Autism as a Predictor of Deviant Peer Association: Testing for Dual Systems Model Mediation Effects.

PURPOSE: Deviant peer association is a robust risk factor for offending, that is, any behavior that violates a criminal law. This can include behaviors like drug use, violence, and theft; among others. Autism is a neurodivergent condition characterized by both atypical cognitive development and challenges with social interaction and communication. Despite these impairments, there is a dearth of research focused on the relationship between autism and deviant peer association. Relatedly, there exists no research examining the role that dual systems model constructs (sensation-seeking and impulse

[Informational needs and information-seeking behaviors of recently diagnosed HIV seropositive gay man].

In recent years, substantial changes in doctor-patient relations and consumer oriented attitudes have occurred. Many patients need more information regarding their diagnosis and treatment for developing positive coping strategies to face their disease. This case report uses an informational needs theoretical framework to present the informational needs and information-seeking behaviors of a recently diagnosed HIV seropositive gay man. The data was collected through continuous contacts with the patient during October 1994 to January 1995. This report's findings indicated that need for informati

Cross-reinstatement between 3,4-methylenedioxypyrovalerone (MDPV) and cocaine using conditioned place preference.

3,4-Methylenedioxypyrovalerone (MDPV) is a new psychoactive substance (NPS) considered to be a cocaine-like psychostimulant. The substitutio … ever, memories associated with MDPV required more time to be extinguished. MDPV and cocaine restore drug-seeking behavior with respect to each other, although relapse into drug-taking is always more pronounced with the conditioning drug. The fact that MDPV-treated mice show increased ΔFosB protein levels correlates with its longer extinction time and points to the acti …

Chemogenetic Inhibition and Optogenetic Depotentiation of the Prelimbic Cortex to Paraventricular Thalamus Pathway Attenuate Abstinence-Induced Plasticity and Heroin Seeking in Rats.

Opioid use disorder (OUD) is characterized by high relapse rates, underscoring an urgent need to identify neurobiological mechanisms of opioid seeking. The paraventricular thalamus (PVT) is a key brain region implicated in drug seeking in humans and rodents. While the role of its downstream targets is beginning to be clarified, the upstream inputs that regulate PVT activity during opioid seeking remain poorly understood. The prelimbic cortex (PL) provides substantial excitatory input to the PVT and has a well-established role in cue-guided behaviors and drug seeking. However, the contribution

Cocaine abuse and midbrain circuits: Functional anatomy of hypocretin/orexin transmission and therapeutic prospect.

Cocaine abuse remains a pervasive public health problem, and treatments thus far have proven ineffective for long-term abstinence maintenance. Intensive research on the neurobiology underlying drug abuse has led to the consideration of many candidate transmitter systems to target for intervention. Among these, the hypocretin/orexin (hcrt/ox) neuropeptide system holds largely untapped yet clinically viable therapeutic potential. Hcrt/ox originates from the hypothalamus and projects widely across the mammalian central nervous system to produce neuroexcitatory actions via two excitatory G-protein

The effects of cannabinoid CB1, CB2 and vanilloid TRPV1 receptor antagonists on cocaine addictive behavior in rats.

There is evidence that indicates that tonic activation of cannabinoid CB1 receptors plays a role in extinction/reinstatement of cocaine seeking-behavior but is not involved in the maintenance of cocaine self-administration. To further explore the importance of other endocannabinoid-related receptors in an animal model of cocaine addiction, the present paper examines cannabinoid CB2 receptor antagonist N-((1S)-endo-1,3,3-trimethylbicyclo(2.2.1)heptan-2-yl)-5-(4-chloro-3-methylphenyl)-1-(4-methylbenzyl)-pyrazole-3-carboxamide (SR144528) and the transient receptor potential vanilloid type-1 (TRPV

Disulfiram attenuates drug-primed reinstatement of cocaine seeking via inhibition of dopamine β-hydroxylase.

The antialcoholism medication disulfiram (Antabuse) inhibits aldehyde dehydrogenase (ALDH), which results in the accumulation of acetaldehyde upon ethanol ingestion and produces the aversive 'Antabuse reaction' that deters alcohol consumption. Disulfiram has also been shown to deter cocaine use, even in the absence of an interaction with alcohol, indicating the existence of an ALDH-independent therapeutic mechanism. We hypothesized that disulfiram's inhibition of dopamine β-hydroxylase (DBH), the catecholamine biosynthetic enzyme that converts dopamine (DA) to norepinephrine (NE) in noradrener

Is It Possible to Shift from Down to Top Rank? A Focus on the Mesolimbic Dopaminergic System and Cocaine Abuse.

Impaired social behavior is a common feature of many psychiatric disorders, in particular with substance abuse disorders. Switching the preference of the substance-dependent individual toward social interaction activities remains one of the major challenges in drug dependence therapy. However, social interactions yield to the emergence of social ranking. In this review, we provide an overview of the studies that examined how social status can influence the dopaminergic mesolimbic system and how drug-seeking behavior is affected. Generally, social dominance is associated with an increase in dop…

Self-destructiveness in adolescence.

This study is an attempt to formulate the dynamics of the group of adolescents involved in major drug use and other antisocial behavior. As consultant to a drug rehabilitation center for teenagers, the author has had occasion to interview and evaluate a large number of youths whose lives had taken such a turn. Certain aspects of the life style and behavior patterns of these young people show striking similarities. In particular, their behavior evidences the presence of a central self-destructive moiety which can be likened to a negative ego ideal; this acts to shape and to direct their lives.

Dopamine agonist and antagonist responders as related to types of nicotine craving and facets of extraversion.

The role of dopamine (DA) in drug seeking behavior has been a matter of debate in the literature: One theory claims that DA triggers incentive motivational behavior, the other one favors the idea that DA itself is the rewarding property induced by the positive stimuli. The present experiment tries to contribute to a solution of the controversy by the approach of relating DA-associated motivational behavior to constellations of hormone response to a DA agonistic and DA antagonistic challenge performed in the same subjects and by relating their responses to different aspects of personality and s

Distinct Behavioral Profiles and Neuronal Correlates of Heroin Vulnerability Versus Resiliency in a Multi-Symptomatic Model of Heroin Use Disorder in Rats.

OBJECTIVE: The behavioral and diagnostic heterogeneity within the opioid use disorder (OUD) diagnosis is not readily captured in current animal models, limiting the translational relevance of the mechanistic research that is conducted in experimental animals. The authors hypothesized that a nonlinear clustering of OUD-like behavioral traits would capture population heterogeneity and yield subpopulations of OUD vulnerable rats with distinct behavioral and neurocircuit profiles. METHODS: Over 900 male and female heterogeneous stock rats, a line capturing genetic and behavioral heterogeneity pre

Revisiting long-access versus short-access cocaine self-administration in rats: intermittent intake promotes addiction symptoms independent of session length.

In rats, continuous cocaine access during long self-administration sessions (6 versus 1-2 hours) promotes the development of behavioral symp … der a progressive ratio schedule of reinforcement and cocaine-induced reinstatement of extinguished drug-seeking behavior. In summary, long IntA-sessions lead to greater cocaine intake than shorter IntA-sessions, but the two conditions are equally effective in evoking the patterns of drug-taking and drug-seeking that define addiction. This suggests that chronic intermi …

eIF2α dephosphorylation in basolateral amygdala mediates reconsolidation of drug memory.

Maladaptive memories elicited by exposure to environmental stimuli associated with drugs of abuse are often responsible for relapse among addicts. Interference with the reconsolidation of drug memory can inhibit drug seeking. Previous studies have indicated that the dephosphorylation of the eukaryotic initiation factor 2 α-subunit (eIF2α) plays an important role in synaptic plasticity and long-term memory consolidation, but its role in the reconsolidation of drug memory remains unknown. The amygdala is required for the reconsolidation of a destabilized drug memory after retrieval of drug-paire

Transcriptional signatures of heroin intake and seeking throughout the brain reward circuit.

Opioid use disorder (OUD) looms as one of the most severe medical crises currently facing society. More effective therapeutics for OUD requires in-depth understanding of molecular changes supporting drug-taking and relapse. Recent efforts have helped advance these aims, but studies have been limited in number and scope. Here, we develop a brain reward circuit-wide atlas of opioid-induced transcriptional regulation by combining RNA sequencing (RNAseq) and heroin self-administration in male mice modeling multiple OUD-relevant conditions: acute heroin exposure, chronic heroin intake, context-indu

Drug assertiveness and sexual risk-taking behavior in a sample of HIV-positive, methamphetamine-using men who have sex with men.

Drug assertiveness skills have been demonstrated to be effective in reducing substance use behaviors among patients with alcohol or heroin use disorders. This study examined the association between drug assertiveness and methamphetamine use, psychological factors, and sexual risk behaviors in a sample of 250 HIV-positive men who have sex with men enrolled in a safer sex intervention in San Diego, CA. Less assertiveness in turning down drugs was associated with greater frequency and larger amounts of methamphetamine use, lower self-esteem, higher scores on a measure of sexual sensation seeking,

Factors impacting military emergency medicine residents' sleep and barriers to seeking help: A sequential exploratory mixed-methods study.

Emergency medicine (EM) resident physicians face unique challenges in adhering to the principles of sleep hygiene due to the nature of EM shift work. They are at higher risk for sleep disorders, sleep-related errors and accidents, and impaired performance due to constantly changing schedules. Military residents also perceive a stigma against seeking mental health care and taking sleeping medications. This mixed-methods study investigated sleep quality and barriers to seeking care among EM residents at a single center, three-year, Active-Duty military EM residency program. A survey was made ava
