Dancing in the dark? The status of late-onset Alzheimer's disease genetics.

Alzheimer's disease (AD) is a genetically complex and heterogeneous disorder. Recent estimates suggest that possibly over 70% of the genetic variance for the disease remains unaccounted for by apolipoprotein E (APOE) and the three known early-onset AD genes (APP, PSEN1, PSEN2). Specifically, one recent segregation analysis predicted the existence of up to four additional susceptibility genes having a similar or greater effect than APOE. However, most of the nearly three dozen putative AD loci proposed to date have only been inconsistently replicated in follow up analyses and more studies are n

Hitch-hiking parasite: a dark horse may be the real rider.

Many parasites engaged in complex life cycles manipulate their hosts in a way that facilitates transmission between hosts. Recently, a new category of parasites (hitch-hikers) has been identified that seem to exploit the manipulating effort of other parasites with similar life cycle by preferentially infecting hosts already manipulated. Thomas et al. (Evolution 51 (1997) 1316) showed that the digenean trematodes Microphallus papillorobustus (the manipulator) and Maritrema subdolum (the hitch-hiker) were positively associated in field samples of gammarid amphipods (the intermediate host), and t

Endocrine secretions under abnormal light-dark cycles and in the blind.

Both endogenous and exogenous factors are involved in regulation of endocrine secretions. Among the exogenous ones, light plays an important role both in animals and in humans. Pineal gland mediates light action on the endocrine system, by means of variations of melatonin (MT) secretion. Here we discuss about the influence of abnormal light-dark cycles and in particular of blindness on pineal and pituitary secretions and on those of correlated glands. MT secretion is usually inhibited by light: thus it reaches the highest levels at night. Exposure to short or long photoperiod causes variations

Dark-adapted thresholds in children with histories of mild retinopathy of prematurity.

PURPOSE: To test the hypothesis that rod-mediated visual thresholds at 10 degrees eccentricity are elevated in children with histories of mild retinopathy of prematurity (ROP). METHODS: Dark-adapted thresholds for detection of 50 msec, 2 degrees diameter spots at a 10 degrees eccentric site, and at a peripheral reference site, 30 degrees eccentric, were measured in 20 children with a history of mild ROP and known courses of refractive development. Ten myopic control subjects also were tested. The thresholds of the ROP and control subjects were compared. RESULTS: Six of the subjects with ROP

Daytime dark exposure increases pineal melatonin in rat pups.

Photic regulation of the pineal melatonin synthesis was studied in 3- to 21-day-old rat pups by exposing the animals to light at night (30-40 min) or to darkness during the day (30-240 min). The pineal melatonin contents were measured by radioimmunoassay. A significant day/night difference in the melatonin content and the nocturnal light-induced decrease were not found until second postnatal week. A novel finding was that at the age of 13-17 days a daytime dark exposure elevated the pineal melatonin content; it was twofold as compared with the normal daytime level and about half of the nocturn…

Ultrastructure of the vestibular dark cell area in patients with acoustic neurinoma.

The human vestibular dark cell (DC) areas of the utricle and ampulla of the lateral semicircular canal were investigated ultrastructurally in 7 patients with acoustic neurinoma. Two types of DCs mainly constituted the epithelial cells of the DC area. One type of DC had basolateral infoldings that were closely interwoven with the melanocyte processes, and the pinocytotic vesicles were frequently found within the basolateral infoldings and the melanocyte processes. The other type of DC had basolateral infolsings stacked upon each other. The former type of DC was more frequently found than the la

Dark adaptation in disabled readers screened for Scotopic Sensitivity Syndrome.

A peripheral retina, photoreceptor, or transient visual-system deficit has been suggested as a basis for dyslexia. We performed dark adaptation using a Goldmann-Weekers adaptometer on 41 dyslexic readers subjected to the Irlen Differential Perceptual Schedule for the Scotopic Sensitivity Syndrome and on 23 volunteers of normal reading ability, all aged between 10 and 20 years. 12 of the 41 disabled readers examined had abnormally poor dark adaptation at peripheral retinal locations consistent with a rod processing-system deficit.

Permeability of the pineal gland of the rat to lanthanum: significance of dark pinealocytes.

The permeability of the pineal gland to lanthanum was investigated by means of transmission electron microscopy and X-ray microanalysis. Different pineal cell populations were studied. Light pinealocytes as well as glial cells were not permeated by the tracer lanthanum, which normally stays extracellularly. Dark pinealocytes showing different degrees of degenerative changes are permeated by lanthanum. The loss of plasma membrane integrity explains the entry of lanthanum into these cells. We conclude: (1) the lanthanum tracer can be used as an indicator of an alteration in membrane permeability

Cyanide and isocyanide abundances in the cold, dark cloud TMC-1.

We have observed emission from HCN, H13CN, HC15N, HN13C, H15NC, HC3N, CH3CN, and possibly CH3NC, and determined an upper limit for NH2CN, toward the cold, dark cloud TMC-1. The abundance ratio [HNC]/[HCN] = 1.55 +/- 0.16 is at least a factor approximately 4 and approximately 100 greater than that observed toward the giant molecular clouds DR 21(OH) and Orion KL, respectively. In contrast, for the corresponding methylated isomers we obtain [CH3NC]/CH3CN] < or approximately 0.1. We also find [NH2CN]/[CH3CN] < or approximately 0.1 and [HC3N]/[CH3CN] = 30 +/- 10. We find no evidence for anomalous

Isolation and properties of lysosomes from dark-grown potato shoots.

A method is described for the isolation of lysosomal fractions from dark-grown potato shoots using a single stage separation on a Ficoll gradient. Peaks of acid hydrolase activity consisting of acid phosphatase, phosphodiesterase, ribonuclease, carboxylic esterase and β-glycerophosphatase were well separated from peaks of mitochondrial and glyoxysomal enzymes. A heavy lysosomal fraction with particle diameters from 0.1 to 1.6 μ and density of 1.10 g cm(-3) containing relatively low hydrolase activity was distinguishable from a light fraction with diameters 0.025 to 0.6 μ and density of 1.07 g

The shadow in the darkness: Case report on adhesive intestinal obstruction secondary to ventriculoperitoneal shunt catheter in an elderly patient.

INTRODUCTION: Shunt placement is an effective therapy for hydrocephalus. Ventriculoperitoneal shunt draining excess cerebrospinal fluid connects the cerebral ventricles to the abdominal cavity. However, intestinal obstruction may ensue as an infrequent complication of the shunt. CASE PRESENTATION: A 65 years old female patient presented with abdominal pain, abdominal bloating, and ceased passage of flatus and stool for six days. She had a history of undergoing a VP shunt procedure due to midbrain obstruction and supratentorial hydrocephalus. Conservative treatment at another local hospital co

Involvement of protein kinase A in the subjective nocturnal rise of melatonin release by chick pineal cells in constant darkness.

Melatonin release by chick cultured pineal cells increases during the dark periods and decreases during the light periods under light-dark cycles, and this rhythmic secretion is maintained under constant conditions with a period of almost 24 hr. The mechanisms by which the circadian oscillator drives the melatonin rhythm under constant conditions have not been elucidated enough. We examined the possibility that cyclic AMP-dependent protein kinase A is involved in the subjective nocturnal increase in melatonin release by chick pineal cells cultured under constant darkness. The subjective noctur…

Effect of exposure to 2,5-hexanediol in light or darkness on the retina of albino and pigmented rats. II. Electrophysiology.

Albino (Sprague-Dawley) and pigmented (Norwegian Brown) male rats were exposed to 2,5-hexanediol (H; 1%) in their drinking water for 5 or 8 weeks, respectively. Half of the rats of each strain were housed in light (average 30 cd/cm2 inside cage, 12 h/day); the other half was kept in constant darkness. Control groups were studied in parallel under identical conditions but without H. Electrophysiological recordings were made 2-5 days and 13 weeks after the end of the exposure to H. Alterations in the visual system, as measured by electroretinography and visual evoked response, were found in grou…

Effects of constant light and darkness on the intrapineal neurons of golden hamsters, stained for tyrosine hydroxylase. A morphometric analysis.

Tyrosine hydroxylase (TH)-positive neurons (TH neurons) were found in the pineal gland of golden hamsters. To examine possible relations between TH neurons and environmental light, we kept male animals under constant light (LL) and darkness (DD) for a week, and morphometrically compared the number, size, and immunoreactivity of TH neurons with those of control animals kept under 12L/12D (LD), using an image processor, Nexus 6400. In LL animals, the number of TH neurons/mm2 of pineal tissue and each cell area were decreased, and immunoreactivity to TH was less than in LD animals. In DD animals,…

Effects of prior exposure to prolonged continuous light on the pattern of melatonin secretion in sheep held under continuous darkness.

To gain insight into the nature of the neural centre regulating melatonin production by the pineal gland we have measured hourly changes in plasma melatonin in 16 ewes held for 42-56 h under continuous darkness following a 28-day period of exposure to continuous light. Plasma melatonin was undetectable while animals were in constant light but increased to normal night time levels (120-2,200 pmol/litre) within 10 min of the onset of darkness in 14 out of the 16 ewes regardless of whether this occurred at 1,200 h (group 1) or 2,400 h (group 2). In four ewes melatonin secretion was maintained for…

Profiles of the Dark Tetrad and the frequency of GAI-related academic misconduct.

The rise of generative artificial intelligence (GAI) presents new challenges to academic integrity, enabling students to engage in academic misconduct with reduced effort and risk of detection. While previous research has examined personality predictors of academic misconduct, very few studies have adopted a person-centered approach to explore how distinct Dark Tetrad personality profiles relate to GAI-related misconduct over time. A total of 781 university students in Taiwan completed the Time 1 survey and 760 completed the follow-up one month later. Latent Profile Analysis based on the Dark
