ocs element promoter sequences are activated by auxin and salicylic acid in Arabidopsis.

ocs elements are a group of promoter elements that have been exploited by two distinct groups of plant pathogens, Agrobacterium and certain viruses, to express genes in plants. We examined the activity of single and multiple ocs elements linked to a minimal plant promoter and the uidA reporter gene in transgenic Arabidopsis. beta-Glucuronidase activity was detected only in root tips and in callus tissue after auxin treatment. A more sensitive assay revealed that auxin treatment also increased ocs element activity in aerial parts of the plant, although the absolute levels of ocs element activit

Cell Death Triggered by the YUCCA-like Bs3 Protein Coincides with Accumulation of Salicylic Acid and Pipecolic Acid But Not of Indole-3-Acetic Acid.

The pepper (Capsicum annuum) resistance gene bacterial spot3 (Bs3) is transcriptionally activated by the matching Xanthomonas euvesicatoria transcription-activator-like effector (TALE) AvrBs3. AvrBs3-induced Bs3 expression triggers a rapid and local cell death reaction, the hypersensitive response (HR). Bs3 is most closely related to plant flavin monooxygenases of the YUCCA (YUC) family, which catalyze the final step in auxin biosynthesis. Targeted mutagenesis of predicted NADPH- and FAD-cofactor sites resulted in Bs3 derivatives that no longer trigger HR, thereby suggesting that the enzymatic

OsMYB8-OsARF12/25 module fine-tunes tiller angle via auxin signaling pathway in rice.

Tiller angle is a critical determinant of rice plant architecture and significantly impacts grain yield by influencing planting density and photosynthetic efficiency. Although auxin signaling is known to affect tiller angle in rice, the detailed regulatory networks remain largely unknown. In this study, we identify OsMYB8, an R2R3-MYB transcription factor, as a positive regulator of rice tiller angle. Functional analyses revealed that loss-of-function mutants of OsMYB8 exhibited reduced tiller angles and a more compact architecture, while overexpression of OsMYB8 resulted in more expanded till

Host-recruited Bacillus and Pseudomonas strains provide potential biocontrol and yield protection against rice bacterial leaf blight.

Bacterial leaf blight (BLB), caused by Xanthomonas oryzae pv. oryzae (Xoo), threatens global rice production while growing bactericide resistance and environmental concerns necessitate sustainable disease management alternatives. We employed a microbiome-guided, habitat-specific isolation strategy targeting naturally recovered plants from BLB-endemic hotspots across ten districts of Punjab, Pakistan operating on the ecological premise that plants under pathogen pressure selectively enrich protective microbial taxa, making disease-affected hosts the most coherent source of adapted biocontrol ag

Design, Synthesis, and Herbicidal Evaluation of Novel Synthetic Auxin Herbicides Containing 6-Indolylpyridine Oxime Ester/Amine.

In this study, a series of 6-indolylpyridine oxime ester/amide derivatives were synthesized as novel synthetic auxin herbicides (SAHs) for postemergence herbicidal applications. At 30 g ai/ha, compounds 9q, 9u, 9v and 9w demonstrated inhibition rates of 90 to 100% against weeds Echinochloa crus-galli (EC) and Digitaria sanguinalis (DS). Even at a reduced 7.5 g ai/ha, these compounds maintained over 90% inhibition against four broadleaf weeds, with effects comparable to those of commercial herbicides halauxifen-methyl (HAM), indolauxipyr (IND) and indolauxipyr-cyanomethyl (INC). Synergistic Effect of Melatonin and Lysinibacillus fusiformis L. (PLT16) to Mitigate Drought Stress via Regulation of Hormonal, Antioxidants System, and Physio-Molecular Responses in Soybean Plants.

Drought is one of the most detrimental factors that causes significant effects on crop development and yield. However, the negative effects of drought stress may be alleviated with the aid of exogenous melatonin (MET) and the use of plant-growth-promoting bacteria (PGPB). The present investigation aimed to validate the effects of co-inoculation of MET and Lysinibacillus fusiformis on hormonal, antioxidant, and physio-molecular regulation in soybean plants to reduce the effects of drought stress. Therefore, ten randomly selected isolates were subjected to various plant-growth-promoting rhizobac

Plant growth-promoting potential of 'Myroides gitamensis' isolated from virgin soils of Punjab.

Phosphate-solubilizing (PS) and phosphate-mineralizing (PM) bacteria are considered vital for augmenting the plant growth through phosphorus mobilization and plant growth-promoting attributes. In the present study, a rhizospheric bacterium was isolated from the virgin land of Punjab, India and identified as 'Myroides gitamensis' BSH-3 through 16S rRNA sequencing. 'M. gitamensis' showed potential halo zone on Pikovskaya agar. The novelty of the study lies in the fact that plant growth-promoting potential of 'M. gitamensis' has not been studied earlier. It was able to solubilize 17.53-106.66 µg/

Overexpressing GH3.1 and GH3.1L reduces susceptibility to Xanthomonas citri subsp. citri by repressing auxin signaling in citrus (Citrus sinensis Osbeck).

The auxin early response gene Gretchen Hagen3 (GH3) plays dual roles in plant development and responses to biotic or abiotic stress. It functions in regulating hormone homeostasis through the conjugation of free auxin to amino acids. In citrus, GH3.1 and GH3.1L play important roles in responding to Xanthomonas citri subsp. citri (Xcc). Here, in Wanjingcheng orange (Citrus sinensis Osbeck), the overexpression of CsGH3.1 and CsGH3.1L caused increased branching and drooping dwarfism, as well as smaller, thinner and upward curling leaves compared with wild-type. Hormone determinations showed that

Roles of Small-Molecule Compounds in Plant Adventitious Root Development.

Adventitious root (AR) is a kind of later root, which derives from stems and leaf petioles of plants. Many different kinds of small signaling molecules can transmit information between cells of multicellular organisms. It has been found that small molecules can be involved in many growth and development processes of plants, including stomatal movement, flowering, fruit ripening and developing, and AR formation. Therefore, this review focuses on discussing the functions and mechanisms of small signaling molecules in the adventitious rooting process. These compounds, such as nitric oxide (NO), h

Metabolite of tryptophan promoting changes in EEG signal and the oxidative status of the brain.

Tryptophan is an essential amino acid precursor of neurotransmitter serotonin and triptamine. During its metabolism, indole-3-acetic acid (IAA) is generated; this substance presents both antioxidant and prooxidant effects in different biological systems in addition to hipoglicemic effects. To date, electroencephalography (EEG) has been used to evaluate the temporal effect of several substances in neurotransmission. The goal of this study was to characterize the effect of IAA in the brain by analysing the EEG signal and evaluate the oxidative status by means of biochemical parameters. The EEG w

Genome-wide characterization of BraABCB transporters reveals their potential roles in hormone responses in Brassica rapa var. parachinensis.

Thirty-six BraABCB genes were identified in Brassica rapa var. parachinensis; expression and interaction analyses suggest BraABCB27 and BraABCB28 as hormone-responsive candidates associated with BRI1-related proteins. ABCB transporters are ATP-dependent membrane proteins that mediate the transport of diverse substrates, including phytohormones, and play important roles in plant development and environmental adaptation. Previous studies in Arabidopsis have shown that several ABCB proteins participate in phytohormone transport, including auxin and brassinosteroid transport, whereas the functions

Auxin and Cytokinins-Mediated Regulation of Adventitious Shoot Regeneration via the RpARR12/RpARF19-RpWOX4-RpLBD4 Pathway in Robinia pseudoacacia.

Woody plants are fast-growing and ecologically important, with widespread applications in urban landscaping and ecological restoration. However, limited understanding of the molecular mechanisms underlying adventitious shoot regeneration has hindered advances in tissue culture and genetic improvement of woody species. Here, we present a comprehensive framework for elucidating adventitious shoot regeneration in Robinia pseudoacacia. Histological analyses delineated key developmental stages, while RNA-sequencing and hormone profiling highlighted the critical roles of auxin and cytokinins. WGCNA

Novel Bacillus and Prestia isolates from Dwarf century plant enhance crop yield and salinity tolerance.

Soil salinity is a major environmental stressor impacting global food production. Staple crops like wheat experience significant yield losses in saline environments. Bioprospecting for beneficial microbes associated with stress-resistant plants offers a promising strategy for sustainable agriculture. We isolated two novel endophytic bacteria, Bacillus cereus (ADJ1) and Priestia aryabhattai (ADJ6), from Agave desmettiana Jacobi. Both strains displayed potent plant growth-promoting (PGP) traits, such as producing high amounts of indole-3-acetic acid (9.46, 10.00 µgml-1), ammonia (64.67, 108.97 µ

N6-adenosine methylation of mRNA integrates multilevel auxin response and ground tissue development in Arabidopsis.

N6-methyl adenosine (m6A) is a widespread internal mRNA modification impacting the expression of numerous genes. Here, we characterize auxin-related defects among the pleiotropic phenotypes of hypomorphic Arabidopsis thaliana mutants with impaired m6A status and reveal that they show strong resistance to exogenously applied auxin. By combining major published m6A datasets, we propose that among high-confidence target transcripts emerge those encoding the main components required for auxin signaling, including the TIR1/AFB auxin receptors and ARF transcriptional regulators. We also observe subt

Biotoxic impact of heavy metals on growth, oxidative stress and morphological changes in root structure of wheat (Triticum aestivum L.) and stress alleviation by Pseudomonas aeruginosa strain CPSB1.

Rapid industrialization and uncontrolled metal discharge into environment is a global concern for crop production. Metal tolerant bacterium isolated from chilli rhizosphere was identified as Pseudomonas aeruginosa by 16S rDNA sequence analysis. Pseudomonas aeruginosa tolerated high concentrations of Cu (1400 μg ml-1), Cd (1000 μg ml-1) and Cr (1000 μg ml-1). Pseudomonas aeruginosa CPSB1 produced multiple plant growth promoting biomolecules in the presence and absence of metals. Strain CPSB1 solubilized P at 400 μg ml-1 of Cd, Cr and Cu. The strain was positive for indole-3-acetic acid (IAA), s

The arabidopsis IDD14, IDD15, and IDD16 cooperatively regulate lateral organ morphogenesis and gravitropism by promoting auxin biosynthesis and transport.

The plant hormone auxin plays a critical role in regulating various aspects of plant growth and development, and the spatial accumulation of auxin within organs, which is primarily attributable to local auxin biosynthesis and polar transport, is largely responsible for lateral organ morphogenesis and the establishment of plant architecture. Here, we show that three Arabidopsis INDETERMINATE DOMAIN (IDD) transcription factors, IDD14, IDD15, and IDD16, cooperatively regulate auxin biosynthesis and transport and thus aerial organ morphogenesis and gravitropic responses. Maize nitrilases have a dual role in auxin homeostasis and beta-cyanoalanine hydrolysis.

The auxin indole-3-acetic acid (IAA), which is essential for plant growth and development, is suggested to be synthesized via several redundant pathways. In maize (Zea mays), the nitrilase ZmNIT2 is expressed in auxin-synthesizing tissues and efficiently hydrolyses indole-3-acetonitrile to IAA. Zmnit2 transposon insertion mutants were compromised in root growth in young seedlings and sensitivity to indole-3-acetonitrile, and accumulated lower quantities of IAA conjugates in kernels and root tips, suggesting a substantial contribution of ZmNIT2 to total IAA biosynthesis in maize. Argiotoxin-636 blocks effects of N-methyl-D-aspartate on lateral line of Xenopus laevis at concentrations which do not alter spontaneous or evoked neural activity.

Activity of primary afferent neurons in acousticolateralis organs can be modulated by excitatory amino acids (EAA) and their antagonists, and EAA are among the better candidates for the transmitter(s) between hair cells and afferent neurons. Argiotoxin-636 (ATX) is a spider venom toxin that is a selective antagonist of the N-methyl-D-aspartate (NMDA) subtype of excitatory amino acid receptors in vertebrates. In the present study, the effects of NMDA alone and in combination with ATX on resting firing rate (spontaneous activity) and mechanically stimulated activity of lateral line afferent neur
