PURPOSE: To establish primary and immortalized cell cultures of human lens epithelial cells for a model system investigating human lens epithelial physiology and cataract.
METHODS: Human lens epithelial cells in culture were grown by isolating epithelium fragments from infant human lenses from patients who underwent treatment for retinopathy of prematurity and by allowing epithelial cells to grow from explants. Migration of epithelial cells is an important feature of wound healing. Components of extracellular matrix stimulate migration of other cells, and we hypothesized that basement membrane components, laminin and type IV collagen, stimulate migration of bovine bronchial epithelial cells (BBEC). Epstein-Barr Virus (EBV) is an oncogenic herpesvirus associated with two human malignancies. Initially linked in the etiology of Burkitt's lymphoma, a B cell lymphoma, it has subsequently been implicated in the causation of nasopharyngeal carcinoma, an epithelial tumor of considerable world health importance. More recently a number of head and neck tumors have been shown to harbor the virus, suggesting a possible role for EBV in their development.
