Acetylcholine release from the carotid body by hypoxia: evidence for the involvement of autoinhibitory receptors.

The purpose of the present study was to investigate whether hypoxia influences acetylcholine (ACh) release from the rabbit carotid body and, … nt for hypoxia-induced inhibition of ACh release. In the presence of either atropine (10 microM) or domperidone (10 microM), hypoxia stimulated ACh release. These results demonstrate that glomus cells of the rabbit carotid body express ACh and that hypoxia overall inhibits ACh release via activation of muscarinic and dopaminergic autoinhibitory receptors in t …

Supraesophageal manifestations of gastroesophageal reflux disease.

An increasing amount of evidence indicates that gastroesophageal reflux disease (GERD) is a contributing factor to hoarseness, throat clearing, throat discomfort, chronic cough, and shortness of breath. The association between GERD and these supraesophageal symptoms may be elusive. Heartburn and regurgitation are absent in more than 50% of patients. Acid reflux should be considered if signs of GERD are present, symptoms are unexplained, or symptoms are refractory to therapy. The diagnosis of GERD may be unclear, despite a careful history and initial evaluation. A high index of suspicion is req

Discriminative stimulus properties of (+)cathine, an alkaloid of the khat plant.

The effects of the psychostimulant (+)cathine (norpseudoephedrine) were examined in a two-choice, food-motivated, drug-discrimination paradigm. Rats were able to discriminate cathine from vehicle and this effect was dose- and time-dependent. Prior administration of cathine resulted in a diminished response (tolerance) to subsequent cathine and this effect developed and dissipated rapidly. Thus, different dose-response curves were generated depending upon whether cathine or vehicle was administered the day before testing. The development of tolerance also shortened cathine's time course of acti

Dispensing of Antiemetics During Pregnancy in Aotearoa New Zealand, 2005-2020.

BACKGROUND: Antiemetic use during pregnancy is rising in high-income countries. Increasing use of antiemetics has also been documented in Aotearoa New Zealand (NZ); however, the specific antiemetics dispensed have not been reported. AIM: To describe patterns of antiemetic dispensing during pregnancy in NZ, 2005-2020. MATERIALS AND METHODS: Community-based antiemetic dispensings were linked with 1.37 million pregnancies in the New Zealand Pregnancy Cohort. The proportions of pregnancies in which there was at least one dispensing of any and of specific antiemetics were calculated for each trim

Orthostatic hypotension: pathophysiology, assessment, treatment and the paradox of supine hypertension.

Both hypertension and orthostatic hypotension (OH) are strongly age-associated and are common management problems in older people. However, unlike hypertension, management of OH has unique challenges with few well-established treatments. Not infrequently, they both coexist, further compounding the management. This review provides comprehensive information on OH, including pathophysiology, diagnostic workup and treatment, with a view to provide a practical guide to its management. Special references are made to patients with supine hypertension and postprandial hypotension and older hypertensiv

Spawning performance of Clarias gariepinus (Burchell, 1822) induced with ethanol preserved and fresh catfish pituitary extract.

This study was conducted to determine the efficacy of preserved and fresh pituitary extract of Clarias gariepinus (Burchell, 1822) to induced spawning in the same species. Growth performance of fry was also monitored for 15 days to determine the possible effect of natural hormone treatment on the fry. Pituitary glands (PG) were obtained from 12 male broodstocks, six extracted PG were preserved in 96% ethanol 24 h before injection, while the other were extracted and used in their fresh state. The PG solutions (1 g ml-1 of saline water) were injected at 1 ml kg-1 of the female broodstock. Ovapri

Role of dopamine and arterial chemoreceptors in thermal tachypnea in conscious cats.

In mammals submitted to a warm environment, intracerebral injection of dopamine (DA) produces no change or an increase in body temperature a … body (CB) activity during tachypneic breathing. CB stimulation by intravenous injection of NaCN or domperidone reduced thermal tachypnea mainly by decreasing the respiratory frequency, whereas CB inhibition by DA tended to increase frequency and thus tachypnea. It is concluded that 1) in a warm environment, central DA receptors are also greatly involved in h …

Effect of prolonged dopamine receptor blockade on lactotroph activity in the rat pituitary gland.

The dopamine antagonist fluphenazine was administered to young rats in the presence or absence of physiological concentrations of oestradiol for periods of 230 days. An important observation arising from the investigation was that no pituitary tumours were produced by the combination of prolonged dopamine receptor blockade and small amounts of oestradiol. Prolactin (Prl) secretion increased rapidly with fluphenazine but in the absence of oestrogen lactotroph activity subsequently declined after 20 days and this decrease in hormone secretion was accompanied by a greatly increased dopamine turno

Different mechanisms mediated by dopamine D1 and D2 receptors are involved etiologically in activity-stress gastric lesion of the rat.

Rats subjected to activity-stress developed gastric lesions and showed excessive running activity with an increase of light/dark ratio. Daily treatment with centrally acting dopamine antagonists, SCH23390 [(R)-(+)-8-chloro-2,3,4,5-tetrahydro-3-methyl-5-phenyl-1H-3- benzazepin-7-ol] (0.1-10 mg/kg), haloperidol (0.1-10 mg/kg), sulpiride (32-320 mg/kg), clozapine (1-100 mg/kg) and metoclopramide (1-100 mg/kg) suppressed the lesion formation: ID50 values were 0.9, 0.4, 53, 8.9 and 60 mg/kg, respectively. On the other hand, domperidone (1-100 mg/kg), a peripherally acting dopamine antagonist, faile…

Serum angiogenesis-hypoxia biomarkers in relapsing-remitting multiple sclerosis.

BACKGROUND: Studies have demonstrated that the hypoxia-angiogenic axis is dysregulated in progressive multiple sclerosis (MS), with an upregulation of hypoxic biomarkers with a paradoxical reduction of angiogenic biomarkers. In this study, we postulated that the hypoxia-angiogenic axis would be relatively preserved in relapsing-remitting MS (RRMS; a predominantly neuroinflammatory driven process) as opposed to its progressive counterpart (predominantly neurodegenerative driven). METHODS: Serum samples were collected from RRMS participants screened for a randomized pilot trial of domperidone a…
