In an attempt to lay the background work of immunodiagnosis of human leishmaniasis in Taiwan, the usefulness of enzyme-linked immunosorbent assay (ELISA) for detection serum antibodies in large regional groups was studied. Antigens were prepared from promastigotes of Leishmania tropica major recently isolated in our laboratory by Chao. One hundred and sixty-five serum samples were collected from medical students of National Yang-Ming Medical College (YM), Taipei, 61 from residents of San-hsing District (SH), I-lan County, 9 from residents of Wu-lai District (WL), Taipei County, 2 from patients

The recently described DIG-ELISA test was evaluated on 38 sera from patients with onchocerciasis, using a soluble, crude extract of Onchocerca volvulus dissected out from glycerol-preserved nodules. Results showed that the diameters of the orange-yellow antibody-antigen reaction zones obtained with infected sera were significantly greater than those obtained with normal human sera. A close correlation was observed between the diameters of the reaction zones and antibody concentration (expressed as log of the serum dilution), indicating the potential utility of this test in the quantitative det

An enzyme-linked immunosorbent assay was evaluated for the detection of antibody in sera of equine naturally infected with Histoplasma farciminosum 'epizootic lymphangitis'. Ten sera from naturally infected horses were tested. A hydrogen peroxide ABTS mixture constituted the substrate. The reactions were read as the absorbance values measured at 405 nm using a spectrophotometer. The standard deviation and the average percentage of the absorbance values of the different serum samples were considered in the interpretation of the results. All sera were proved positive with variations in the diffe

The development and evaluation of an indirect ELISA for IgG and IgM antibodies in human sera against the genus-specific chlamydial antigen is described. Application of commercial anti-immunoglobulins for production of conjugates usually resulted in direct conjugate/antigen reactions. Therefore, all immunoreagents had to be prepared by the authors. Using anti-immunoglobulins purified by affinity chromatography for the production of anti-human IgG(H + L)-peroxidase conjugates, these conjugates showed high specific activity and could be diluted 1/10,000 for the test, whereby direct reactions with

In a prospective study paired sera of 14 patients suffering from Bell's palsy were examined for antibodies against Varicella-zoster Virus (VZV), Herpes-simplex Virus (HSV), Cytomegalovirus (CMV) and Epstein-Barr Virus (EBV). For determination of antibodies against VZV, HSV and CMV an enzyme-linked immunosorbent assay was carried out. Indirect immunofluorescence was carried out to determine antibodies against EBV. Serum samples were taken within 5 days of Bell's palsy having been diagnosed and were compared with further serum samples taken 4 weeks later. No evidence of significant differences b

Tay-Sachs disease results from a loss of activity of hexosaminidase A (HEXA) in body tissues and fluids. Heterozygotes for the disease are usually identified by their relatively low ratio of heat-labile HEX A to total hexosaminidase. During pregnancy an intermediate isoenzyme (HEX I) increases in activity in serum and obscures the heterozygote status. HEX I dose not increase in leucocytes, tears and other body tissues but because of technical difficulties in these assays we examined the feasibility of using a radioimmunoassay for HEX A. By univariate analysis, the heat denaturation assay gave

We have developed an enzyme-linked monoclonal antibody inhibition assay to detect bacterial antigens in cerebrospinal fluid. The monoclonal antibody used in this immunodiagnostic test was produced by continuous cultures of hybrid myeloma cell lines. Using this assay, type III GBS antigen was detected in CSF specimens from 11 culture-proven cases of GBS meningitis and in the knee aspirate from an infant with GBS septic arthritis. Five spinal fluid specimens from meningitis due to other bacterial pathogens and ten other control samples were negative. The ELMIA detected streptococcal antigen at a

BACKGROUND: Heparin-induced thrombocytopenia (HIT) is a potentially fatal complication of therapy with unfractionated or low molecular weight heparins that can result in thrombocytopenia and thrombosis. Timely clinical and laboratory diagnoses are imperative to prevent further morbidity and mortality.
OBJECTIVES: To evaluate a rapid high-sensitivity chemiluminescence luminescence enzyme immunoassay (HISCL HIT IgG assay) compared with automated and ELISA antigen assays and heparin-induced platelet activation (HIPA) functional assays on samples healthy donors, patients suspected of having HIT,

INTRODUCTION: Serum protein electrophoresis and serum free light chain (SFLC) assays are standard methods for monitoring patients with multiple myeloma (MM). However, patients with non-secretory MM often require invasive bone marrow biopsies to monitor treatment response and disease progression. Recently, serum soluble B-cell maturation antigen (sBCMA) has been proposed as an alternative biomarker for monitoring of MM, including non-secretory disease. We aimed to optimize the performance of and validate a serum sBCMA enzyme-linked immunosorbent assay (ELISA) from R&D Systems for research and e

Hepatitis E virus (HEV) is transfusion-transmissible and therefore poses a risk to blood transfusion safety. Seroprevalence studies are useful for estimating disease burden and determining risk factors. Considerable variability in the sensitivity of HEV antibody detection assays exists. This study aimed to compare the performances of commercially available HEV enzyme-linked immunosorbent assays (ELISA) in Australian blood donor samples. Plasma samples that tested positive (n = 194) or negative (n = 200) for HEV IgG (Wantai HEV IgG ELISA) were selected. Of the 194 HEV IgG positive samples, 4 we

This study was undertaken to detect pregnancy in Iraqi riverine buffalo (Bubalus bubalis) using three different methods (rectal palpation, plasma progesterone concentration and detection of the presence of pregnancy-specific protein B (PSPB) with the BioPRYN(®) enzyme-linked immunosorbent assay (ELISA) test. The aim of the study was to identify the most sensitive, early and accurate method for detecting pregnancy. Twenty-two female riverine buffalo that were 6.0 ± 0.93 years old were used. Four blood samples per buffalo were taken via jugular venipuncture at days 22-24, 32-34, 42-44 and 58-61

In the present study, diagnostic methods for canine G(M1)-gangliosidosis were examined by comparing a DNA mutation assay with an enzyme assay. Sixty-two Shiba dogs of a pedigree with G(M1)-gangliosidosis were differentiated into 3 genotypes, i.e., normal, heterozygous, and homozygous affected dogs, using a DNA mutation assay, which consists of polymerase chain reaction amplification and the determination of restriction fragment length polymorphisms. The beta-galactosidase activity in leukocytes, umbilical cords, and plasma was measured using 4-methylumbelliferyl beta-D-galactoside and p-nitrop

Total immunoglobulin G (IgG) and IgG subclass reactivities with purified mumps glycoproteins (GP) and nucleoprotein (NP), measured in enzyme-linked immunosorbent assays (ELISAs), were compared with titers in a mumps microneutralization assay (NT). For determination of mumps immunity, the sensitivity of both ELISAs was 100% in comparison with the NT and the specificity was 90%. IgG1 was the dominant subclass against the two antigens found in seropositive healthy individuals. In samples from patients with clinical mumps infections and positive mumps IgM, titer rises of total IgG against NP were

Pegmolesatide is a novel synthetic erythropoietin-mimetic agent that binds to and activates the erythropoietin receptor (EPOR), thereby stimulating erythropoiesis. Owing to its erythropoiesis-stimulating activity, it has been included in the latest World Anti-Doping Agency (WADA) Prohibited List as an erythropoietin receptor agonist (S2.1.1). The current analytical approach for pegmolesatide in doping analysis mainly relies on bottom-up nano-liquid chromatography-high-resolution tandem mass spectrometry (LC-HRMS) in serum. Although accurate and sensitive, this approach involves extensive sampl
