelle in infected Vero cells. This led to modifications of the glycoproteins which could not be efficiently used in infectious virion formation. In contrast, as much as 10 micrograms/ml of BFA in culture medium did not affect either glycoprotein formation or production of infectious particles in C6/36 cells. The results showed that

Three commonly used antitussive compounds were tested for their ability to block epileptiform activity recorded extracellularly from hippocampal and olfactory cortex slices maintained in vitro. Antitussives were bath-applied to brain slices either before or after epileptiform activity was induced. Dextromethorphan (DM) prevented electrically evoked epileptiform afterdischarges and arrested spontaneous bursting induced by exposure to added NMDA or to Mg2(+)-free medium. In contrast, caramiphen (CM) and carbetapentane (CB) were effective against epileptiform activity induced by Mg2(+)-free mediu

The effect of the non-opioid antitussive caramiphen was studied in the rat maximal electroshock test. Caramiphen produced a dose- and time-dependent blockade of tonic hindlimb extension and was nearly twice as potent as the prototypical anticonvulsant drug diphenylhydantoin. Pretreatment with a subthreshold-effective dose of caramiphen potentiated the anticonvulsant effects of diphenylhydantoin, lowering its ED50 33-fold. The anticonvulsant effects of caramiphen were not associated with its cholinolytic activity since (a) its anticonvulsant effects were not antagonized by physostigmine and (b)

Sugarcane (Saccharum spp.) is a major sugar crop, yet its growth and yield are frequently limited by soil salinization. Receptor-like cytoplasmic kinases (RLCKs) act as central signaling hubs in plants, regulating growth, development, immunity, and abiotic stress responses. However, the evolution of the RLCK gene family in sugarcane and its role in salt tolerance remain unclear. Using the high-quality genome of elite cultivar 'Zhongzhe No.1', we identified 262 ScRLCK genes, grouped into 15 subfamilies, all of which contain a typical protein kinase domain. Synteny analysis revealed that whole-g

Plant oxylipins are a diverse group of oxygenated fatty acid derivatives that function as important signaling molecules in plant responses to abiotic stress. Although jasmonates, particularly jasmonic acid (JA) and its derivatives, have been extensively studied, increasing evidence demonstrates that other oxylipin classes, including 12-oxophytodienoic acid (OPDA), green leaf volatiles (GLVs), reactive electrophilic oxylipins (RES), and peroxygenase (PXG)-derived oxylipins, also contribute to stress adaptation. This review summarizes current understanding of the molecular mechanisms underlying

Insect herbivory triggers rapid local and systemic signaling in plants, including membrane depolarization, cytosolic Ca2+ elevation, and activation of jasmonate-mediated defense pathways. These early events are shaped by calcium-permeable ion channels and regulatory components that generate stimulus-specific Ca2+ signatures. The glutamate receptor-like channel-centric model of long-distance Ca2+ signaling is insufficient to explain the robustness of systemic responses to diverse damage and herbivore cues, indicating the involvement of additional Ca2+ channels. Here, we identify CYCLIC NUCLEOTI

Rice (Oryza sativa), a staple cereal crop, is significantly threatened by rising temperatures, which impact all stages of growth, including the crucial early seedling establishment phase. Despite being critical in determining overall growth and productivity, response to acute (short-term intense) heat stress during the early seedling stage remains understudied. Here, by using gradients of heat stress duration and intensity, we identified a critical threshold, beyond which acute heat stress significantly impedes seedling growth. Transcriptome analysis of seedlings exposed to growth-inhibiting t

Climate models predict that episodes of extreme heat will intensify in both frequency and severity in the upcoming decades, significantly impacting plant growth, productivity, and survival. In addition, plants in natural environments are often exposed to many other stress factors simultaneously, triggering responses even more complex and difficult to predict. Understanding how plants integrate multiple abiotic stress signals is essential for improving resilience to increasingly complex environmental conditions. In this study, we investigated how heat stress (HS) modulates hormonal signaling ne

CgMYC2 links jasmonate signaling to naringin biosynthesis by binding G-box motifs and activating flavonoid-pathway promoters in Citrus grandis 'Tomentosa' Naringin, the predominant bitter-flavanone glycoside in Citrus grandis 'Tomentosa', has well-characterized biosynthetic enzymes, yet the transcriptional regulators coupling hormonal signals to pathway activation remain poorly understood. We demonstrate that CgMYC2, a jasmonate-responsive bHLH transcription factor, functions as a central activator of naringin biosynthesis. Exogenous methyl jasmonate (MeJA) treatment increased naringin content

Inflammation is a key factor leading to secondary brain injury after intracerebral hemorrhage (ICH). Neddylation, a post-translational modification that attaches NEDD8 (neuronal precursor cell-expressed developmentally downregulated protein 8) to targets, regulates diverse cellular processes. Neddylation is overactivated in various cancers and linked to atherosclerosis and inflammatory disorders, but its role in ICH remains unclear. In this study, mice were subjected to ICH by injection of collagenase. The NEDD8-activating enzyme inhibitor MLN4924 was administered subcutaneously post-ICH. We f

Plant-parasitic nematodes deploy secreted effector proteins that reprogram host cellular processes to establish parasitism. Here, we characterized Mi1D08B, a putative dorsal-gland effector from the root-knot nematode Meloidogyne incognita and defined its role in subverting soybean jasmonate-mediated immunity. Mi1D08B is conserved across several Meloidogyne species and localizes to the plant nucleus and cytoplasm. Overexpression of Mi1D08B significantly increased galling and egg production, demonstrating a strong virulence role. A gall-specific yeast two-hybrid screen identified the NINJA-famil

Delaying plant aging and death and enhancing immune defenses of plant have beneficial effects on crop production and the continued development of the agricultural economy. In this study, we focused on AtALMT9, which is located in the vacuolar membrane. Specifically, we investigated its functions in Arabidopsis thaliana leaf development and disease resistance. Our physiological analysis revealed that AtALMT9 negatively regulates leaf development and the immune response to pathogenic microorganisms. Moreover, RNA-seq and qRT-PCR analyses indicated leaf aging- and immunity-related genes were sign

BACKGROUND: Mediterranean diet may enhance cognitive function and delay the progression of Alzheimer's disease (AD). We conducted a systematic review to investigate the effect of oleocanthal (OC) from extra-virgin olive oil (EVOO) on amyloid-β (Aβ) burden in preclinical models of AD, considering the anti-inflammatory and neuroprotective effects of EVOO biophenols, which are key components of the Mediterranean dietary model.
METHODS: The literature was searched through six electronic databases until February 2023. Screening of 52 retrieved articles for inclusion criteria resulted in 7 preclini

Phosphate (Pi) is a critical nutrient for plants and is often a limiting factor in food production, as many agricultural soils are limited in available Pi. Inositol pyrophosphates (PP-InsPs) are signaling molecules involved in Pi sensing and jasmonic acid (JA)-regulated plant defense. Here, we report that overexpression of 1,3,4-trisphosphate 5/6-kinase 1 (ITPK1) and the kinase domain of the dual-domain diphosphoinositol pentakisphosphate kinase 2 (VIP2KD) in Arabidopsis thaliana results in unique elevations in PP-InsPs, accompanied by altered leaf growth and senescence patterns, as well as de

Nerve agents are lethal chemical weapons and highly potent chemoconvulsants. For the treatment of nerve agent-induced status epilepticus (SE), the FDA has approved the use of benzodiazepines-initially diazepam and recently midazolam (MDZ); however, benzodiazepines are not neuroprotective, particularly if not administered promptly. Here, we compared the antiseizure and neuroprotective efficacy of MDZ with that of tezampanel (LY293558; an AMPA/GluK1 receptor antagonist) administered with caramiphen (CRM; an antagonist of muscarinic and NMDA receptors), 30 min after the onset of SE induced by exp

New antimicrobial molecules effective against Pseudomonas aeruginosa, known as an antibiotic-resistant "high-priority pathogen", are urgently required because of its ability to develop biofilms related to healthcare-acquired infections. In this study, for the first time, the anti-biofilm and anti-virulence activities of a polyphenolic extract of extra-virgin olive oil as well as purified oleocanthal and oleacein, toward P. aeruginosa clinical isolates were investigated. The main result of our study was the anti-virulence activity of the mixture of oleacein and oleocanthal toward multidrug-resi

Plants have developed various resistance mechanisms against herbivorous insects through prolonged coevolution. Plant defence responses can be triggered by specific compounds present in insect saliva. Apyrase, a known enzyme that catalyzes the hydrolysis of adenosine triphosphate (ATP) and adenosine diphosphate (ADP) into adenosine monophosphate (AMP) and inorganic phosphorus, has recently been identified in some herbivorous insects. However, whether insect salivary apyrase induces or inhibits plant responses remains poorly understood. In this study, we identified an apyrase-like protein in the
