Epigenetic mechanisms governing cell type specific somatic expansion and toxicity in Huntington's disease.

Huntington's disease (HD) is characterized by neuronal dysfunction and degeneration that varies markedly by brain region and cell type. We previously showed that CAG repeat expansion in exon 1 of the mHTT gene correlates with increased expression of the mismatch repair genes MSH2 and MSH3 in striatal medium spiny neurons1, and demonstrated that, in the striatum and cerebral cortex of individuals with HD, hundreds of genes are dysregulated in neuronal cell types carrying somatically expanded CAG repeat in mHTT 1,2. Here we employ comprehensive epigenetic profiling in specific neuronal and glial

Mechanisms associated with temozolomide resistance in U87MG cell line: in silico and in vitro approaches.

IDH-wildtype grade diffuse glioma is the most aggressive primary brain tumor in adults, with limited survival despite standard therapy with temozolomide (TMZ). Resistance to TMZ is driven by DNA repair pathways, tumor heterogeneity, and microenvironmental adaptations, indicating the need for novel therapeutic strategies. Using a systems biology approach, a compound-protein and protein-protein interaction networks was constructed with STRING 12.0 and STITCH 5.0 databases, identifying hub-bottleneck proteins associated with TMZ resistance. Topological and gene ontology analyses revealed 5 functi

Nucleases and Their Inhibitors: Exploring Biological Roles, Industrial Applications, and Challenges in Heterologous Expression.

Nucleases hydrolyze phosphodiester bonds and participate in numerous cellular and metabolic processes. Intracellular nucleases repair nonfunctional or damaged DNA using DNA base excision repair (BER), mismatch repair (MMR), and homologous recombination (HR). Apoptotic nucleases systematically degrade cellular DNA during programmed cell death (PCD). Non-apoptotic nucleases support DNA repair and replication. Small noncoding RNAs (sncRNAs) degrade the RNA of viral particles. Extracellular and membrane-associated nucleases replenish nucleotides, especially in biofilms where cells rely on addition

The ATF2-KNSTRN Axis Harnesses Lactate to Modulate MLH1/MSH2 Expression and Gemcitabine Resistance in Lung Adenocarcinoma.

Gemcitabine (GEM) is a cornerstone of chemotherapy for lung adenocarcinoma (LUAD). However, acquired resistance severely limits its clinical efficacy, leading to relapse and metastasis. This study aims to investigate the role of KNSTRN in GEM resistance and its underlying molecular mechanisms in LUAD. Bioinformatics analysis revealed the expression levels of KNSTRN in LUAD tissues and the enriched pathways, and analyzed the prognostic impact of KNSTRN in LUAD, predicting the upstream transcription factor (TF) ATF2. The study examined the correlation and binding sites between KNSTRN and ATF2, a

Mismatch repair genes Mlh1 and Mlh3 modify CAG instability in Huntington's disease mice: genome-wide and candidate approaches.

The Huntington's disease gene (HTT) CAG repeat mutation undergoes somatic expansion that correlates with pathogenesis. Modifiers of somatic expansion may therefore provide routes for therapies targeting the underlying mutation, an approach that is likely applicable to other trinucleotide repeat diseases. Huntington's disease Hdh(Q111) mice exhibit higher levels of somatic HTT CAG expansion on a C57BL/6 genetic background (B6.Hdh(Q111) ) than on a 129 background (129.Hdh(Q111) ). Linkage mapping in (B6x129).Hdh(Q111) F2 intercross animals identified a single quantitative trait locus underlying

Substrate spectrum of human excinuclease: repair of abasic sites, methylated bases, mismatches, and bulky adducts.

Nucleotide-excision repair is the repair system for removing bulky lesions from DNA. Humans deficient in this repair pathway suffer from xeroderma pigmentosum (XP), a disease characterized by photodermatoses, including skin cancers. At the cellular level, XP patients fail to remove cyclobutane pyrimidine dimers and pyrimidine(6-4)pyrimidone photoproducts induced by UV light, as well as other bulky DNA lesions caused by various genotoxic agents. XP cells are not particularly sensitive to ionizing radiation or to alkylating agents that cause mostly nonbulky DNA lesions. Therefore, it has general

Sarcomas associated with hereditary nonpolyposis colorectal cancer: broad anatomical and morphological spectrum.

Hereditary nonpolyposis colorectal cancer (HNPCC) is primarily linked to colorectal and endometrial cancer, but is associated with a broad tumor spectrum. Though not formally part of the syndrome, occasional sarcomas have been reported in individuals with HNPCC. We used the national Danish HNPCC-register to identify HNPCC families in which sarcomas had been diagnosed. Fourteen sarcomas were identified in families with mutations in MSH2, MSH6, and MLH1. The median age at sarcoma diagnosis was 43 (15-74) years. Soft tissue sarcomas predominated followed by uterine sarcomas and eight histopatholo

Familial and hereditary non-polyposis colorectal cancer: issues relevant for surgical practice.

About 15% of patients with colorectal cancer report a family history of this disease. An estimated 1%-5% of patients have hereditary non-polyposis colorectal cancer (HNPCC). Recently, DNA mismatch repair genes associated with this syndrome were identified. For about 50% of families in which HNPCC occurs, DNA-based diagnosis and presymptomatic DNA testing are now feasible. Diagnosis of a hereditary tumour syndrome is relevant for both the patient with cancer and his or her close relatives. The complexities of family studies warrant the forming of a multidisciplinary team which may choose to wor…

Highly efficient base editing at PCSK9 and normal human embryo development.

Cas9-based tools enable programmable DNA lesions for studying repair outcomes, gene function, and genome correction. In human embryos, Cas9-induced DNA double-strand breaks are genotoxic, causing frequent aneuploidy and large deletions1,2. Here, we evaluate DNA repair outcomes at nicks and mismatches introduced by base editors at the PCSK9 and HBG loci in human embryos. Delivering ABE8e-V106W as a protein at fertilization achieved editing at all PCSK9 alleles, supporting development to the blastocyst stage and the derivation of homozygous edited stem cell lines. No insertions or deletions were

hSSB2 (NABP1) is required for the recruitment of RPA during the cellular response to DNA UV damage.

Maintenance of genomic stability is critical to prevent diseases such as cancer. As such, eukaryotic cells have multiple pathways to efficiently detect, signal and repair DNA damage. One common form of exogenous DNA damage comes from ultraviolet B (UVB) radiation. UVB generates cyclobutane pyrimidine dimers (CPD) that must be rapidly detected and repaired to maintain the genetic code. The nucleotide excision repair (NER) pathway is the main repair system for this type of DNA damage. Here, we determined the role of the human Single-Stranded DNA Binding protein 2, hSSB2, in the response to UVB e

Family history and molecular features of children, adolescents, and young adults with colorectal carcinoma.

BACKGROUND: Colorectal cancer is extremely rare in childhood. Published case series reporting children and adolescents with colorectal cancer have not focused on the underlying genetic aspects of the tumour or genetic susceptibility of the families. AIMS: We examined a cohort of patients with early onset colorectal cancer to determine whether a specific genetic predisposition could be elucidated. In particular, we focused on whether DNA mismatch repair gene deficiency which causes hereditary non-polyposis colorectal cancer (HNPCC) could be elucidated. METHODS: Patients with colorectal cancer…

Bioprospecting potential genetic biomarkers of gallbladder cancer.

BACKGROUND: Gallbladder cancer (GBC) is a rare and aggressive cancer of the biliary tract with a very low survival rate. The availability of diagnostic biomarkers and targeted therapies for its management is limited. The study identifies potential genetic biomarkers of GBC by analyzing differentially expressed genes (DEGs) through microarray profiling and constructing regulatory networks using systems biology techniques. METHODS: We used Clariom™ D Array in gallbladder cancer, cholelithiasis, and normal tissues (10 cases in each group), identifying DEGs and key biological pathways. Functional

Intersection of the fragile X-related disorders and the DNA damage response.

The Repeat Expansion Diseases (REDs) are a large group of human genetic disorders that result from an increase in the number of repeats in a disease-specific tandem repeat or microsatellite. Emerging evidence suggests that the repeats trigger an error-prone form of DNA repair that causes the expansion mutation by exploiting a limitation in normal mismatch repair. Furthermore, while much remains to be understood about how the mutation causes pathology in different diseases in this group, there is evidence to suggest that some of the downstream consequences of repeat expansion trigger the DNA da

Photoactivation of DNA thiobases as a potential novel therapeutic option.

The thiopurines, 6-thioguanine and 6-mercaptopurine, are antileukemic agents that are incorporated into DNA following retrieval by the purine salvage pathway (see [1] for a review). Their toxicity requires active DNA mismatch repair (MMR), and thiopurine resistance is an acknowledged phenotype of MMR-defective cells [2, 3]. In addition to these direct cytotoxic effects, DNA thiobases have distinctive photochemical properties [4], the therapeutic potential of which has not been extensively evaluated. We report here that the thiopyrimidine nucleoside 4-thiothymidine is incorporated into DNA. It …

Molecular basis of UV lesion binding and repair inhibition by ETS-family transcription factors.

Mutation hotspots in melanoma frequently occur at DNA binding sites of E26 transformation-specific (ETS)-family transcription factors, as ETS factors stimulate the formation of UV-induced cyclobutane pyrimidine dimers (CPDs) while suppressing repair at ETS-bound DNA sites. To elucidate the molecular mechanism by which ETS factors bind to damaged DNA sites and inhibit repair, we investigated the binding of members from the three major classes of the ETS superfamily (Ets1, ELF1, and PU.1) to cognate DNA containing a cis-syn TpT CPD. These site-specific CPDs modulated ETS recognition and repair b

The mitochondrial DNA-cGAS-STING axis in colorectal cancer: a focused review of context-dependent roles and therapeutic opportunities.

Immune checkpoint blockade produces durable benefit mainly in microsatellite instability-high/mismatch repair-deficient (MSI-H/dMMR) colorectal cancer (CRC), whereas most microsatellite-stable/mismatch repair-proficient (MSS/pMMR) tumors remain poorly inflamed and resistant to immunotherapy. This focused narrative review examines how mitochondrial DNA (mtDNA)-derived danger signals engage the cGAS-STING pathway in CRC, distinguishes canonical DNA sensing from noncanonical STING activation and STING-independent cGAS functions, and evaluates therapeutic strategies that modulate this axis. Optimisation of pembrolizumab therapy for de novo metastatic MSI-H/dMMR colorectal cancer using data-driven delay integro-differential equations.

Colorectal cancer (CRC), the third most commonly diagnosed cancer worldwide, presents a growing public health concern, with 20% of new diagnoses involving de novo metastatic disease and up to 80% of these patients presenting with unresectable metastatic lesions. Microsatellite instability-high (MSI-H) CRC and deficient mismatch repair (dMMR) CRC constitute 15% of all CRC, and 4% of metastatic CRC, and, while less responsive to conventional chemotherapy, exhibit notable sensitivity to immunotherapy, especially programmed cell death protein 1 (PD-1) checkpoint inhibitors such as pembrolizumab.
