Hippocampal and subcortical alterations of first-episode, medication-naïve major depressive disorder with panic disorder patients.

The authors conducted a subcortical analysis of first-episode, medication-naïve patients with major depressive disorder comorbid with panic disorder. A sample of 15 patients were compared with 15 healthy controls to explore volumetric differences of subcortical regions and hippocampus, including the putamen and thalamus. The patient group had smaller volumes of left hippocampus, thalamus, and putamen. Subcortical and hippocampal volumes in the patient group showed significant negative correlations with symptom severity. Structural alterations of the putamen, hippocampus, and thalamus might rep…

Effect of tryptophan hydroxylase gene polymorphism on aggression in major depressive disorder and undifferentiated somatoform disorder.

OBJECTIVE: Aggression and anger have been linked with depression, and anger suppression has been linked with somatic symptoms of somatoform disorders. However, the relationship between aggression or anger and genes in patients with depression and somatoform disorders has not been clearly elucidated. The objective of this study was to examine the effect of serotonin-related gene polymorphism on aggression in depressive disorders and somatoform disorders. METHOD: A serotonin-related polymorphic marker was assessed by using single nucleotide polymorphism (SNP) genotyping. 106 outpatients with ma…

Interleukin 10 family gene polymorphisms are not associated with major depressive disorder and panic disorder phenotypes.

Genetic regulation of immune system and inflammatory response may be related to the pathogenesis and manifestations of mood and anxiety disorders. In the present study we examined a range of single-nucleotide polymorphisms (SNP) in chromosomal region 1q32, the locus of interleukin 10 (IL10) gene, in patients with major depressive disorder (n=312) and panic disorder (n=210), and matched healthy controls (n=356). We found no significant associations of the SNPs in IL10 family genes with either diagnostic group. Haplotype analysis revealed seven haplotype blocks, but their frequencies did not dif…

New developments in the management of major depressive disorder and generalized anxiety disorder: role of quetiapine.

Quetiapine has demonstrated efficacy in schizophrenia, bipolar disorder and in the treatment of specific symptom clusters such as agitation and sleep problems in mood disorders. In this review, randomized controlled studies demonstrating efficacy, safety and tolerability of quetiapine in major depressive disorder (MDD) and general anxiety disorder (GAD) are evaluated. The results show that quetiapine monotherapy and quetiapine augmentation of antidepressant treatment in MDD and GAD are efficacious for short-term and maintenance treatment at a dose range between 50 and 300 mg/day. Quetiapine ap…

Elderly patients with major depressive disorder and delusional disorder are at increased risk of subsequent dementia.

All the patients (n=150) admitted to psychogeriatric clinic during a 1-year period were followed up until their death or for 10 years. Fifteen of these were lost during the 10-year follow-up. Thus, follow-up data were available for 135 patients. Twenty-four of these fulfilled at baseline the DSM-III-R diagnostic criteria of major depressive disorder and 18 of delusional disorder after careful medical examination and neuropsychological tests to eliminate organic causes. In the follow-up, the diagnoses were gathered from the death certificates of those patients who died during the 10-year period…

Personality changes in adult subjects with major depressive disorder or obsessive-compulsive disorder treated with paroxetine.

BACKGROUND: Human and animal studies point to 3 dimensions of personality that change during pharmacotherapy with a selective serotonin reuptake inhibitor (SSRI). Specifically, harm avoidance has been found to decrease, social dominance has been found to increase, and hostility in social situations has been found to decrease with SSRI treatment. We sought to determine personality changes in subjects with either major depressive disorder (MDD) or obsessive-compulsive disorder (OCD) treated with paroxetine. We also sought to determine whether or not these personality changes were associated with…

[Sleep electroencephalography in depression and mental disorders with depressive comorbidity].

Traditional scoring of sleep EEG in depressed patients shows abnormalities in sleep maintenance, sleep architecture, REM sleep, the distribution of slow wave and REM sleep during the night. Computerized analysis that comprises the period-amplitude analysis procedure and spectral analysis discloses changes in delta activity and distribution of delta activity. However, these methods of analysing EEG sleep are not able to distinguish the various concepts of depression: endogenous and non-endogenous depression, unipolar and bipolar depression, psychotic and non-psychotic depression. Heart rate variability in generalized anxiety disorder, major depressive disorder and panic disorder: A network meta-analysis and systematic review.

BACKGROUND: Heart rate variability (HRV) in patients with emotional disorders and healthy controls (HCs) has been investigated in many studies but the difference between these emotional disorders was unclear. METHODS: The PubMed, Embase, Medline and Web of Science databases were systematically searched for studies published in English that compared HCs with generalized anxiety disorder (GAD), major depressive disorder (MDD), panic disorder (PD) patients in HRV. We conducted a network meta-analysis to compare HRV in patients with GAD, MDD, PD and HCs. HRV outcomes, including time domain indice…

Maternal stress, prenatal medical illnesses and obstetric complications: Risk factors for schizophrenia spectrum disorder, bipolar disorder and major depressive disorder.

Maternal stress and medical illnesses during early life are well-documented environmental indicators of an increased risk of schizophrenia. Few studies, conversely, have confirmed an association with major affective disorders. The present study examined the impact of maternal stress, medical illnesses and obstetric complications on the development of severe mental disorder in 240 patients with a diagnosis of schizophrenia spectrum disorder, bipolar disorder, or major depressive disorder and matched with 85 controls. Mothers of participants were asked about stressful events during pregnancy usi…

Heterogeneity in the longitudinal course of depressive symptomatology in adults with a history of recurrent major depressive disorder.

BACKGROUND: Depression is highly heterogeneous. Characteristics of previous depressive episodes, additional clinical difficulties, family history of depression and exposure to stressors are associated with poorer outcomes. However, the role of these factors has not been tested simultaneously to explain depression persistence. METHODS: Using longitudinal data from 337 adults with a history of recurrent MDD, we characterised depressive symptom trajectories over 13-years using growth mixture modelling. We tested two partially competing hypotheses as to why depression persists: a) persistent depr

Altered coordination between frontal delta and parietal alpha networks underlies anhedonia and depressive rumination in major depressive disorder.

BACKGROUND: A hyperactive default mode network (DMN) has been observed in people with major depressive disorder (MDD), and weak DMN suppression has been linked to depressive symptoms. However, whether dysregulation of the DMN contributes to blunted positive emotional experience in people with MDD is unclear. METHODS: We recorded 128-channel electroencephalograms (EEGs) from 24 participants with MDD and 31 healthy controls in a resting state (RS) and an emotion-induction state (ES), in which participants engaged with emotionally positive pictures. We combined Granger causality analysis and dat…

Vortioxetine improves symptomatic and functional outcomes in major depressive disorder: A novel dual outcome measure in depressive disorders.

BACKGROUND: With symptomatic remission and functional recovery as the overarching therapeutic objectives of antidepressant therapy, composite endpoint measures that conjointly consider both aspects of treatment are needed. This analysis evaluated the combined effect of vortioxetine on depressive symptoms and functional capacity in adults with MDD. METHODS: NCT01564862, a multinational, double-blind, placebo-controlled, duloxetine-referenced study, conducted between April 2012 and February 2014, in 602 adult outpatients (18-65 years) with moderate-to-severe MDD (Montgomery-Åsberg Depression Ra

Multimodal Machine Learning-Based Prediction of Relapse in Major Depressive Disorder.

IMPORTANCE: Early identification of patients at high risk of relapse is key to personalized preventive strategies in major depressive disorder (MDD). However, current predictive models do not adequately account for the multimodal factors contributing to MDD relapse and fail to provide clinically meaningful predictions. OBJECTIVE: To predict relapse within 2 years in patients with MDD by using a comprehensive machine learning pipeline that integrates an unprecedented set of multimodal data. DESIGN, SETTING, AND PARTICIPANTS: This naturalistic longitudinal prognostic study used data from the M…

Association Between Tctex1 Domain Containing 1 Expression and Major Depressive Disorder.

INTRODUCTION: Major depressive disorder (MDD) involves peripheral immune-inflammatory dysregulation. Tctex1 Domain Containing 1 (TCTEX1D1), a dynein light chain expressed predominantly in monocytes among circulating immune cells, was recently nominated as a candidate risk gene for MDD by brain-derived transcriptome-wide association studies, but its peripheral mRNA expression in MDD has not been examined. We aimed to quantify TCTEX1D1 mRNA in peripheral blood mononuclear cells (PBMCs) of acute-phase MDD patients and to track its trajectory over four weeks of antidepressant treatment. METHODS: …

Neuroimmune dysregulation and comorbidity mechanisms of major depressive disorder and migraine.

BACKGROUND: Although major depressive disorder (MDD) and migraine often present together, their comorbidity mechanisms are complex. This study aims to identify potential biomarkers and common mechanisms of MDD-migraine through neuroimmune microenvironment and transcriptome data, which is crucial for understanding its pathogenesis and developing diagnostic and treatment strategies. METHODS: We constructed a dynamic atlas of neural-peripheral immune cells in patients experiencing a MDD-migraine comorbidity. Differentially expressed genes (DEGs) were identified and performed functional enrichment…

Ethanolamine as a potential biomarker and therapeutic target for depressive disorder.

We investigated cerebrospinal fluid (CSF) ethanolamine (EA) levels in major depressive disorder (MDD), to validate our previous findings on EA (Ogawa et al., 2015) and broaden its biological context and translational relevance. Using human samples (sets A-C) and animal models, we explored the implications for replication, translational potential, therapeutic development, and mechanistic insight. In set A (n = 380), CSF EA levels were low in patients with MDD (P = 0.00047, Cohen's d = -0.59), negatively correlating with depression severity (Spearman's ρ = -0.29, P = 0.00015) and positively corr…

Top 50 Cited Articles on the Treatment of Major Depressive Disorder.

Major depressive disorder (MDD) is a common mental health disorder and is one of the leading causes of disability. Numerous treatment modalities have been studied for MDD, from interventions to psychotherapies. However, no exhaustive investigation has yet provided an overview of influential research on MDD treatments. Studies do not cover influential efforts occurring in the form of a comprehensive bibliometric review. This bibliometric study aims to fill this gap by thoroughly examining the characteristics of the 50 most cited articles on MDD treatment and providing an essential understanding…

Emotional processing biases in the longitudinal course of major depressive disorder.

Negative biases in emotional processing are central to cognitive models of Major Depressive Disorder (MDD), yet it remains unclear whether negatively biased valence attribution covaries with depressive symptoms or represents a more stable vulnerability. In this longitudinal study, we examined emotional valence ratings during a subliminal affective priming task in n = 232 MDD patients and n = 496 healthy controls (HC's) across multiple follow-up assessments, yielding 1395 observations. Multilevel models tested effects of depressive symptom severity, diagnostic group (MDD vs. HC), affective prim…
