Parasite and vertebrate host genetic heterogeneity determine the outcome of infection by Schistosoma mansoni.

Intraspecific variation in Schistosoma mansoni infection and modulation of its expression by vertebrate host genetics was studied by evaluation of some biological parameters of the infection in BALB/c and C57BL/6 mice infected with one Brazilian (BH) and two Venezuelan (YT and SM) laboratory strains of the parasite. Mice infected with 60 cercariae of each parasite strain were euthanized at 5, 6, 8, and 12 weeks. Parameters recorded included the number of adult worms recovered by portal perfusion (infectivity); the number of eggs in the feces, the intestine, and the liver; and the ability of th

Genetic and environmental determinants of plasma nitrogen oxides and risk of ischemic heart disease.

Endothelial dysfunction, caused in part by reduced NO bioavailability, is a feature of hypercholesterolemia, hypertension, smoking, and atherosclerosis. We examined whether cholesterol, blood pressure, smoking status, and polymorphisms in the endothelial NO synthase gene (NOS 3) influence NO production (as assessed by the plasma levels of nitrogen oxides, NO(x)) in middle-aged men. We also determined whether plasma NO(x) or NOS 3 genotype predicted the risk of is chemic heart disease (IHD). We studied 3052 men who were initially free of IHD and recruited from 9 UK primary care practices. Blood

Surfactant proteins A and B as interactive genetic determinants of neonatal respiratory distress syndrome.

Prematurity is the most important risk factor predisposing to neonatal respiratory distress syndrome (RDS). Genetic factors are likely to contribute to the risk of this complex disease. The present study was designed to investigate whether the surfactant protein B (SP-B) gene or interaction between the SP-A and SP-B genes has a role in the genetic susceptibility to RDS. The genotype analyses were performed on 684 prematurely born neonates, of whom 184 developed RDS. Of the two SP-B polymorphisms genotyped, the Ile131Thr variation affects a putative N-terminal N:-linked glycosylation site of pr

Genetic polymorphism of CYP2D6 and CYP2C19 metabolism determined by phenotyping Israeli ethnic groups.

Genetic polymorphism of the cytochrome P450 isoenzymes CYP2D6 and CYP2C19 was determined by phenotyping four ethnic groups of the Israeli population. The groups consisted of Ethiopian subjects, Yemenite subjects, and Russian subjects representing first-generation new immigrants and an Israeli Arab group. Dextromethorphan was used as the probe for CYP2D6 activity and mephenytoin was used for CYP2C19 activity. The two drugs were administered simultaneously and urine samples were collected over a period of 8 hours. The CYP2D6 phenotype was determined from the ratio of dextromethorphan conversion

Clinical experience of sex determination by fluorescent in-situ hybridization for preimplantation genetic diagnosis.

In our centre we started using fluorescent in-situ hybridization (FISH) technique for sexing in couples with sex-linked diseases in May 1995. Probes specific for chromosomes X, Y and 18 were applied, allowing us to detect simultaneously both gender and ploidy status. The efficiency of the FISH procedure is 90.4% per biopsied blastomere or 95.2% per biopsied blastomere with a distinct nucleus visible at spreading. Up to December 1997, we treated 15 couples (20 treatment cycles) at risk for X-linked recessive disease and two couples with Yq deletion (two treatment cycles) with the aim of transfe

COMPLEX COLONY STRUCTURE IN SOCIAL INSECTS: I. ECOLOGICAL DETERMINANTS AND GENETIC CONSEQUENCES.

For social insect species, intraspecific variation in colony social structure provides an opportunity to relate the evolution of social behavior to ecological factors. The species Myrmica punctiventris is a cavity-dwelling forest ant that exhibits very different colony structures in two populations in the northeastern United States. Combined data from seasonal censuses, allozyme electrophoresis, and worker hostility tests showed that a population of M. punctiventris in Vermont was strictly monogynous and seasonally polydomous. The same procedures showed that a population of M. punctiventris in

Antibody-mediated autoimmune myocarditis depends on genetically determined target organ sensitivity.

Injury to cardiac myocytes often leads to the production of anti-myosin antibodies. While these antibodies are a marker of myocardial injury, their contribution to pathogenesis in diseases such as autoimmune myocarditis or rheumatic fever is much less clear. We demonstrate in this report that monoclonal anti-myosin antibodies can mediate myocarditis in a susceptible mouse strain. Additionally, we show disease susceptibility depends on the presence of myosin or a myosin-like molecule in cardiac extracellular matrix. This study demonstrates that susceptibility to autoimmune heart disease depends

Roles of gamma interferon and interleukin-4 in genetically determined resistance to Coccidioides immitis.

The profiles of gamma interferon (IFN-gamma) and interleukin-4 (IL-4) production were evaluated during the course of coccidioidomycosis in two inbred mouse strains which differ in their susceptibility to Coccidioides immitis. Cytokine responses, measured at the molecular and protein levels, showed increased levels of IFN-gamma in lung extracts from mice of the resistant DBA/2 strain after a pulmonary challenge, whereas the susceptible BALB/c strain manifested a predominant IL-4 response. The importance of these cytokines in host defense against C. immitis was established by treating the mice w

The resident faecal flora is determined by genetic characteristics of the host. Implications for Crohn's disease?

Recently, an "abnormal" faecal flora has been associated with Crohn's disease, a disease of unknown origin but with pertinent genetic predisposition. Therefore, it was investigated whether the faecal flora is under genetic influence in groups of monozygotic and dizygotic twins. Faecal floras of monozygotic twin siblings were found to be much more alike than those of dizygotic twin siblings. It is concluded that the resident faecal flora is under genetic influence and may be useful parameter to study possible genetic backgrounds of other diseases.

Glycogen-storage disease in rats, a genetically determined deficiency of liver phosphorylase kinase.

Rats from an inbred strain (NZR/Mh) were found to have high concentrations of glycogen in their livers, even after 24 h of starvation. Despite this, blood glucose concentrations were well maintained on starvation for up to 72 h. The primary defect is a deficiency of liver phosphorylase kinase, causing a lack of active glycogen phosphorylase, although total phosphorylase is normal. The intravenous injection of glucagon caused a rapid activation of cyclic AMP-dependent protein kinase in the liver, but no increase in either phosphorylase kinase or phosphorylase a activity. Although total glycogen

Cellular basis for genetically determined enhanced resistance of certain mouse strains to listeriosis.

The characteristics of the mononuclear phagocytes mediating resistance to infection with Listeria during the early phase (0 to 48 h) of the response have been investigated in genetically determined susceptible (A/J) and resistant (C57BL/6, B10.A/SgSn) strains of mice. Irradiation immediately before infection profoundly enhanced the bacterial growth in the resistant strain, while having no effect in the susceptible strain, over a wide range (3 x 10(3) to 10(5)) of infective doses. This effect of irradiation is demonstrable at low-dose radiation (200 roentgens) and can be reversed by repopulatio

Aspergillus fumigatus isolated from diverse wildfowl exhibit distinct antifungal susceptibility profiles driven by genetic and non-genetic determinants.

Invasive aspergillosis (IA) is a fungal infection caused by Aspergillus species affecting humans and animals, including birds. Such infections have severe impacts on host health, with the efficacy of current treatment options dwindling against rising rates of antifungal resistance. This scenario represents a critical One Health challenge influenced by climate change at the intersection of animal, human, and environmental health. In this study, we isolated and identified four fungal isolates from infected wildfowl in Southern Ontario, Canada, as Aspergillus fumigatus. Antifungal susceptibility

Genetic and environmental determinants of violence risk in psychotic disorders: a multivariate quantitative genetic study of 1.8 million Swedish twins and siblings.

Patients diagnosed with psychotic disorders (for example, schizophrenia and bipolar disorder) have elevated risks of committing violent acts, particularly if they are comorbid with substance misuse. Despite recent insights from quantitative and molecular genetic studies demonstrating considerable pleiotropy in the genetic architecture of these phenotypes, there is currently a lack of large-scale studies that have specifically examined the aetiological links between psychotic disorders and violence. Using a sample of all Swedish individuals born between 1958 and 1989 (n=3 332 101), we identifie

Genetically determined very low lipoprotein(a) is not associated with type 2 diabetes and glycemic traits.

BACKGROUND: Lipoprotein (a) [Lp(a)] is an independent atherosclerotic cardiovascular disease risk factor, with levels largely determined by variation in the LPA gene. Despite interests in therapeutic Lp(a) lowering, the phenotypic effects of very low Lp(a) are incompletely characterized, with some studies suggesting increased type 2 diabetes risk. OBJECTIVES: Estimate associations between very low Lp(a), inferred by LPA loss of function variant rs41272114, with type 2 diabetes and glycemic biomarkers. METHODS: A multi-population study of European, Hispanic/Latino, and South Asian populations

Genetic Determinants of Vascular Dementia: Blood Metabolite Associations and Candidate Therapeutic Target Prioritization.

Vascular dementia (VaD), caused by reduced cerebral blood flow, is a common form of dementia lacking effective treatments. This study used genome-wide association study (GWAS) summary data, Mendelian randomization (MR), and colocalization analyzes to prioritize blood metabolites and candidate therapeutic targets associated with VaD. Key analyzes included metabolic pathway enrichment, a descriptive Phenome-Wide Association Study (PheWAS) of the broader curated association profiles of prioritized genes, mediation analysis, and in-silico compound prioritization with molecular docking. Twenty-four

Preimplantation genetic testing to determine polygenic disease risk: Will we know when to stop?

Recent advances in genomics technologies have facilitated the introduction of new tools that broaden the scope of applications for genetic data. Preimplantation genetic testing using polygenic risk scores (PGT-P), a technique based on the fact that complex traits are influenced by multiple genes, is one such technology. Polygenic risk scoring involves the analysis of many genes in a single embryo to assess the odds of expressing diseaseand non-disease-related factors. This technique could alleviate subsequent burdens on healthcare systems, as it could lead to a population with reduced polygeni

Biomarkers and genetic determinants of cardiac sarcoidosis: current status, the unmet needs and future perspectives.

Sarcoidosis is a systemic disorder driven by genetic predisposition, environmental exposures, and immune dysregulation, resulting in the formation of noncaseating granulomas across multiple organs. In cardiac sarcoidosis (CS), immune cell infiltration of the myocardium, epicardium, and endocardium may lead to conduction disturbances, ventricular arrhythmias, and heart failure. While overt cardiac involvement was historically considered rare, affecting only 5% of sarcoidosis patients, the wider availability and improved sensitivity of contemporary cardiac imaging have revealed a substantially h
