A review of the mechanisms and effectiveness of dietary polyphenols in reducing oxidative stress and thrombotic risk.

Dietary sources of polyphenols, which are derivatives and/or isomers of flavones, isoflavones, flavonols, catechins and phenolic acids, possess antioxidant properties and therefore might be important in preventing oxidative-stress-induced platelet activation and attenuating adverse haemostatic function. Free radicals, including reactive oxygen and nitrogen species, promote oxidative stress, leading to platelet hyperactivation and the risk of thrombosis. The consumption of antioxidant/polyphenol rich foods might therefore impart anti-thrombotic and cardiovascular protective effects via their in…

Optimisation of an extraction procedure and chemical characterisation of Croatian propolis tinctures.

Three spectrophotometric methods for the quantitative determination of different flavonoid groups and total phenolics in Croatian propolis samples were optimised and validated. The assay based on the formation of aluminium chloride complex (with galangin as a standard) was applied to the quantification of flavones and flavonols, while the 2,4-dinitrophenylhydrazine method (with pinocembrine as a reference) was used for the quantification of flavanones. Total phenolic content was measured by the Folin-Ciocalteau method using reference solution of caffeic acid:galangin:pinocembrine (1:1:1). Thro…

A rapid screening assay for inhibitors of 11beta-hydroxysteroid dehydrogenases (11beta-HSD): flavanone selectively inhibits 11beta-HSD1 reductase activity.

A rapid screening assay for chemicals inhibiting 11beta-hydroxysteroid dehydrogenase (11beta-HSD) type 1 or type 2 using lysates from stably … tive activity. We observed a reduced inhibitory effect of hydroxylated flavanone derivatives and of flavones containing a double-bond between atom C2 and C3. Flavanone was specific for 11beta-HSD1 and did not inhibit 11beta-HSD2. Our results reveal that a variety of environmental compounds exert distinct inhibitory effects on 11beta-HSD1 and 11beta-HSD2, o …

Probing the polyphenolic flavonoid, morin as a highly efficacious inhibitor against amyloid(A4V) mutant SOD1 in fatal amyotrophic lateral sclerosis.

Deposition of misfolded protein aggregates in key areas of human brain is the quintessential trait of various pertinent neurodegenerative disorders including amyotrophic lateral sclerosis (ALS). Genetic point mutations in Cu/Zn superoxide dismutase (SOD1) are found to be the most important contributing factor behind familial ALS. Especially, single nucleotide polymorphism (SNP) A4V is the most nocuous since it substantially decreases life expectancy of patients. Besides, the use of naturally occurring polyphenolic flavonoids is profoundly being advocated for palliating amyloidogenic behavior o

Integrated virtual screening and experimental validation identify dietary phytochemicals as PI3Kα inhibitors with antiproliferative activity.

The phosphoinositide 3-kinase (PI3K)/AKT/mTOR pathway is a central regulator of cancer progression, with frequent activation of the PI3Kα catalytic subunit (p110α) in breast cancer. Here, an integrated in silico to experimental workflow was applied to systematically identify dietary phytochemicals capable of inhibiting PI3Kα and suppressing cancer cell proliferation. A curated PhytoHub library comprising 1966 food-derived compounds was screened by molecular docking against the PI3Kα catalytic domain, followed by isoform-specific enzymatic validation and cellular evaluation. Several flavonoids

Discovery of an isoflavone biosynthetic pathway in Iridaceae plants.

Isoflavones, characterized by their distinctive C6-C3-C6 skeleton, represent an important class of plant secondary metabolites with considerable pharmacological and physiological relevance. Although their biosynthetic pathways have been extensively elucidated in Fabaceae species, the molecular mechanisms governing isoflavone production in other plant families remain largely unexplored. Herein, we uncovered an isoflavone biosynthetic pathway in Iridaceae plants. Notably, it involves an uncharacterized CYP736D family isoflavone synthase, a non-CYP450 (iso)flavone 6-hydroxylase, two cation-depend…

A Novel Nrf2 Activator Suppresses Osteoclastogenesis and Ovariectomy-Induced Bone Loss by Directly Interfering Keap1-Nrf2 Protein-Protein Interaction.

Drug-targeting osteoclasts is a mainstream strategy to treat osteoporosis. The marketed antiosteoporotic medications present various adverse side effects and limited clinical responses. Activating Nrf2 attenuates osteoclastogenesis, and it is considered as a promising strategy for osteoporosis therapy. Currently, no Nrf2 activators have progressed to clinical trial for the treatment of osteoporosis. In this work, a series of 5-selenyl-flavone were efficiently prepared, and their inhibitory effects on RANKL-induced osteoclastogenesis were tested. Compound 5c was identified as the most potent co

Revealing the dynamic changes in the metabolites and sensory quality of Citri Reticulatae Pericarpium during aging using feature-based molecular networks and metabolomics.

The contribution of non-volatile metabolites to the unique flavor formation in Citri Reticulatae Pericarpium (CRP) during aging remains poorly characterized. This study employed sensory evaluation, electronic tongue (E-tongue) assessment, untargeted metabolomics in conjunction with feature-based molecular networks (FBMN) to elucidate flavor differentials and metabolic mechanisms during the aging process of CRP. The results showed that extended aging time enhanced the bitterness, astringency, aftertaste-bitterness, aftertaste-astringency, and sweetness of the CRP. FBMN tool used for the first t

Apigenin attenuated sepsis induced acute lung injury via polarizing macrophage towards M2 by blocking miR-146a →TLR7 interaction.

TLR7 (Toll-like receptor 7) has been indicated as an important sensor for single -stranded RNA contributes to systemic inflammation and mortality in acute lung injury (ALI), which is an acute diffuse inflammatory lung injury. Cumulative results show that macrophages contribute to the development and progression of ALI through the secretion of inflammatory cytokines/chemokines. Here we show that macrophage polarizes towards M1 phenotype and TLR7 signaling is activated in septic mice. Moreover, TLR7 deficiency promotes macrophage polarized towards M2 phenotype and attenuates ALI. Strikingly, the

Extremely Rare Flavonoid Glycosides Identified in the Stems of Ephedra gerardiana by HPLC-MS and Their Antioxidant Activity.

The plants of the genus Ephedra are mainly known for the contents of alkaloids; however, it has recently been found that they also contain phenolic constituents that show interesting bioactivities, e.g., antioxidative or antimicrobial. From among the Ephedrae herba, the Ephedra gerardiana seems to be relatively poorly researched in terms of flavonoid presence. In this study, on the basis of the results of high-pressure liquid chromatography-mass spectrometry (HPLC-MS) with cone voltage-induced fragmentation analysis, which are discussed in detail, the flavonoid glycosides present in Ephedra ge

The influence of rolling pressure on the changes in non-volatile compounds and sensory quality of congou black tea: The combination of metabolomics, E-tongue, and chromatic differences analyses.

Rolling represents an essential stage in congou black tea processing. However, the influence of rolling pressure on tea flavor and non-volatile compounds remains unclear. Herein, a combination of untargeted metabolomics, tea pigments quantification, E-tongue, colorimeter and sensory evaluation was used to evaluate the effect of rolling pressure on black tea quality. As the rolling pressure increased, theaflavins (TFs), thearubigins (TRs), and theabrownins (TBs) significantly elevated. The tea metabolic profiles fluctuated and 47 metabolites were identified as key differential metabolites inclu
