Analysis of CCR5-Delta 32 and CCR2-V64I polymorphisms in a cohort of Spanish HCV patients using real-time polymerase chain reaction and fluorescence resonance energy transfer technologies.

Nowadays it is clear that chemokine-chemokine receptor interactions are important in chronic hepatitis C virus (HCV) infection. The objective of the present study was to elucidate the involvement of the CCR5-Delta 32 and CCR2-V64I polymorphisms in the response to the HCV infection, as well as in the histological damage and the outcome of the infection. A cohort of 139 patients with hepatitis C and 100 healthy blood donors were analysed for both polymorphisms using real-time polymerase chain reaction (PCR) and LightCycler technology. We have detected the CCR5-Delta 32 allele in 15 of 278 HCV ch

Impact of dose and dose-averaged linear energy transfer on oro-pharyngeal-mucosal toxicity in patients with non-squamous head and neck cancers treated with carbon-ion radiotherapy.

BACKGROUND: In head and neck carbon-ion radiotherapy (CIRT), high dose-averaged LET (LETd) and relative biological effectiveness weighted dose (DRBE) often overlap the oro-pharyngeal-mucosa (mucosa of oral cavity, oro-pharynx, nasopharynx, pharynx) beyond the target, resulting in serious complications. We investigated clinical and dosimetric predictors for late severe mucosal-toxicity/ oro-antral fistula (OAF) post-CIRT. MATERIALS AND METHODS: CIRT plans for 51 patients with non-squamous head and neck cancers (NSHNCs) were evaluated and correlated with clinical outcome. Prescription was DRBE

Fluorescence resonance energy transfer analysis of the RANTES polymorphisms -403G --> A and -28G --> C: evaluation of both variants as susceptibility factors to HIV type 1 infection in the Spanish population.

The identification of genetic factors predisposing or protecting against HIV-1 infection has been an important aim in AIDS research. Two of these factors are located in the promoter region of the CCL5 gene, which encodes the RANTES (regulated on activation, normal T cells expressed and secreted) chemokine, an inhibitor agent for M-tropic HIV-1 strains. More specifically, the role of single-nucleotide polymorphisms (SNPs) -403G --> A and -28C --> G has been evaluated in the course of HIV-1 infection in several populations with different genetic, geographic, and ethnic backgrounds. Here we prese

Longitudinal Tumor, Vascular, and Immune Cell Response in Two Rat Prostate Carcinomas After Isoeffective Photon, Proton, and Carbon Ion Irradiation: Impact of Linear Energy Transfer, Dose Level, and Hypoxia.

Background/Objectives: High linear energy transfer (LET) carbon ions achieved more effective and biologically robust tumor control than photons in preclinical prostate cancer models; however, the longitudinal development of histopathological parameters remains insufficiently characterized, limiting the selection of the optimal treatment modality in patients. This study analyzed the temporal histological patterns after isoeffective photon, proton, and carbon ion irradiations. Methods: Two Dunning R3327 prostate carcinoma sublines (H, HI) grown subcutaneously in male Copenhagen rats received sin

A Turn-On-Type Fluorescence Resonance Energy Transfer Eco-friendly Method for Nitazoxanide Quantification in Pharmaceutical Dosage Form and Spiked Plasma: Evaluation of Greenness Profile Using Different Assessment Tools.

A brand-new class of anti-infective drugs that work against bacteria, viruses, and protozoan parasites is nitazoxanide and related thiazolides. Thiazolides have also been shown to cause cell cycle arrest and apoptotic cell death in cancer cells in recent years. In this study, an eco-friendly, spectrofluorimetric technique that is verified, easy, and sensitive has been proposed for quantifying nitazoxanide (NTZ), a broad-spectrum antiparasitic drug. When NTZ is reduced with zinc (Zn) powder in an acidic media, a highly fluorescent product is produced. To get the highest sensitivity, different e

Detection of Babesia canis vogeli and Hepatozoon canis in canine blood by a single-tube real-time fluorescence resonance energy transfer polymerase chain reaction assay and melting curve analysis.

A real-time fluorescence resonance energy transfer polymerase chain reaction (qFRET PCR) coupled with melting curve analysis was developed for detection of Babesia canis vogeli and Hepatozoon canis infections in canine blood samples in a single tube assay. The target of the assay was a region within the 18S ribosomal RNA gene amplified in either species by a single pair of primers. Following amplification from the DNA of infected dog blood, a fluorescence melting curve analysis was done. The 2 species, B. canis vogeli and H. canis, could be detected and differentiated in infected dog blood sam

Metallophilic interactions in closed-shell d10 metal-metal dicyanide bonded luminescent systems Eu[Ag(x)Au(1-x)(CN)2]3 and their tunability for excited state energy transfer.

We report on the heterobimetallic system, Eu[Ag(x)Au(1-x)(CN)(2)](3) (x = 0-1) in which sensitization of europium luminescence occurs by energy transfer from [Ag(x)Au(1-x)(CN)(2)](-) donor excited states. The donor states have energies which are tunable and dependent on the Ag/Au stoichiometric ratio. These layered systems exhibit interesting properties, one of which is their emission energy tunability when excited at different excitation wavelengths. In this paper, we report on their use as donor systems with Eu(III) ions as acceptor ions in energy transfer studies. Luminescence results show

One-step rapid reverse transcription-PCR assay for detecting and typing dengue viruses with GC tail and induced fluorescence resonance energy transfer techniques for melting temperature and color multiplexing.

BACKGROUND: Dengue fever is an arthropod-borne infection caused by dengue viruses (DVs; DEN-1 to DEN-4). Early diagnosis is critical to prevent severe disease progression and the spreading of DV because no vaccine or specific treatment is available; therefore, a rapid and specific diagnostic assay capable of detecting and typing all serotypes would be ideal. METHODS: We amplified RNA samples from all 4 DV serotypes and Japanese encephalitis virus with 4 serotype-specific forward primers and a universal species-specific reverse primer. DEN-1 and DEN-3 forward primers were labeled at their 5' e

Naked-eye and fluorescence resonance energy transfer based ratiometric fluorescent probe for rapid, sensitive and selective detection of hydrazine and its applications in imaging of environmental samples and living systems.

Hydrazine, a compound recognized for its carcinogenic and genotoxic properties, presents a significant threat to human health via environmental exposure and drug metabolism. The detection of hydrazine is essential for safeguarding human health. However, a tool capable of accurately detecting hydrazine across diverse sample types, such as soil, water sources, and plant specimens contaminated by hydrazine leakage, as well as cells and live mice containing endogenously generated hydrazine from drug metabolism, is still lacking. In this study, we have designed and synthesized a ratiometric fluores

Induction and rejoining of DNA double-strand breaks in normal human skin fibroblasts after exposure to radiation of different linear energy transfer: possible roles of track structure and chromatin organization.

DNA double-strand breaks are nonrandomly induced by high-LET radiation. Differences in the induction and rejoining of DSBs after irradiation with ions having different LET were detected by fragment analysis. The data obtained indicate that the track structure of the traversing particle and its interaction with the different chromatin structures of the cellular DNA influence the yield as well as the distribution of the induced damage. The induction and rejoining of clustered DSBs induced by the same nitrogen ion fluence at LETs of 80-225 keV/microm were investigated by a detailed analysis of th

Aggregation-Induced Electrochemiluminescence Based on a Zinc-Based Metal-Organic Framework and a Double Quencher Au@UiO-66-NH2 for the Sensitive Detection of Amyloid β 42 via Resonance Energy Transfer.

A novel sandwich electrochemiluminescence (ECL) biosensor based on aggregation-induced electrochemiluminescence resonance energy transfer (AIECL-RET) was designed for the sensitive detection of amyloid β42 (Aβ42). The synthesized silver nanoparticle-functionalized zinc metal-organic framework (Ag@ZnPTC) and gold nanoparticle-functionalized zirconium organic framework (Au@UiO-66-NH2) were used as the ECL donor and acceptor, respectively. AgNPs were generated in situ on the surface of ZnPTC, which further improved the ECL intensity and the loading of antibody 1 (Ab1). Under the optimized experim

Ultrasensitive Photoelectrochemical Biosensor for microRNA-155 Based on Energy Transfer between Au Nanocages and Red Emission Carbon Dot-Assembled Nanosheets Coupled with the Duplex-Specific Nuclease Enzyme-Assisted Target Recycling Strategy.

Energy transfer (ET) is an effective tool to construct photoelectrochemical (PEC) biosensors for its high sensitivity. Since the materials to develop ET systems are limited, exploring new and universal ET systems is significant. Herein, new photoactive nanosheets (R-CDs NS) formed by self-assembling of red emission carbon dots (R-CDs) have been synthesized, which exhibit wide visible light absorption and stable photocurrent response and have an obvious sensitization effect for TiO2. Gold nanocages (AuNCs), whose absorption overlap well with the R-CDs' emission, were synthesized and served as P

Syntheses and studies of electron/energy transfer of new dyads based on an unsymmetrical perylene diimide incorporating chelating 1,10-phenanthroline and its corresponding square-planar complexes with dichloroplatinum(ii) and dichloropalladium(ii ).

Perylene diimides (PDIs) are among the most versatile and functional dyes for supramolecular structures displaying characteristic high absorptions and photo-luminescence properties as the prerequisite for optoelectronic thin film devices. Despite intense investigations into these semi-conducting and electro-active materials, details of their electronic structure are still under examination. In particular, non-planar twisted PDIs as an electron acceptor is a promising model system for efficient charge generation and transport processes. Therefore, a new dyad, an unsymmetrical PDI, N'-(2-ethylhe

Synthesis, characterization and energy transfer studies of fluorescent dye-labeled metal-chelating polymers anchoring pendant thiol groups for surface modification of quantum dots and investigation on their application for pH-responsive controlled release of doxorubicin.

We describe the synthesis of an end-functionalized fluorescent dye-labeled poly(N-(2-thiolethyl methacrylamide) as a metal chelating polymer with a very narrow size distribution and controllable molecular weights by reversible addition-fragmentation chain transfer polymerization. In following, we apply this water-soluble metal-chelating polymer for surface modification of cadmium telluride quantum dots (CdTe QDs) by ligand exchange method. The obtained poly(N-(2-thiolethyl methacrylamide)/CdTe QDs hybrid was fully characterized using Fourier transform infrared spectroscopy (FT-IR), thermo-grav

Improved detection of JC virus in AIDS patients with progressive multifocal leukoencephalopathy by T-antigen specific fluorescence resonance energy transfer hybridization probe real-time PCR: evidence of diverse JC virus genotypes associated with progressive multifocal leukoencephalopathy in Southern Africa.

JC virus (JCV) causes progressive multifocal leukoencephalopathy under conditions of immunosuppression, especially associated with HIV. Despite the high prevalence of HIV-1 infection, few cases of progressive multifocal leukoencephalopathy have been reported and only a small number of JCV strains have been characterized in AIDS patients in Southern Africa. Diagnosis of progressive multifocal leukoencephalopathy through PCR detection of JCV DNA in CSF may result in false negative results if variable regions of the genome are targeted. Characterization of circulating JCV genotypes in Southern Af
